혈청 Fructosamine에 대한 임상적 유용성에 관한 연구

Diabetes mellitus is a universal health problem and may occur at any age. The traditional biochemical measurements for initially detection patients with diabetes mellitus are random estimations of blood and urine glucose concentrations. Despite their common use both tests are fairly non-specific, being influenced by wide variety of condition. Measurement of the HbA_(1c) in a random blood specimen is a more simple screening test for diabetes, and widely used as an index of diabetic control. But unfortunately, it has difficulties with methodology. Fructosamine has been described as a new parameter for the estimation of glycosylated protein, It is based on the reduction potential of ketoamines in an alkaline medium and measures colorimethcally the concentration of all glycosylated semm proteins belonging to the class of fructosamine. It reflects the time-integrated glucose concentration over the previous 1 to 3 weeks and can be used for the follow up of diabetes mellitus. We measured serum fructosamine concentration and compared with any other items. The results are summehzed as follows : 1. Within-run and between-run imprecision were assessed by 20 analysis of 3 differential fructosamine concentration in control sera. The respective coefficients of variation were 2.9%, 6.6% in low concentration, 2.2%, 5.1% in medium concentration and 3.3%, 4.7% in high concentration. 2. The serum fructosamine concentration were 2.26±0.21 mmol/L in normal control subjects(n=29) and 3.35±0.65 mmol/L (n=62) in patients with diabetes mellitus (p<0.001). 3. In diabetic patients (n=62), the serum fructosamine concentrations correlated less significantly with glucose (Y(fructosamine) = 0.01×(glucose)+2.52, r=0.54, P<0.001) and more significantly with HbA_(1c) (Y(fructosamine) = 0.22×(HbA_(1c))+1.71 , r=0.63, P<0.001). 4. The concentrations of fructosamine in serum and EDTA plasma were not significently diferent (P<0.01). So, Plasma is as well suited to fructosamine determinations as serum(Y(serum) = 0.95×(Plasma)+0.25, r=0.96). 5. No interference was observed with lipemic sera (cholesterol) in detection of serum fructosamine(P=0.393).

Apple mosaic virus (ApMV), a member of the genus Ilarvirus, was detected and isolated from diseased 'Fuji' apple (Malus domestica) in Korea. The coat protein (CP) genes of two ApMV strains, denoted as ApMV-Kl and ApMV-K2, were amplified by using the reverse transcription and polymerase chain reaction (RT-PCR) and were analyzed thereafter. The objectives were to define the molecular variability of genomic information of ApMV found in Korea and to develop virus-derived resistant gene source for making virus-resistant trans-genic apple. RT-PCR amplicons for the APMVS were cloned and their nucleotide sequences were determined. The CPs of ApMV-Kl and ApMV-K2 consisted of 222 and 232 amino acid residues, respectively. The identities of the CPs of the two Korean APMVS were 93.1% and 85.6% at the nucleotide and amino acid sequences, respectively. The CP of ApMV-Kl showed 46.1-100% and 43.2-100% identities to eight different ApMV strains at the nucleotide and amino acid levels, respectively. When ApMV-PV32 strain was not included in the analysis, ApMV strains shared over 83.0% and 78.6% homologies at the nucleotide and amino acid levels, respectively. ApMV strains showed heterogeneity in CP size and sequence variability. Most of the amino acid residue differences were located at the N-termini of the strains of ApMV, whereas, the middle regions and C-termini were remarkably conserved. The APMVS were 17.(1-54.5% identical with three other species of the genus Ilarviyus. ApMV strains can be classified into three subgroups (subgroups I, II, and III) based on the phylogenetic analysis of CP gene in both nucleotide and amino acid levels. Interestingly, all the strains of subgroup I were isolated from apple plants, while the strains of subgroups II and III were originated from peach, hop, or pear, The results suggest that ApMV strains co-evolved with their host plants, which may have resulted in the CP heterogeneity.

Background: To evaluate the efficacy of peri-urethral thrombin instillation at the surgical site after trans-urethral prostate surgery in benign prostatic hyperplasia (BPH), with the objective of reducing post-operative hematuria and preserving voiding function. Methods: The study was retrospectively analyzed a total of 122 patients who had trans-urethral prostate surgery between January 2019 and March 2023. Among these, 33 received thrombin instillation, while 89 did not receive it. In the test group, a mixture of 5 cc of saline solution and 5,000 IU of thrombin was instilled below the balloon, targeting the surgical site by inserting a 5 Fr feeding tube along with the Foley catheter. We compared hematologic data and uroflowmetry (UFM) between pre and post-operative periods, the duration of continuous bladder irrigation (CBI) and Foley catheter indwelling period. Results: The mean age and prostate size in the test group were 71.52±7.15 years and 54.05±24.73 g, while 71.38±8.22 years and 64.52±28.43 g in the control group. The prostate resected weight was 24.97±15.77 g and 27.58±17.00 g respectively. The hemoglobin and platelet levels were −1.05±0.81 g/dL and −42.79±35.59×103/L in the test group, and −1.08±0.97 g/dL and −27.93±40.49×103/L, in the control group. The test group showed better outcomes in CBI application time (21.31±12.15 hours) and Foley catheter indwelling time (50.97±14.63 hours) compared to the control group (34.42±22.68 hours and 70.33±33.95 hours) (P<0.001). Conclusion: After trans-urethral prostate surgery, thrombin agent instillation can prevent post-operative hematuria and facilitate quicker recovery to daily activities.
Functional modification of Drosophila intestinal stem cells by ionizing radiation.

<P>Although the diverse effects of ionizing radiation on biological and pathological processes at various levels ranging from molecular to whole body are well studied, the effects on adult stem cells by ionizing radiation remain largely unknown. In this study, we characterized the functional modifications of adult Drosophila midgut intestinal stem cells after ionizing radiation treatment. A dose of 10 Gy of radiation decreased the proliferative capacity of intestinal stem cells. Interestingly, after irradiation at 2 Gy, the intestinal stem cells exhibited increased proliferative activity, misdifferentiation and γH2AvD and 8-oxo-dG levels. In addition, the guts irradiated with 2 Gy showed increased JNK and AKT activities. Furthermore, we showed that 2 Gy of ionizing radiation induced centrosome amplification in intestinal stem cells of adult midguts. Our data gives molecular insights into the effects of ionizing radiation on functional modifications of stem cells. The adult Drosophila midgut intestinal stem cells offer a potentially rich new system for the exploration of the biological effects of ionizing radiation.</P>
