        Human Placenta Extract Could Promote Proliferating Cell Nuclear Antigen Expression during Liver Regeneration Induced by Partial Hepatectomy in Rats

        Human placenta extract (hPE) has therapeutic potential against certain diseases such as burn injury, liver cirrhosis and chronic wound through stimulating tissue repair processes. However, the effects of hPE on liver regeneration in animals are unknown. This study investigated the effect of hPE on the expression of proliferating cell nuclear antigen (PCNA) during liver regeneration induced by partial hepatectomy (PH) in rats. The activities of AST, ALT and ALP increased during a few days after PH. A high level of ALP was particularly seen at day 3 in the control group. All the levels of experimental groups were normalized by day 5 after PH. On immunohistochemistry, the expression of PCNA increased at the early days, showed a peak at day 3 after PH. The PCNA staining was more obvious in the experimental group over the whole period. By western blotting, PCNA seemed to be more strongly expressed in the hPE injected group in the early stage and fell to almost undetectable levels at day 7. On immunocytochemical observations, the number of PCNA-gold particles in the nuclei at day 1 of the hPE treated groups was more than those of the untreated groups. The results suggest that hPE could accelerate liver regeneration induced by PH involving the expression of PCNA in rats.

        류마티스관절염으로 오인된 Mycobacterium Marinum 감염에 의한 다발성 관절염

        Mycobacterium marinum (M. marinum) is a nontuberculosis mycobacterium that is normally associated with cutaneous lesions. Most cases occur after an individual has suffered minor trauma in association with contaminated fresh or salt water. In immunocompromised patients, M. marinum can cause osteomyelitis, synovitis and/or disseminated infection, which can be mistaken for rheumatoid arthritis (RA) or remitting seronegative symmetrical synovitis with pitting edema (RS3PE). Therefore, precise history taking and specimen culture are very important for early diagnosis and appropriate treatment. We report an unusual case of septic polyarthritis due to M. marinum that was diagnosed by culture and improved with antibiotic therapy.

        Sebekia benihana에서 Streptomyces coelicolor SCO4967 유전자 도입을 통한 하이드록실 사이클로스포린 A의 생전환

        Actinomycetes are Gram-positive soil bacteria and one of the most important industrial microorganisms due to superior biosynthetic capabilities of many valuable secondary metabolites as well as production of various valuable bioconversion enzymes. Among them are cytochrome P450 hydroxylase (CYP), which are hemoproteins encoded by a super family of genes, are universally distributed in most of the organisms from all biological kingdoms. Actinomycetes are a rich source of soluble CYP enzymes, which play critical roles in the bioactivation and detoxification of a wide variety of metabolite biosynthesis and xenobiotic transformation. Cyclosporin A (CyA), one of the most commonly-prescribed immunosuppressive drugs, was previously reported to be hydroxylated at the position of 4th N-methyl leucine by a rare actinomycetes called Sebekia benihana, leading to display different biological activity spectrum such as loss of immunosuppressive activities yet retaining hair growth-stimulating side effect. In order to improve this regio-selective CyA hydroxylation in S. benihana, previously-identified several secondary metabolite up-regulatory genes from Streptomyces coelicolor and S. avermitilis were heterologously overexpressed in S. benihana using an ermE * promoter-containing Streptomyces integrative expression vector. Among tested, SCO4967 encoding a conserved hypothetical protein significantly stimulated region-specific CyA hydroxylation in S. benihana, implying that some common regulatory systems functioning in both biosynthesis and bioconversion of secondary metabolite might be present in different actinomycetes species.

        해표지증: 국내 환자들의 임상적 양상, 분류 및 수술적 치료

        목적: 해표지증은 상완골, 요골, 척골이 부분적 혹은 전체적으로 결손되고 수부는 남아 있는 매우 드문 기형이다. 국내에서 해표지증 환자에 대한 임상적 연구는 5건의 출생 보고를 제외하고는 찾아볼 수 없다. 본 연구에서는 저자들이 경험한 해표지증의 임상적 양상 및 분류, 수술적 치료에 대하여 보고하고자 한다. 대상 및 방법: 1993년 1월부터 2007년 8월까지 본 병원에서 상지의 해표지증으로 진단받은 7명의 환자의 9예의 이환된 상지를 대상으로 하였다. 수술적 치료는 5명에 대하여 동반된 수부 기형에 대하여 시행하였다. 대상군의 의무기록 및 방사선 영상을 검토하여 그 특징을 살펴보았고 기존의 Frantz와 O'Rahilly 분류법 및 Tytherleigh-Strong과 Hooper에 의해 추가된 분류법 및 Goldfarb 등에 의해 제시된 분류법에 따라 각각 분류해보았다. 수술적 치료의 결과를 평가하기 위하여 맞섬 및 집기 기능의 향상 여부를 확인하였고, 부모들의 수술에 대한 만족도를 VAS (visual analog scale)를 이용하여 정량화 하였다. 결과: 태아기의 문제점이나 유전양식은 발견되지 않았다. 견관절은 1예를 제외하고는 모두 비정상적이었으며 주관절은 비정상적인 2예를 제외하고는 존재하지 않았으며 완관절 및 수부도 모든 예에서 기형을 보였으며 무지는 없거나 저형성되어 있었다. 편측이 이환된 경우에 비해 양측이 이환된 경우는 하지를 비롯한 다른 동반 기형을 가지는 차이를 보이고 있었다, 기존의 Frantz와 O'Rahilly 분류법으로 분류가 가능한 경우는 없었으며 Tytherleigh-Strong과 Hooper에 의한 추가 분류법으로는 type A가 1예, type B가 1예, type C가 7예에 해당되었다. Goldfarb 등에 의한 분류법으로는 proximal radial longitudinal dysplasia가 2예에 해당되었으며 proximal ulnar longitudinal dysplasia가 7예에 해당되었다. 무지화 수술을 시행한 3예에서는 수술 후 맞섬 및 손끝집기가 가능해졌으며, 합지증 분리술을 시행한 2예에서는 모두 수술 후 가쪽집기가 가능해졌다. 수술적 치료에 대한 부모의 만족도는 수술 후 평균 8.2였다. 결론: 해표지증은 편측 및 양측 이환 여부에 따라서 임상 양상의 차이를 보이고 있었다. 해표지증은 기존의 Frantz와 O'Rahilly의 분류에 의하여 단순 분류하기는 어려우며, 분류상 횡적 개재성 기형이 아닐 수 있다는 점을 시사한다. 수술적 치료는 팔 길이보다는 수부 기능을 향상시키기 위한 무지화 수술과 합지증 분리술을 통해 기능적 개선을 얻을 수 있었다. Purpose: Phocomelia is an extremely rare congenital anomaly of the upper extremity. There have been no clinical reports about phocomlia in Korea except for five birth reports. We present here the clinical features, classifications and surgical treatments of our phocomelia cases. Materials and Methods: From January 1993 to August 2007, seven patients were diagnosed as having phocomelia in 9 upper extremities at our clinic. Surgical treatments were performed for five patients on their hand anomalies. We retrospectively reviewed the medical records and radiographs of our cases, and we tried to classify them by the previously suggested systems. We evaluated the functional improvement and measured the VAS scale for parental satisfaction with the operative outcomes. Results: We could not find any problems during the fetal periods or any hereditary features. The bilaterally-affected patients also had deformities of the lower extremity, while the unilaterally-affected patients did not. We couldn't classify our cases according to the Frantz and O'Rahilly system. We found that the classifications suggested by Tytherleigh-Strong and Hooper (2003) and Goldfarb et al. (2005) could be promising alternatives for classification. One upper extremity was classified as type A, one as type B, and 7 as type C by Tytherleigh-Strong and Hooper's system. Using the Goldfarb's system, two upper extremities were classified as proximal radial longitudinal dysplasia, and seven were classified as proximal ulnar longitudinal dysplasia. Three patients who underwent pollicization showed opposition and tip pinch. Two patients who underwent syndactyly division could do lateral pinch. The VAS scale for parental satisfaction with the functional improvement averaged 8.2 postoperatively. Conclusion: The bilateral cases had different clinical features from unilateral ones. Phocomelia could not simply be classified by the Frantz and O'Rahilly system, and it may not be a true transverse intercalary deficiency. We could gain functional improvement after operations on the hand anomalies.

        Recombinant human granulocyte macrophage colony-stimulating factor (rhGM-CSF) could accelerate burn wound healing in hamster skin

        Burns are one of the most devastating forms of trauma and wound healing is a complex and multicellular process, which is executed and regulated by signaling networks involving numerous growth factors, cytokines, and chemokines. Recombinant human granulocyte macrophage colony-stimulating factor (rhGM-CSF) was specifically produced from rice cell culture through use of a recombinant technique in our laboratory. The effect of rhGM-CSF on promotion of deep second-degree burn wound healing on the back skin of a hamster model was evaluated through a randomized and double-blind trial. As macroscopic results, hamster skins of the experimental groups showed earlier recovery by new epidermis than the control groups. Immunohistochemical reactions of proliferating cell nuclear antigen and transforming growth factor-b1, which are indicators of cell proliferation, were more active in the experimental group, compared with the control group. On electron microscopy, basal cells in the epidermis of the experimental group showed oval nuclei, prominent nucleoli, numerous mitochondria and abundant free ribosomes. In addition, fibroblasts contained well-developed rough endoplasmic reticulum with dilated cisternae. Bundles of collagen fibrils filled the extracellular spaces. Particularly, ultrastructural features indicating active metabolism for regeneration of injured skin at 15 days after burn injury, including abundant euchromatin, plentiful free ribosomes, and numerous mitochondria, were observed. These findings suggest that use of rhGM-CSF could result in accelerated deep second-degree burn wound healing in animal models.

        주관절에 발생한 신경병성 관절병증

        목적: 주관절의 신경병성 관절병증은 매우 드문 질환으로 관절의 고유 감각을 상실하는 질환과 동반되어 무통성의 관절 파괴를 특징으로 하는 질환이다. 저자들은 주관절의 신경병성 관절병증 5예를 경험하여, 그 치료 결과를 문헌 고찰과 함께 보고하파 한다. 대상 및 방법: 주관절의 신경병성 관절병증으로 진단 받고 추시 중인 5명의 환자를 대상으로 하였다. 남자가 3명, 여자가 2명이었고, 평균 나이는 38.6세(범위: 13-50세)였으며, 기저 질환은 척수 공동증 3예, 척수 매독증 1예, 신경섬유종증 1예였다. 이들 중 1명은 보존적 치료를 시행하였고 4명은 수술적 치료를 시행하였다. 평균 추시 기간은 8.5년(범위: 1-19년)이었고, 최종 추시에서 술 전 증상과 운동 범위의 변화 및 주관절의 기능에 대한 주관적 만족도를 평가하였다. 결과: 다발성 관절증을 보인 1명에서는 특별한 치료 없이 주관절 기능을 유지하였다. 척골 신경 감압술을 시행한 1명과 주두 골절로 골유합술을 시행한 1명에서는 증상과 운동 범위가 호전되었으나 관절 성형술을 시행한 2명에서는 심부 감염이 합병되었고 술 후 운동 범위가 감소하였다. 결론: 주관절의 신경병성 관절병증은 관절의 심한 변형과 불안정성이 동반되지만 비교적 기능을 잘 유지하므로, 파괴되고 불안정한 관절의 교정보다는 주관절 변형에 의해 나타나는 이차적인 증상을 위주로 한 치료가 바람직하다고 생각한다. Purpose: Neuropathic arthropathy of the elbow is extremely rare and is characterized by painless destruction of the joint due to diminished proprioception. We report five cases of neuropathic arthropathy of the elbow with review of pertinent literature. Materials and Methods: We analyzed records of five patients who were diagnosed with neuropathic arthropathy of the elbow. There were three men and two women with a mean age of 38.6 years (range, thirteen to fifty years). The conditions associated with the arthropathy were three cases of syringomyelia, one tabes dorsalis, and one neurofibromatosis. One patient was conservatively treated and was followed periodically and others underwent operative procedures. The mean duration of follow-up was 8.5 years (range, one to nineteen years). Changes of symptoms, range of motion (ROM), and functional satisfaction were assessed. Results: The conservatively treated patient maintained the function of the elbow. In the surgically treated group, one case of ulnar nerve decompression and one case of osteosynthesis of the fractured olecranon showed improvement of symptoms, ROM, and function. But two patients who underwent reconstructive procedures had decreased ROM after complication of deep infection. Concluson: Neuropathic arthropathy of the elbow causes few functional problems despite of the radiologic destruction and instability of the elbow. Therefore symptomatic management is considered more preferable than surgical correction of destruction or instability of the elbow.

      • Achievements and Perspectives of GWAS Case Study in Rice Core Set

        In order to breakthrough upcoming challenges for the food production, the efficient use of rice germplasm would be a indispensible. These rice germplasm, adapted from diverse eco-systems, are undiscovered treasures for rice breeders/researchers, potentially providing a broad array of useful alleles that enrich gene pools of current cultivated rice varieties. Although growing ex-situ conservation efforts are an important for preserving diverse rice genetic resources, the activity on finding the novel and favorable genetic variants from the vast genebank collection is greatly challenging, requiring extensive screening processes. Therefore, rice core collection is a powerful solution to accelerate utilizations of the exotic germplasm of the entire population. In addition, The application of whole genome re-sequencing technology would establish a potent platform for fast forward genetic study, such as genome wide association study (GWAS). The GWAS has been implemented to efficiently identify candidate genes related to various useful agricultural traits in many crop species including rice. Given the significant associations between genetic variations and phenotypic diversity does not require prior knowledge, GWAS using high genome coverage of SNP markers provides a genomics platform to dissect previously unknown adaptive or other useful genetic variation accumulated in plant germplasm resources over the times. Once pinpointing candidate genes, GWAS allows informed choice of parents for QTL analysis based on the haplotype information, along with suggesting targets for following mutagenesis and transgenics. Here, we are to report our current achievements and perspectives from GWAS and post-GWAS undertaken to dissect and exploit useful alleles underlying many agricultural traits from Rice core set, including PHS (Pre-Harvest Sprouting), salt tolerance and disease resistance and so forth. Also, we will introduce the integrated Omics based GWAS case study using transcriptomes, proteomes, metabolomes and ionomes of our rice core set.
