Ginsenoside Rb1 exerts neuroprotective effects through regulation of Lactobacillus helveticus abundance and GABAA receptor expression

        Huimin Chen,Jiajia Shen,Haofeng Li,Xiao Zheng,Dian Kang,Yangfan Xu,Chong Chen,Huimin Guo,Lin Xie,Guangji Wang,Yan Liang 고려인삼학회 2020 Journal of Ginseng Research Vol.44 No.1

Background: Ginsenoside Rb1 (Rb1), one of the most abundant protopanaxadiol-type ginsenosides, exerts excellent neuroprotective effects even though it has low intracephalic exposure. Purpose: The present study aimed to elucidate the apparent contradiction between the pharmacokinetics and pharmacodynamics of Rb1 by studying the mechanisms underlying neuroprotective effects of Rb1 based on regulation of microflora. Methods: A pseudo germ-free (PGF) rat model was established, and neuroprotective effects of Rb1 were compared between conventional and PGF rats. The relative abundances of common probiotics were quantified to reveal the authentic probiotics that dominate in the neuroprotection of Rb1. The expressions of the gamma-aminobutyric acid (GABA) receptors, including GABAA receptors (α2, β2, and γ2) and GABAB receptors (1b and 2), in the normal, ischemia/reperfusion (I/R), and I/RþRb1 rat hippocampus and striatum were assessed to reveal the neuroprotective mechanism of Rb1. Results: The results showed that microbiota plays a key role in neuroprotection of Rb1. The relative abundance of Lactobacillus helveticus (Lac.H) increased 15.26 fold after pretreatment with Rb1. I/R surgery induced effects on infarct size, neurological deficit score, and proinflammatory cytokines (IL-1β, IL-6, and TNF-α) were prevented by colonizing the rat gastrointestinal tract with Lac.H (1×109 CFU) by gavage 15 d before I/R surgery. Both Rb1 and Lac.H upregulated expression of GABA receptors in I/R rats. Coadministration of a GABAA receptor antagonist significantly attenuated neuroprotective effects of Rb1 and Lac.H. Conclusion: In sum, Rb1 exerts neuroprotective effects by regulating Lac.H and GABA receptors rather than through direct distribution to the target sites.

A Type of Subsection Model for a Permanent Magnet Bar and its Leakage Permeance Calculation Method in an Open Magnetic Circuit

        Huimin Liang,Jiaxin You,Wenying Yang,Guofu Zhai 한국자기학회 2014 Journal of Magnetics Vol.19 No.1

The equivalent model of a permanent magnet (PM) plays an important role in electromagnetic system calculation. A type of subsection model for a PM bar is established, to improve the accuracy of the traditional equivalent circuit method. The mathematical expression, and its end verification condition, are presented. Based on the analytical method and finite element method, the leakage permeance calculation of a PM bar in an open magnetic circuit is investigated. As an example, for a given certain type of PM bar, the magnetic flux of each section is validated by experiment, and by simulation. This model offers a foundation for building a high accuracy equivalent magnetic PM model in an electromagnetic system.

In vitro differentiation of spermatogonial stem cells using testicular cells from Guangxi Bama mini-pig

        Huimin Zhao,Junyu Nie,Xiangxing Zhu,Yangqing Lu,Xingwei Liang,Huiyan Xu,Xiaogan Yang,Yunkai Zhang,Kehuan Lu,Shengsheng Lu 대한수의학회 2018 Journal of Veterinary Science Vol.19 No.5

In this study, we attempted to establish a culture system for in vitro spermatogenesis from spermatogonial stem cells (SSCs) of Bama mini-pig. Dissociated testicular cells from 1-month-old pigs were co-cultured to mimic in vivo spermatogenesis. The testicular cells were seeded in minimum essential medium alpha (a-MEM) supplemented with Knockout serum replacement (KSR). Three-dimensional colonies formed after 10 days of culture. The colonies showed positive staining for SSC-associated markers such as UCHL1, PLZF, THY1, OCT4, Dolichos biflorus agglutinin, and alkaline phosphatase. Induction of SSCs was performed in a-MEM + KSR supplemented with retinoic acid, bone morphogenetic protein 4, activin A, follicle-stimulating hormone, or testosterone. The results showed that STRA8, DMC1, PRM1, and TNP1 were upregulated significantly in the colonies after induction compared to that in testis from 1-month-old pigs, while expression levels of those genes were significantly low compared to those in 2-month-old testis. However, upregulation of ACROSIN was not significant. Replacement of a-MEM and KSR with Iscove’s modified Dulbecco’s medium and fetal bovine serum did not upregulate expression of these genes significantly. These results indicate that SSCs of Bama mini-pig could undergo differentiation and develop to a post-meiotic stage in α-MEM supplemented with KSR and induction factors.

Rehmannioside D mitigates disease progression in rats with experimental-induced diminished ovarian reserve via Forkhead Box O1/KLOTHO axis

        Yan Liang,Huimin Wang,Jin Chen,Lingyan Chen,Xiaoyong Chen 대한약리학회 2023 The Korean Journal of Physiology & Pharmacology Vol.27 No.2

This study aims to explore the impact of Rehmannioside D (RD) on ovarian functions of rats with diminished ovarian reserve (DOR) and its underlying mechanisms of action. A single injection of cyclophosphamide was performed to establish a DOR rat model, and fourteen days after the injection, the rats were intragastrically administrated with RD for two weeks. Rat estrus cycles were tested using vaginal smears. Ovarian tissues were histologically evaluated, the number of primordial, mature, and atretic follicles was calculated, and the apoptotic rate of granulosa cells. Follicle-stimulating hormone (FSH), luteinizing hormone (LH), and estradiol (E2) levels were determined by ELISA assays. Protein levels of Forkhead Box O1 (FOXO1), KLOTHO, Bcl-2, and Bax were investigated in ovarian tissues of DOR rats. The binding between FOXO1 and KLOTHO was verified by ChIP assay. High-dose administration of RD into DOR rats improved their estrus cycles, increased ovarian index, enhanced the number of primordial and mature follicles, reduced the number of atretic follicle number, and ovarian granulosa cell apoptosis in addition to inhibiting FSH and LH levels and upregulating E2 expression. FOXO1 and KLOTHO were significantly suppressed in DOR rats. FOXO1 knockdown partially suppressed the protective effects of RD on DOR rats, and KLOTHO overexpression could restore RD-induced blockade of DOR development despite knocking down FOXO1. FOXO1 antibody enriched KLOTHO promoter, and the binding between them was reduced in DOR group compared to that in sham group. RD improved ovarian functions in DOR rats and diminished granulosa cell apoptosis via the FOXO1/KLOTHO axis

      • In Situ Assembly of Antifouling/Bacterial Silver Nanoparticle-Hydrogel Composites with Controlled Particle Release and Matrix Softening

        Baek, Kwanghyun,Liang, Jing,Lim, Wan Ting,Zhao, Huimin,Kim, Dong Hyun,Kong, Hyunjoon American Chemical Society 2015 ACS APPLIED MATERIALS & INTERFACES Vol.7 No.28

<P>Controlling bacterial contamination has been a major challenge for protecting human health and welfare. In this context, hydrogels loaded with silver nanoparticles have been used to prevent biofilm formation on substrates of interest. However, such gel composites are often plagued by rapid loss of silver nanoparticles and matrix softening, and thus the gel becomes less effective for antifouling. To this end, this study demonstrates that in situ photoreaction of an aqueous mixture of silver nitrates, poly(ethylene glycol) diacrylate, and vinylpyrrolidone results in a silver nanoparticle-laden hydrogel composite with minimal nanoparticle loss and matrix softening due to enhanced binding between nanoparticles and the gel. The resulting gel composite successfully inhibits the bacterial growth in media and the bacterial adhesion to surfaces of interest. We suggest that the results of this study serve to advance quality of materials with antifouling/bacterial activities.</P><P>An advanced, antifouling/bacterial hydrogel composite is structurally stable and sustainably sequesters encapsulated silver nanoparticles. A Non-Linear Permanent Magnet Working Point Migration Model and its Application to Simulation of a Polarized Magnetic System

        Bo Li,Huimin Liang,Jiaxin You,Fangyuan Xiong,Jan K. Sykulski 한국자기학회 2019 Journal of Magnetics Vol.24 No.3

The paper addresses the issue of the working point migration in non-linear permanent magnets (PM). Starting from the considerations of energy, a novel working-point migration (WPM) model is proposed which can be incorporated into a magnetic equivalent circuit (MEC). The basic theory of working-point migration is discussed which focus on the second quadrant of the magnetic hysteresis loop. The calculation method of the WPM model is proposed based on the relationship between the recoil line model and the affine transformation hysteresis loop. The establishment method for the resultant working point is described by combining the demagnetization curve and the recoil line models in the minor hysteresis loop. The static characteristic of a bistable polarized magnetic system (BPMS), as used in actuators, is calculated using the magnetic circuit method based on the WPM, while a finite element model (FEM) is also derived. The calculation method for the WPM used in MEC is also discussed. The WPM based MEC model yields reasonable results, compared with FEM, of the latching force but with much faster calculation speeds. Furthermore, the working-point state of the PM is clearly illustrated. The test system of the BPMS prototype is established. The test data and WPM model calculation results are compared. It is shown that the WPM model provides accurate prediction of static characteristics of an electromagnetic system.

      • Order Imbalances in Options and Volatility Risk Premium on Equity Index

        Chin-Ho Chen,Huimin Chung,Wen-Liang G. Hsieh,Shu-Fang Yuan 한국재무학회 2011 한국재무학회 학술대회 Vol.2011 No.09

This study explores the impact of aggregate daily order imbalances in options on volatility risk premium on its underlying index. Two types of volatility risk premium, ex-post and ex-ante volatility risk premiums, are included to address this issue. Based on Taiwan stock index and options (TXO) data over the period from January 1, 2005 to December 31, 2009, several interesting results emerge. First, order imbalances in near-month options, especially for call options’, have a predominant influence on ex-post and ex-ante volatility risk premiums. Next, order imbalances of near-month options in either direction, excess buy orders or excess sell order, have distinct effect on volatility risk premium. Overall, the increased level of excess buy orders drives ex-post and ex-ante volatility risk premiums upward, but downward only for excess sell orders in call options. Finally, ex-post and ex-ante volatility risk premiums are positively related to order imbalances of near-month options in respond to continuous price variations. Nonetheless, there is no significant relation for index price jumps.

Effect of Postmortem Phases on Lamb Meat Quality: A Physicochemical, Microstructural and Water Mobility Approach

        Yue Ge,Dequan Zhang,Huimin Zhang,Xin Li,Fei Fang,Ce Liang,Zhenyu Wang 한국축산식품학회 2021 한국축산식품학회지 Vol.41 No.5

To investigate the effect of postmortem phases on lamb meat quality, the physicochemical quality, microstructure and water mobility of oyster cut, short loin, knuckle and silverside muscles from Small-Tail Han sheep were evaluated in the pre-rigor, rigor mortis and post-rigor phases. Pre-rigor lamb meat had higher pH and water holding capacity (WHC), whereas lower CIE L*, b*, hue angle values than rigor mortis and postrigor meat (p<0.05). The Warner-Bratzler shear force (WBSF) values were higher in rigor mortis short loin and silverside than their pre-rigor and post-rigor counterparts, pre-rigor short loin had lower WBSF value than its post-rigor counterpart (p<0.05). Muscle fibers shrank laterally and longitudinally during the onset of rigor mortis. Rigor mortis and postrigor lamb meat exhibited wide I-bands, dark A-bands, short sarcomeres and large intermyofibrillar spaces. The shift of immobilized water to free water and repulsion from the intra-myofibrillar space to the extracellular space result in the increase of water loss in rigor mortis and post-rigor lamb meat. The results of the principal component analysis (PCA) indicated that rigor mortis and post-rigor lamb meat had similar quality properties but different from pre-rigor lamb meat. In conclusion, the lamb meat in the pre-rigor phase had good tenderness, color and WHC. The results of this research could provide some theoretical references for lamb meat production and processing.
