      Transcriptomic analysis of zonula occludens-1 (ZO-1) knockout in ovarian cancer cell lines

      Background Zonula occludens-1 (ZO-1) is a crucial tight junction protein that regulates intercellular permeability and adhesion, thereby preserving the integrity of epithelial and endothelial barriers. ZO-1 is associated with tumorigenesis and the progression of epithelial–mesenchymal transition (EMT), invasion, and metastasis. In our previous study, knockout (KO) of ZO-1 using clustered regularly interspaced short palindromic repeats (CRISPR) reduced proliferation but increased migration and invasion, suggesting that ZO-1 may have a dual role. Therefore, this study aimed to elucidate the role of ZO-1 in ovarian cancer by analyzing transcriptomic changes associated with ZO-1. Objective This study aims to elucidate the impact of ZO-1 KO on gene expression in ovarian cancer cells by performing comparative RNA sequencing (RNA-seq) analysis on two distinct ZO-1 KO ovarian cancer cell lines, SKOV3 and SNU119. Methods ZO-1 was knocked out in SKOV3 and SNU119 cells using CRISPR-Cas9 technology. After identifying differentially expressed genes (DEGs) through RNA sequencing, Gene Ontology (GO) and pathway enrichment analyses were performed. The selected targets were subsequently validated using reverse transcription quantitative PCR (RT-qPCR) and Western blot analysis to assess both transcript- and protein-level expression changes. Results Transcriptomic analysis revealed over 400 DEGs in each cell line. Of these, 14 genes were consistently upregulated in both cell lines, while 24 genes were consistently downregulated. The common DEGs were visualized using a heatmap, and a subset of these genes was further validated by RT-qPCR and Western blot analyses. TGFB2 expression was consistently altered at both the mRNA and protein levels following ZO-1 KO in both cell lines. Similar expression patterns were observed for THBS1, VCAN, ITGB8, SEMA3A, and GAS6. The concordant changes observed in transcriptomic and protein analyses suggest a consistent association between ZO-1 KO and TGFB2 expression. Conclusion ZO-1 KO in ovarian cancer cells induces substantial transcriptional reprogramming, particularly affecting genes associated with extracellular matrix organization and signaling pathways. Multiple candidate genes showed consistent alterations at both the mRNA and protein levels, supporting the robustness of the observed transcriptional changes. These findings provide a framework for understanding ZO-1–associated regulatory networks in ovarian cancer.       선종(禪宗)의 교단(敎壇) 성립에서 『능가경(楞伽經)』의 공능(功能)

      현대의 세계불교에서 선불교의 중추는 인도에서 중국으로의 전래를 넘어 한국에서 위상을 담당한다고 본다. 중국의 선불교를 받아들이는 과정에서 여러 한국 禪門들이 성립되었는데, 어느 하나 맹목적이지 않고 계승을 넘은 발전을 이루었다. 모든 면에서 그렇지는 않았더라도 지속적인 연구와 수행을 통해 다듬어졌고, 窮極을 위한 禪觀의 ‘반야사상’을 중심으로 그 점검과 修禪으로 교육해 온 점은 오히려 후대에 고착되고 있다. 수승함이라는 측면에서 선사상의 수준이 높아졌지만 교육학적 눈높이는 점차 배제되었고, 이는 현대 선불교를 중심으로한 한국불교의 불자 감소에 영향을 준 간접적 요소라 판단된다. 언제나 붓다의 설법은 중생에 맞추어 설해졌고, 그 입문의 효과는 여러 아라한의 증득으로 증명되었다. 중국 선불교의 여러 祖師와 각 사상의 법맥을 계승하되 그 수준은 더욱 높여 가장 수준 높은 선사상을 보유했지만, 후학의 제도를 점차 초점에서 예외로 치부해서는 안된다. 출ㆍ재가를 막론하고 불교 입문과 수행을 위한 첫 발심에서 ‘禪’을 통한 大悟를 추구하고자 한다면 그 단계는 더욱 공고히 해야 할 과제일 것이다. 본고는 초기 선종 교단의 성립 과정에서 크게 역할을 하였으나 여러 이유로 점차 이원화된 禪經으로 배태된 『금강경』에 비해 소외된 『능가경』의 習禪이 주효한 단계적 기능을 해줄 것이라는 추론에서 착안한 연구다. 과거 맥락과 사상을 살펴봄으로써 우선 그 원인과 결과를 밝히고, 작금 현대불교에서 기대되는 역할과 가능성을 제시하고 방향성을 모색하였다. In modern world Buddhism, the nucleus of Seon(禪) Buddhism is believed to have the status of being in charge of Korea beyond the transition from India to China. In the process of accepting Seon(禪) Buddhism in China, many Korean civilizations were established, and none of them were blind and developed beyond succession. In all respects, although it has been refined through continuous study and practice it is rather entrenched in later generations that it has been trained around the view of the Seon(禪)'s thought Prajñā-Paramitā(般若) for extremity. The level of outstanding enlightenment has increased, but pedagogical consideration has been gradually reduced. This is an indirect factor that influenced the decline of Buddhism in Korea, centered on modern Seon(禪) Buddhism. Buddha has always taught at the level of people, and the effect has been proven to be many Arahat. While inheriting various teachers of Zen Buddhism in China and the legal context of each ideology, the level was raised further to hold the highest level of prehistory. However, the system of junior studies should not be removed from the focus. Regardless of monks and believers, it is necessary to further systematize to pursue the great realization of "Seon(禪)" in the first trial for introduction and practice of Buddhism. This study was based on the reasoning that Laṅkāvatāra(楞伽)-sūtra, which played a significant role in the early establishment of Zen Buddhism, would play a major step-by-step role in the learning process, which was gradually dismissed as a dualized scripture for various reasons. By examining the context and ideas of the past, I first identified the cause and effect, and further sought the direction by suggesting whether I could play a new role and possibility in modern Buddhism.

      Evaluation of the Efficacy, Safety, and Treatment-Related Skin Irritation of Adapalene-Benzoyl Peroxide Combination Gel and Adapalene Gel in Acne Patients: A Randomized Comparative Study

      Background: While adapalene and benzoyl peroxide have both been demonstrated to be effective for the treatment of acne vulgaris, they can also cause skin irritation, resulting in patient incompliance. In addition, the irritation may be severe when adapalene is co-administered with benzoyl peroxide. Objective: The intention of this study was to evaluate the efficacy and safety of adapalene 0.1%-benzoyl peroxide 2.5% fixed-dose combination gel, and adapalene 0.1% gel monotherapy, for the treatment of mild to moderate acne. Any changes in the skin barrier function were also measured. Methods: A total of 52 patients applied adapalene-benzoyl peroxide gel or adapalene gel once daily. The changes in lesion counts, investigator.s global assessment, patient satisfaction, and adverse events were recorded for 6 weeks. The biophysical profiles of the skin, including transepidermal water loss and redness, were measured for 3 weeks. Results: Both agents were effective for the treatment of acne lesions. Adapalene-benzoyl peroxide showed a faster onset of action than adapalene, especially on inflammatory lesions. However, at week 6, both agents showed a similar effect in reducing lesion counts. While most patients in both groups experienced skin irritation from week 1, this gradually reduced. Likewise, the changes in transepidermal water loss and redness showed peak rates at week 1 and 2, but decreased over time. Conclusion: Adapalene-benzoyl peroxide gel is effective for the treatment of acne with a safety profile comparable to adapalene gel. The skin irritation experienced may be associated with the changes in skin barrier disruption, which lasts for 1.2 weeks.

      Case Report : Cheilitis Granulomatosa Successfully Treated with Minocycline

      Cheilitis granulomatosa, a rare disease of unknown cause, is characterized clinically by recurrent or persistent swelling of the lip. Histopathologically, the dermis contains non-necrotizing granuloma, lymphangiectasia, and perivascular lymphocytic infiltration. Treatment of cheilitis granulomatosa is difficult, due to its unknown etiology. Several therapeutic methods have been attempted, including systemic and intralesional corticosteroids, oral metronidazole, non-steroidal anti-inflammatory drugs (NSAIDs), hydroxychloroquine, and surgical reduction, but unfortunately no treatment method has yet proven reliable.
