        Hydro-mechanical interaction of reinforced concrete lining in hydraulic pressure tunnel

        The reinforced concrete lining of hydraulic pressure tunnels tends to crack under high inner water pressure (IWP), which results in the inner water exosmosis along cracks and involves typical hydro-mechanical interaction. This study aims at the development, validation and application of an indirect-coupled method to simulate the lining cracking process. Based on the concrete damage plasticity (CDP) model, the utility routine GETVRM and the user subroutine USDFLD in the finite element code ABAQUS is employed to calculate and adjust the secondary hydraulic conductivity according to the material damage and the plastic volume strain. The friction-contact method (FCM) is introduced to track the lining-rock interface behavior. Compared with the traditional node-shared method (NSM) model, the FCM model is more feasible to simulate the lining cracking process. The number of cracks and the reinforcement stress can be significantly reduced, which matches well with the observed results in engineering practices. Moreover, the damage evolution of reinforced concrete lining can be effectively slowed down. This numerical method provides an insight into the cracking process of reinforced concrete lining in hydraulic pressure tunnels.

        Pipe-Soil Interaction and Sensitivity Study of Large-Diameter Buried Steel Pipes

        Large-diameter buried steel pipes (BSPs) have been widely utilized in water diversion and hydropower fields. This paper presents a comprehensive numerical investigation on the interaction between the large-diameter pipe and soil under three typical working conditions, to capture the structural mechanical behaviors of pipe deformation, soil displacement, and soil pressure. Furthermore, a parametric analysis of pipe diameter is conducted, along with the sensitivity study of soil parameters based on the orthogonal test method. The results show that the water weight increases the pipe deformation significantly by 14%, while the high internal water pressure decreases the deformation greatly with the maximum effect of 39%. The pattern of soil pressure at the top of the pipe changes from parabola to double-hump as the pipe diameter varies from 0.5 m to 5.0 m. The pipe deformation and soil pressure keep increasing with the pipe diameter, but the safety margin for prism load decreases gradually. Pipe deformation and soil pressure are very sensitive to the elastic modulus and Poisson's ratio of backfill. Friction coefficients of soil-soil (trench sidewall) and pipe-soil have great influences on the soil pressure, while the bedding material and backfill cohesion have small effects on it.

        Cubic time-spline fitting and interpolation for five-axis CNC machining

        In CNC machining, G01 codes are widely used to represent the tool path. Directly interpolating these G01 codes is time-consuming and may cause discontinuities. In this paper, we propose a time-spline curve fitting method that combines tool path fitting and feedrate scheduling into a single step for five-axis CNC machining. The input for this method consists of a three-dimensional linear path of the tool tip in the workpiece coordinate system and two-dimensional tool orientations in the machine coordinate system (MCS). The output is a fitted tool path in the MCS represented by a five-dimensional smooth time-parametric B-spline curve, simply referred to as the time-spline curve. The time-spline curve provides not only position information but also kinematic information, including velocity, acceleration, and jerk for each axis, directly derived from the first, second, and third derivatives of the curve. To meet fitting error constraints and axial kinematic constraints, our objective is to find the time-spline curve that is time-optimal. We formulate the optimization problem as a nonlinear optimization model and design a recursive algorithm to solve it. The resulting time-spline curve demonstrates high accuracy and fully utilizes the machine’s kinematic capabilities. Along the tool path defined by the time-spline curve, exact interpolation points can be straightforwardly obtained according to the interpolation period. Simulations and experimental results indicate that the proposed method yields a time-optimal time-spline curve with the desired precision and kinematic constraints.

        Low-stiffness spring element constraint boundary condition method for machining deformation simulation

        The 3-2-1 constraint principle has been widely applied as the boundary conditions for the finite element method (FEM) to simulate machining deformation of aerospace structure components. However, this principle is inconsistent with the actual contact surface between workpieces and worktables because it provides only three constraint points. These points have rarely been studied in terms of positions and distances. In addition, the applicability of the principle is limited for the workpiece with geometric centers difficult to find or centers without nodes. Therefore, this study proposed a new boundary condition method, low-stiffness spring element constraint method (SECM), drawing on FEM theories and related mechanic theories. With the method proposed this study established the FE model of machining deformation, and then compared the simulation results with both the analytical results and the experimental results of milling and deformation measurements. Good agreement is found between the three results. Finally, this study examined the effect of the three constraint points on simulation of the 3-2-1 principle in terms of point positions and distances. A comparison between SECM and the 3-2-1 principle revealed that SECM is closer to the actual working conditions and more reliable with wider application, which suggests that SECM can replace the 3-2-1 principle as the boundary conditions for the workpiece.

      • A meta-analysis of genome-wide association studies for adiponectin levels in East Asians identifies a novel locus near <i>WDR11-FGFR2</i>

        <P>Blood levels of adiponectin, an adipocyte-secreted protein correlated with metabolic and cardiovascular risks, are highly heritable. Genome-wide association (GWA) studies for adiponectin levels have identified 14 loci harboring variants associated with blood levels of adiponectin. To identify novel adiponectin-associated loci, particularly those of importance in East Asians, we conducted a meta-analysis of GWA studies for adiponectin in 7827 individuals, followed by two stages of replications in 4298 and 5954 additional individuals. We identified a novel adiponectin-associated locus on chromosome 10 near <I>WDR11-FGFR2</I> (<I>P</I> = 3.0 × 10<SUP>−14</SUP>) and provided suggestive evidence for a locus on chromosome 12 near <I>OR8S1-LALBA</I> (<I>P</I> = 1.2 × 10<SUP>−7</SUP>). Of the adiponectin-associated loci previously described, we confirmed the association at <I>CDH13</I> (<I>P</I> = 6.8 × 10<SUP>−165</SUP>), <I>ADIPOQ</I> (<I>P</I> = 1.8 × 10<SUP>−22</SUP>), <I>PEPD</I> (<I>P</I> = 3.6 × 10<SUP>−12</SUP>), <I>CMIP</I> (<I>P</I> = 2.1 × 10<SUP>−10</SUP>), <I>ZNF664</I> (<I>P</I> = 2.3 × 10<SUP>−7</SUP>) and <I>GPR109A</I> (<I>P</I> = 7.4 × 10<SUP>−6</SUP>). Conditional analysis at <I>ADIPOQ</I> revealed a second signal with suggestive evidence of association only after conditioning on the lead SNP (<I>P</I><SUB>initial</SUB> = 0.020; <I>P</I><SUB>conditional</SUB> = 7.0 × 10<SUP>−7</SUP>). We further confirmed the independence of two pairs of closely located loci (<2 Mb) on chromosome 16 at <I>CMIP</I> and <I>CDH13</I>, and on chromosome 12 at <I>GPR109A</I> and <I>ZNF664</I>. In addition, the newly identified signal near <I>WDR11-FGFR2</I> exhibited evidence of association with triglycerides (<I>P</I> = 3.3 × 10<SUP>−4</SUP>), high density lipoprotein cholesterol (HDL-C, <I>P</I> = 4.9 × 10<SUP>−4</SUP>) and body mass index (BMI)-adjusted waist–hip ratio (<I>P</I> = 9.8 × 10<SUP>−3</SUP>). These findings improve our knowledge of the genetic basis of adiponectin variation, demonstrate the shared allelic architecture for adiponectin with lipids and central obesity and motivate further studies of underlying mechanisms.</P>

        Comparison of the Distribution Pattern of 21-Gene Recurrence Score between Mucinous Breast Cancer and Infiltrating Ductal Carcinoma in Chinese Population: A Retrospective Single-Center Study

        Purpose This retrospective study aimed to evaluate the distribution pattern and prognostic value of 21-gene recurrence score (RS) in Chinese patients with mucinous breast cancer (MC) and compared with infiltrating ductal carcinoma (IDC). Materials and Methods Patients diagnosed with MC or IDC from January 2010 to January 2017 were retrospectively recruited. Reverse transcriptase–polymerase chain reaction assay of 21 genes was conducted to calculate the RS. Univariate and multivariate analyses were performed to assess the association between RS and clinicopathological factors. Survival outcomes including disease-free survival (DFS) and overall survival (OS) were estimated by Kaplan-Meier method and compared by log-rank test. Results The MC cohort included 128 patients and the IDC cohort included 707 patients. The proportions of patients with a low (RS < 18), intermediate (18-30), or high risk (RS > 30) were 32.0%, 48.4%, and 19.5% in MC cohort, and 26.9%, 46.8% and 26.3% in IDC cohort. The distribution of RS varied significantly according to different Ki-67 index and molecular subtype in both cohorts. Moreover, the receipt of chemotherapy was associated with RS in both cohorts. Among patients with MC, tumor stage was related to the DFS (p=0.040). No significant differences in DFS and OS were found among MC patients in different RS risk groups (OS, p=0.695; DFS, p=0.926). Conclusion RS was significantly related to Ki-67 index and molecular subtypes in MC patients, which is similar in IDC patients. However, RS was not able to predict DFS and OS in patients with MC.

        Cloning and analysis of b-amyrin synthase gene in Bupleurum chinense

        Bupleurum chinense DC. is one of the source plants of a well-known crude drug, Chai hu (Radix Bupleuri), producing triterpenoid saponins (saikosaponins) with a wide-spectrum of pharmacological applications. The biosynthesis of triterpenoid saponins involved with the cyclizing of the precursor 2,3-oxidosqualene to produce the first committed triterpene b-amyrin catalyzed by b-amyrin synthase (b-AS), whereafter diverse of triterpenoid saponins was biosynthesized. In addition, 2,3-oxidosqualene could be catalyzed by cycloartenol synthase directing to the synthesis of phytosterol. b-AS was thus defined as an important branch point between primary and secondary metabolisms, and may play a regulating role in the control of triterpenoid saponins biosynthesis. In this study, the promoter and protein-encoding regions of a b-AS gene (designated bcAS1) were isolated by genome walking and PCR from B. chinense. Several important cis-acting elements for gene regulation were identified within the promoter region including light-responsive, hormoneresponsive and various other stress-related elements. Approximate 0.8 kb fragment on upstream of ATG start codon of bcAS1 was sub-cloned into pAN580 vector to replace the 35S promoter driving the expression of green fluorescent protein (GFP) gene. The promoter activity was detected by transient expression in onion epidermis cells by the expression of GFP. Approximately 6 kb length of bcAS1 gene was cloned, containing 18 exons and 17 introns. Although a dozen of b-AS cDNA was isolated, seldom the promoter and gene of it was reported. This work was a valuable foundation for further studies on the regulatory role of b-AS in biosynthesis of saikosaponins.

        Saponins from Panax japonicus ameliorate age-related renal fibrosis by inhibition of inflammation mediated by NF-κB and TGF-β1/Smad signaling and suppression of oxidative stress via activation of Nrf2-ARE signaling

        Background: The decreased renal function is known to be associated with biological aging, of which the main pathological features are chronic inflammation and renal interstitial fibrosis. In previous studies, we reported that total saponins from Panax japonicus (SPJs) can availably protect acute myocardial ischemia. We proposed that SPJs might have similar protective effects for aging-associated renal interstitial fibrosis. Thus, in the present study, we evaluated the overall effect of SPJs on renal fibrosis. Methods: Sprague-Dawley (SD) aging rats were given SPJs by gavage beginning from 18 months old, at 10 ㎎/㎏/d and 60 ㎎/㎏/d, up to 24 months old. After the experiment, changes in morphology, function and fibrosis of their kidneys were detected. The levels of serum uric acid (UA), β2-microglobulin (β2-MG) and cystatin C (Cys C) were assayed with ELISA kits. The levels of extracellular matrix (ECM), matrix metalloproteinases (MMPs), tissue inhibitors of metalloproteinases (TIMPs), inflammatory factors and changes of oxidative stress parameters were examined. Results: After SPJs treatment, SD rats showed significantly histopathological changes in kidneys accompanied by decreased renal fibrosis and increased renal function; As compared with those in 3-month group, the levels of serum UA, Cys C and β2-MG in 24-month group were significantly increased (p < 0.05). Compared with those in the 24-month group, the levels of serum UA, Cys C and β2-MG in the SPJ-H group were significantly decreased. While ECM was reduced and the levels of MMP-2 and MMP-9 were increased, the levels of TIMP-1, TIMP-2 and transforming growth factor-β1 (TGF-β1)/Smad signaling were decreased; the expression level of phosphorylated nuclear factor kappa-B (NF-κB) was down-regulated with reduced inflammatory factors; meanwhile, the expression of nuclear factor erythroid 2-related factor 2-antioxidant response element (Nrf2-ARE) signaling was aggrandized. Conclusion: These results suggest that SPJs treatment can improve age-associated renal fibrosis by inhibiting TGF-β1/Smad, NFκB signaling pathways and activating Nrf2-ARE signaling pathways and that SPJs can be a potentially valuable anti-renal fibrosis drug.
