        당밀의 첨가가 갈근탕박 사일리지의 품질과 산양의 기호성에 미치는 영향

        Two experiments were conducted to investigate the effects of molasses supplementation to silage materials in ensiling Galgeuntang (herbal medicine) meal on the quality of silage, and its palatability in Korean native goats. In experiment Ⅰ, molasses was added at the levels of 0, 0.5, and 1.0% to Galgeuntang meal with three replicates per treatment. In experiment Ⅱ, three 1.5-yr-old Korean native goats were used to measure palatability. In experiment Ⅰ, lactic acid contents in molasses treatments were significantly(p<0.05) higher, and pH and butyric acid contents were lower than those of non-molasses treatment. In addition, molasses treatment increased total microbial cell counts in MRS medium for lactobacillus, but decreased total microbial cell counts in PDA medium for fungi. Molasses supplementation to silage materials increased in vitro dry matter disappearance. Molasses supplementation tended to increase silage intake in Korean native goat (experiment Ⅱ), but the difference was not significant(p>0.05). It is concluded that molasses supplementation to silage materials in ensiling herbal medicine meal could improve its preservability and palatability, the higher the better.

      • 나프록센 함유 방출제어형 패취의 제제설계 및 평가

        The purpose of this study is to prepare he controlled release adhesive patch containing naproxen. Pressure-sensitive adhesive (PSA)-type patch was fabricated by casting of polyisobutylene (PIB) and mineral oil in toluene. Membrane-controlled release (MCR)-type patch was prepared by the attachment of the controlled release membrane on the PSA-type patch. The membrane was mainly composed of Eugragit. polyethylene glycol(PEG) and glycerin. The drug release profile and skin permeation test with various patches were evaluated in vitro. The release of naproxen from PIB-based PSA-type patch with various loading dosed fitted Higuchi's diffusion equation. However, the permeation of naproxen through hairless mouse skin from PSA-type patch followed zero-order kinetics. In MCR-type patch, thickness of controlled release membrane affected on the drug release rate highly. In the composition of membrane the release rate was decreased as the ratio of Eudragit increased. The drug release from the MCR-type patch followed zero order kinetics. The permeation of naproxen through hairless mouse skin from MCR-type patch showed lag time for the intial release period and didn't fit the zero-order kinetics.

        On the Assessment of Compressibility Effects of Two-Equation Turbulence Models for Supersonic Transition Flow with Flow Separation

        An assessment of two-equation turbulence models, the low Reynolds k-${\varepsilon}$ and k-${\omega}$ SST models, with the compressibility corrections proposed by Sarkar and Wilcox, has been performed. The compressibility models are evaluated by investigating transonic or supersonic flows, including the arc-bump, transonic diffuser, supersonic jet impingement, and unsteady supersonic diffuser. A unified implicit finite volume scheme, consisting of mass, momentum, and energy conservation equations, is used, and the results are compared with experimental data. The model accuracy is found to depend strongly on the flow separation behavior. An MPI (Message Passing Interface) parallel computing scheme is implemented.

        정자 점자의 사용에 관한 일 연구

        시각장애 아동에게 있어 점자는 그중요성을 아무리 강조하더라도 지나치지 않을 만큼, 지난 150여 년 동안 이들의 읽기, 쓰기에서 중추적인 역할을 담당해왔다. 프랑스에서 처음 개발된 점자가 영어권으로 옮겨오면서, 약점자의 필요성이 대두되었으며, 현재 189개의 약점자를 공식적으로 사용하고 있다. 초기 읽기, 쓰기 학습에 있어, 약점자를 어떻게 아동에게 지도할 것인가는 지금까지 교제 제작의 편의성이라는 문제와 연관되면서, 교육의 효율성 측면에서보다는 사용 교제의 약점자 사용 형태에 따라 결정된 것이 사실이다. 즉, 1950년대에 대부분의 점자 출판물 제작 업체들이 약점자를 사용한 교제만을 제작함으로서 교사들은 약점자를 초기 점자 학습에서 부터 지도하도록 강요되어 온 것이 미국의 현실이다. 본 연구는, 16명의 점자를 읽는 아동 중에서 8명은 약점자부터 배우는 집단, 나머지 8명을 정자 점자부터 배운 집단으로 나누어 이들 집단 사이에 읽기, 쓰기에서의 수행 능력 차이가 있는가를 검사한 결과를 보고한 것이다. 연구 결과, 아동은 어떤 형태의 점자를 초기에 학습하였더라도 읽기, 쓰기에서 수행 능력상 별다른 차이를 보이지 않았음이 밝혀졌다. 대다수의 시각장애 아동이, 시각 이외의 기타 중복 장애를 갖고 있는 미국 시각장애 교육의 현실을 참작할 때, 정자 점자를 초기에 교육하는 것은 보다 많은 시각장애 아동의 능동적 학습에 도움이 될 것이며, 컴퓨터와 기타 보조 공학 장치를 사용한 점자 출판이 보다 용이하여진 현실에서, 초기 학습에서의 점자는 교제의 제작 편의성과는 분리하여 고려되어야할 사항임을 문헌연구와 실험 연구를 통하여 밝혔다.

        Extracts from Rhizopus oryzae KSD-815 of Korean Traditional Nuruk Confer the Potential to Inhibit Hypertension, Platelet Aggregation, and Cancer Metastasis in vitro

        Rhizopus oryzae KSD-815 was isolated from nuruk that has been used to make Korean traditional wines. This study was performed to investigate the effect of cultures of R. oryzae KSD-815 on cardiovascular disorders and cancer metastasis. Firstly, these cultures were sequentially fractionationed with n-hexane (TAHe), ethylacetate (TAE), n-butanol (TAB), and H₂O (TAW). The TAE inhibited the activity of angiotensin-converting enzyme (ACE) and TAB suppressed platelet aggregation in vitro. TAE and TAB inhibited cell motility of human breast cancer cells. Furthermore, TAW interrupted the formation of neovasculature and tube-like structure, and down-regulated the expression of angiogenic factors, basic fibroblast growth factor (bFGF), tumor necrosis factor-α (TNF-α), and hypoxia-inducible factor-1α (HIF-1α) in breast cancer cells. These results indicated that cultures of R. oryzae KSD-815 display the inhibitory activities on hypertension, platelet aggregation, and metastasis, and suggest that these cultures might be further probed for the purposes as therapeutic agents or dietary supplements.

      • Unemployment and Underemployment of people with visual impairments : Literature review on recent trends and issues

        Unemployment and underemployment issues for people with visual impairments have long been recognized by practitioners and professionals in the field as one of the primary factors in fully enjoying independent life in this society. In spite of continuous effort devoted to the topic, immediate improvement for employment rate and status has not been observed. By surveying recent literature, this study was aimed at reviewing topical development on this subject, observing current trends and issues, and itemizing major findings reported in various studies. Fifteen recent articles related to employment of people with visual impairments were reviewed and were analyzed.

        Performance characteristics of a pintle nozzle using the conformal sliding mesh technique

        <P><B>Abstract</B></P> <P>Numerical and non-linear internal ballistics analyses are conducted to investigate the performance characteristics of a pintle reciprocating in a nozzle. A conformal sliding mesh is successfully implemented to maintain smooth and continuous flow properties at the boundary of a moving pintle block. The proper mesh size is determined via a grid dependency test. The pintle nozzle operates under over-expansion conditions so that flow separation occurs at the nozzle wall at certain pintle stroke positions. Both the chamber pressure and thrust have hysteresis loci to allow for the reciprocation of the pintle's insertion and extraction motions, which have different trace-to-pintle speeds. The hysteresis loci reduce as the pintle speed increases. The thrust coefficient (rather than the thrust itself) is able to precisely determine the effects of the nozzle flow separation on overall performance.</P>

      • Limiting Dilution Assay를 이용한 Long-term Culture-Initiating Cells의 정량

        연구배경: LTC-IC는 5주간의 장기액상배양 후에도 집락을 형성할 수 있는 세포를 말하며 조혈모세포의 이식능을 가장 잘 대변할 수 있는 세포이다. 본 연구에서는 Limiting Dilution Assay를 통해 LTC-IC를 정량하고자 하였다. 대상 및 방법: 제대혈, 가동화된 말초혈 및 골수형을 대상으로 일정수의 단핵구에 포함된 CD34^(+) 세포수, CD34^(+)/38- 세포수, 단기배양시 형성된 집락의 수 및 LTC-IC의 수를 구하였다. LTC-IC의 수는 96-well plate에서의 장기액상배양 및 Poisson 분포를 이용한 limiting dilution assay를 이용하였다. 결과: 10^(5)개의 단핵구에 포함된 CD34^(+) 세포, CD34^(+)/38^(-) 세포수는 제대혈에서 960±493, 43.0±20.0, 가동화된 말초혈에서 720±432, 42.5±18.8, 골수혈에서 2,505±805, 805±305였고, CFU-GM의 수는 제대혈에서 24.6±9.9, 가동화된 말초혈에서 20.7±13.0이었고, LTC-IC의 수는 제대혈에서 10.4±5.7, 가동화된 말초혈에서 5.8±5.3, 골수혈에서 13.9±4.2였다. 결론: 96-well plate에서의 장기액상배양 및 Poisson 분포를 이용한 limiting dilution assay를 이용하여 성공적으로 LTC-IC의 수를 정량하였고 이를 통해 이식후의 장기생착능에 대한 정보를 얻을수 있다. Background: Long-term culture-initiating cells(LTC-IC) are defined as those cells responsible for the presence of clonogenic progenitors in 5-week-old long-term culture. LTC-IC are the best available approximation of stem cells in human. we compared the number of LTC-IC in cord blood(CB), mobilized peripheral blood(PB), and bone marrow(BM) using limiting dilution assay(LDA). Methods: Samples were obtained from CB, mobilized PB, and BM. Mononuclear cells(MNC) and CD34^(+) cells were separated by ficoll-hypaque centrifugation and method using immunomagnetic beads, respectively. Flow cytometric analysis was done for determination of CD34^(+) and CD34^(+)/38 cells. Clonogenic progenitors were assayed in methylcellulose media. For LDA, MNCs or CD 34^(+) cells were seeded onto irradiated murine stromal feeder layer in 96-well plate at 3~5 different cell concentrations. Wells that did not contain cobblestone area after 5 weeks of culture were counted and the number of LTC-IC was determined using Poisson distribution. Results: The number of CD34^(+) and CD34^(+)/38^(+) cells per 10^(5) MNCs in CB, PB, and BM were 960±493, 43.0±20.0, 720±432, 42.5±18.8, and 2,505±805, 805±305, respectively. The number of CFU-GM per 10^(5) MNCs in CB and PB were 24.6±9.9 and 20.7±13.0, respectively. The number of LTC-IC per 10^(5) MNCs in CB, PB, and BM were 10.4±5.7, 5.8±5.3, and 13.9±4.2, respectively. Conclusion: In this report, we successfully determined LTC-IC frequencies using LDA with long-term culture system. Determination of LTC-IC frequencies is useful for measurement of long-term in vivo repopulating potential.
