        STUDIES ON IMMUNOTOXIC POTENTIAL OF METHAMPHETAMINE (MA) IN Balb/C MICE II. The Functional Alteration of Effector Cells in Immune System

        Several populations of lymphocytes possess receptors for autonomic neurotransmitter, which make lymphocytes susceptible to autonomic stimulation. This study was to evaluate the functional alternation of effector cells of the immune system. Female Balb/C mice, 15-20 g, were injected with MA subcutaneously under various conditions. Mixed lymphocyte reaction (MLR) showed certain T cell subsets were affected by MA. The level of interleukin-2 (IL-2) production was inhibited due to a defect in expression of the IL-2 receptor. In mice injected with 20 mg MA/kg, 1 day before assay, phagocytosis of peritoneal macrophages showed $14.07\pm3%$, which was similar degree to 5 mg MA/kg treatment for 4 consecutive days. Phagocytosis was almost recovered to that of control after 4 day in 20 mg/kg injected mice. Maximum inhibition of plaque forming cell (PFC) occurred when MA was given early, indicating the inductive time point of antibody production was affected. The cortisol level increased in the MA treated group (0.05, 0.20, and $0.08{\mu}g$/dl for control, low, and high dose-MA treated mice, respectively). Based on these results, MA has general suppression effects on the immune systems by functional alteration of effector cells. Considering the increment of serum cortisol levels, MA partially impacts the neuroendocrine system to lead to failure of immune response.

      • A novel adipokine CTRP1 stimulates aldosterone production

        <P>Complement-C1q TNF-related protein 1 (CTRP1), a member of the CTRP superfamily, is expressed at high levels in adipose tissues of obese Zucker diabetic fatty (fa/fa) rats, and CTRP1 expression is induced by proinflammatory cytokines, including TNF-alpha and IL-1beta. In the present study, we investigated stimulation of aldosterone production by CTRP1, since it was observed that CTRP1 was specifically expressed in the zona glomerulosa of the adrenal cortex, where aldosterone is produced. Increased aldosterone production by CTRP1 in cells of the human adrenal cortical cell line H295R was dose-dependent. Expression levels of aldosterone synthase CYP11B2 were examined to investigate the molecular mechanisms by which CTRP1 enhances the production of aldosterone. The expression of CYP11B2 was greatly increased by treatment with CTRP1, as was the expression of the transcription factors NGFIB and NURR1, which play critical roles in stimulation of CYP11B2 gene expression. It was also revealed that angiotensin II-induced aldosterone production is, at least in part, mediated by the stimulation of CTRP1 secretion, not by the increase of CTRP1 mRNA transcription. In addition, the levels of CTRP1 were significantly up-regulated in hypertensive patients' serum. As CTRP1 was highly expressed in obese subjects as well as up-regulated in hypertensive patients, CTRP1 may be a newly identified molecular link between obesity and hypertension.</P>

        Triptolide suppresses interleukin-1beta-induced human beta-defensin-2 mRNA expression through inhibition of transcriptional activation of NF-kappaB in A549 cells.

        <P>The immunosuppressive effect of triptolide has been associated with suppression of T-cell activation. However, the immunosuppressive effects of triptolide on innate immunity in the epithelial barrier remain to be elucidated. Human beta-defensin (HBD)-2 is an inducible antimicrobial peptide and plays an important role in the innate immunity. We have previously demonstrated that IL-1beta induced HBD-2 mRNA expression in A549 cells through activation of nuclear factor-kappaB (NF-kappaB) transcriptional factor as well as p38 mitogen-activated protein kinase (MAPK), c-Jun N-terminal kinase (JNK), or phosphatidylinositol-3-kinase (PI3K). In this study, we investigated effects of triptolide on IL-1beta-induced HBD-2 mRNA expression in A549 cells. Triptolide inhibited IL-1beta-induced HBD-2 mRNA expression in a dose-dependent manner. Addition of triptolide did not suppress activation of p38 MAPK, JNK, or PI3K in response to IL-1beta. Triptolide inhibited IL-1beta-induced MAPK phosphatase-1 expression at the transcriptional level and resulted in sustained phosphorylation of JNK or p38 MAPK, explaining the little effect of triptolide on IL-1beta-induced phosphorylation of these kinases. Although triptolide partially suppressed IL-1beta-mediated degradation of IkappaB-alpha and nuclear translocation of p65 NF-kappaB, triptolide potently inhibited NF-kappaB promoter-driven luciferase activity in A549 cells. These results collectively suggest that the inhibitory effect of triptolide on IL-1beta-induced HBD-2 mRNA expression in A549 cells seems to be at least in part mediated through nuclear inhibition of NF-kappaB transcriptional activity, but not inhibition of p38 MAPK, JNK, or PI3K. This inhibition may explain the ability of triptolide to diminish innate immune response.</P>

        <P><B>Abstract</B></P><P>CTRP1, a member of the CTRP superfamily, consists of an N-terminal signal peptide sequence followed by a variable region, a collagen repeat domain, and a C-terminal globular domain. CTRP1 is expressed at high levels in adipose tissues of LPS-stimulated Sprague-Dawley rats. The LPS-induced increase in CTRP1 gene expression was found to be mediated by TNF-α and IL-1β. Also, a high level of expression of CTRP1 mRNA was observed in adipose tissues of Zucker diabetic fatty (<I>fa/fa</I>) rats, compared to Sprague-Dawley rats in the absence of LPS stimulation. These findings indicate that CTRP1 expression may be associated with a low-grade chronic inflammation status in adipose tissues.</P>

        <P>NK cells are a key component of innate immune systems, and their activity is regulated by cytokines and hormones. Adiponectin, which is secreted from white adipose tissues, plays important roles in various diseases, including hypertension, cardiovascular diseases, inflammatory disorders, and cancer. In this study the effect of adiponectin on NK cell activity was investigated. Adiponectin was found to suppress the IL-2-enhanced cytotoxic activity of NK cells without affecting basal NK cell cytotoxicity and to inhibit IL-2-induced NF-kappaB activation via activation of the AMP-activated protein kinase, indicating that it suppresses IL-2-enhanced NK cell cytotoxicity through the AMP-activated protein kinase-mediated inhibition of NF-kappaB activation. IFN-gamma enhances NK cell cytotoxicity by causing an increase in the levels of expression of TRAIL and Fas ligand. The production of IFN-gamma, one of the NF-kappaB target genes in NK cells, was also found to be suppressed by adiponectin, accompanied by the subsequent down-regulation of IFN-gamma-inducible TRAIL and Fas ligand expression. These results clearly demonstrate that adiponectin is a potent negative regulator of IL-2-induced NK cell activation and thus may act as an in vivo regulator of anti-inflammatory functions.</P>

      • 升麻葛根湯加味方이 消炎작용에 미치는 영향

        This study was carried out to investigate the effects of SMG on the in vitro and in vivo inflammatory reactions. In experiment I, in vitro tests, ethanol extract of SMG showed potent radical scavenging activity tested by DPPH(I,1-diphenyl-2-picryl-hyrazyl) method and inhibited the lipopolysaccharide-induced gene expressions of interleukin-1${\beta}$, interleukin-6 and tumor necrosis factor-${\alpha}$ (above 50$\%$ at a concentration of 50㎍/㎖) by the macrophage RAW 246.7 cells. Among the herbal ingredients of SMG, ethanol extracts of Scutellaria baicalensis, Paeonia lactiflora, Glycyrrhiza glabra showed potent radical scavenging activity. And Glycyrrhiza glabra and Scutellaria baicalensis showed potent inhibitory activity of nitric oxide production. Especially, ethanol extract of Scutellaria baicalensis inhibited the gene expression of IL-1${\beta}$, IL-6 and TNF-${\alpha}$. In cyclooxygenase-2 assay, Scutellaria baicalensis and Glycyrrhiza glabra showed the potent inhibition of prostaglandin E2 generation. In experiment 2, in vivo tests, SMG showed inhibitory effects on vascular permeability (28.7$\%$) and leukocyte migration (11.5$\%$). These results mean that SMG has a anti-inflammatory effects by it's inhibitory effects of leukocyte migration and vascular permeability as well as it's inhibitory effects of lipopolysaccharide-induced gene expression of IL-1${\beta}$, IL-6 and TNF-${\alpha}$, and radical scavenging activity. Therefore, I expect that SMG may be used as a effective drug for treatment on inflammation.

        This paper is concerned with and efficient model for the structural analysis of threaded connections which is used to assemble the tubular connections such as pipes. Advanced mechanics has been used to develop the model which yields acceptably accurate solutions for stresses when compared to other methods such as experimental or finite element analysis procedures. Beam-on-elastic-foundation equations are used to compute stresses due to assembly of the connection while bolt-nut equation and Lame' equation are used for axial loads and pressures. Numerical results show that our model is computationally efficient and valid in terms of stress distribution for make-up. The current model can provide the quick and inexpensive analysis tools to the production engineer with reasonable accuracy.

        응급실에 내원한 약물중독환자의 지역별 비교분석

        We reviewed retrospectively 230 patients who came to emergency room of Kang Nam St. Mary's Hospital and Uijongbu St. Mary's Hospital between 1. January and 31. December 1994. We classified them according to their age, sex, a sort of drug, the cause of poisoning, prehospital care, time-lapse to arrive at hospital after poisoning, psychiatric problem the status of patients. *We conclude as follows; 1. The male to female ratio was totally 1 : 1.98, 1 : 1.24 in a urban area, 1 : 2.43 in a rural area. 2. On age of the patients in this study, the peake incidence was between 20 and 39(55.2%). 3. On seasonal prevalence, the poisoning was most common in summer and spring, in urban area(35.7%) and rural area(37.0%) respectively. 4. The common drug were as follow; Doxylamine, Organophosphate, Acetaminophen in urban area, Paraquat, Organophospate, Doxylamine in rural area 5. The percentage of suicidal attempt were 81.8% and 87.7% in urban area and rural area respectively. 6. The poisoning patients had psychatric problem in 19.4% and 24.6%, in urban area and rural area respectively. 7. Before arrival to emergency room, 27.7% of patient was done prehospital care in rural area. 8. On arrival to emergency room, 95.1% and 73.8% of patients show stable vital sign in urban area and rural area respectively.

        Behc¸et의 患者의 單球細胞에서 溫淸飮의 cytokine 分泌抑制 효과

        Behc¸ets disease is a systemic inflammatory disorder. The etiology and pathogenesis of Behc¸ets disease has yet been fully elucidated but might involve immune dysfunction. Cytokines involved in the regulation of inflammatory reactions and immune responses may play a role in the pathogenesis of Behc¸ets disease (BD). Onchungeum is an Oriental herbal medication, which has been successfully used in Korea for the treatment of BD. This report describes modulation effects of Onchungeum on cytokine production from phytohaemagglutinin (PHA)-stimulated peripheral blood mononuclear cells (PBMC) of Behc¸ets patients by ELISA. Onchungeum significantly inhibited the production of pro-inflammatory cytokines, TNF-α and IL-1β, compared to absence of Onchungeum (by 52.3 1.4% inhibition for TNF-α and 113.5 3.3% for IL-1β, p < 0.001). Onchungeum also inhibited the production of IFN-γ, immunoregulatory Th1 cytokine, by 89.4 0.8% (p < 0.001). The inhibitory effects of Onchungeum on cytokine production showed dose-dependent manner, and the pre-treatment of 1㎎/㎖ Onchungeum had better effects than immunosuppressive drug for treatment of BD, cyclosporin A. Our results suggest that Onchungeum treatment for Behc¸ets disease patients may have pharmacologic activities and abilities of regulation of immune and inflammatory responses by cytokine modulation.

        As the function Ⅰ(hkl) of the X-ray intensity has a center of symmetry assuming anomalous scattering is negligible, Ⅰ(hkl) shows the symmetry of a centrosymmetric point group, i.e., a Laue group. The symmetry of each Laue group gives rise to its equivalent intensity relation, from which a scheme for identification of Laue group by means of oscillation and Weissenberg zeroth layer photographs has been obtained.
