<P><B>Abstract</B></P> <P>Isoliquiritigenin (ISL) is a natural flavonoid that exhibits anticancer properties in various carcinoma cell types. However, the precise mechanism responsible for its anticancer activity has not been elucidated fully. In the present study, we examined ISL-mediated apoptotic mechanisms in colorectal cancer (CRC) cells. ISL induced apoptosis in human HCT-116 cells and caused marked induction of p62/SQSTM1 mRNA and protein expression. Similarly, ISL potently inhibited <I>in vivo</I> tumor growth and induced p62/SQSTM1 expression in xenograft tumor tissues. In a p62/SQSTM1 siRNA transfection study, ISL-induced p62/SQSTM1 expression attenuated ISL-mediated apoptosis by reducing caspase-8 activation. ISL potentiated the apoptotic effects of 5-fluorouracil (5-FU) on HCT-116 cells. However, ISL-induced p62/SQSTM1 expression also attenuated the potency of apoptosis induced by the combination of 5-FU and ISL. Our results demonstrate that ISL-induced p62/SQSTM1 upregulation affects ISL-mediated apoptotic potential through attenuation of caspase-8 activation in CRC cells. These findings broaden the understanding of the molecular basis of ISL-mediated apoptosis.</P>

The disengaging between invert filling and segment affects the safety of train operation. A 3D numerical model of a three-ring staggered assembled shield tunnel is established, which is verified by a full-scale test. On this basis, cohesive element is inserted between the invert filling and tunnel segment to simulate the fracture process of invert filling-segment interface, and explore the relationship between the deformation of staggered assembled shield tunnel and the disengaging of invert filling. The results show that: 1) The development of invert filling disengaging affects the internal forces of shield tunnel segments. 2) The relationship between the development of invert filling disengaging and the deformation of shield tunnel can be four stages. Different segment assembly methods do not affect the variability of the disengaging ratio. Tensile failure occurs in 89% of the interface layer, which is the main form of invert filling disengaging. 3) The relationship between the development of disengaging distance and the deformation of shield tunnel also can be four stages. The faster the horizontal convergent deformation develops in the segment assembly method, the slower the growth rate of the disengaging distance. 4) The faster the horizontal convergent deformation develops in the segment assembly method, the slower the growth rate of the disengaging volume.

OBJECTIVE:: Gallic acid, a natural chemical found in plants, has been reported to show antioxidant, anticancer, and anti-inflammatory effects. We investigated the efficacy of a short-term or long-term treatment with gallic acid in N-nitro-L-arginine methyl ester (L-NAME)-induced hypertensive mice and the underlying regulatory mechanism. METHODS:: Hypertension was sufficiently induced after 2 weeks of L-NAME administration. Cardiac remodeling was assessed by echocardiography. Hypertrophic markers, transcription factors, and fibrosis-related gene expression were evaluated by quantitative real-time polymerase chain reaction and western blotting. RESULTS:: Gallic acid effectively lowered SBP, regardless of the administration route (intraperitoneal or oral). L-NAME increased the left ventricular (LV) thickness without an increase in the total heart weight. Weekly echocardiography demonstrated that gallic acid significantly reduced LV posterior wall and septum thickness in chronic L-NAME mice from 3 to 7 weeks. The administration of gallic acid to mice showed a dual preventive and therapeutic effect on the L-NAME-induced LV remodeling. The effect was associated with the suppression of the gene expression of hypertrophy markers and the GATA-binding factor 6 (GATA6) transcription factor. Short-term or long-term treatment with gallic acid attenuated cardiac fibrosis and reduced the expression of histone deacetylase 1 and 2 in H9c2 cells and in rat primary cardiac fibroblasts, as well as in vivo. Small interfering RNA knockdown confirmed the association of these enzymes with L-NAME-induced cardiac remodeling and fibrosis. CONCLUSION:: These results suggested that gallic acid may be a potential therapeutic agent for the treatment of cardiovascular diseases with hypertension and cardiac fibrosis.

Pediococcus acidilactici M76 (PA-M76) showed outstanding probiotic properties with high tolerance in acidic GIT environments, broad antimicrobial activity, and high adhesion capability in the intestinal tract of Caenorhabditis elegans. PA-M76 treatment resulted in significant increases of pro-inflammatory cytokine mRNA expression in macrophages. PA-M76 produced 1.62 g/L EPS under optimal lactic acid fermentation conditions. Lactic acid fermentation of BRE by PA-M76 did not significantly affect the total anthocyanin and flavonoid content, except for a significant increase in total polyphenol content compared to non-fermented BRE (NfBRE). However, fBRE exhibited increased DPPH radical scavenging activity, linoleic acid peroxidation inhibition rate, and ABTS scavenging activity of fBRE compared to NfBRE. Among the 28 compounds identified in the GC-MS analysis, esters were present as the major groups. The total concentration of volatile compounds was higher in fBRE than that in NfBRE. However, the undesirable flavor of terpenes decreased. PA-M76 might be useful for preparing functionally enhanced fermented beverages with a higher antioxidant activity of EPS and enhanced flavors.

The binding abilities of scutellarin (Scu) andscutellarein (Scue) with bovine serum albumin (BSA) wereinvestigated using equilibrium dialysis, high performanceliquid chromatography, fluorescence spectroscopy, competitivesite marker and molecular docking. The resultsshowed that the average protein binding ratios of Scu andScue with BSA were (79.85 ± 1.83) and (85.49 ± 1.21) %respectively. Under simulated physiological conditions, thefluorescence data indicated that Scu and Scue bound withBSA through a static mechanism. The thermodynamicparameters indicated that the interactions of Scu-BSA andScue-BSA mainly occurred by van der Waals forces andhydrogen bonds and it was easier for Scue to bind withBSA than Scu, indicating that the glucuronic acid moleculein Scu decreased the binding affinity. Site competitivemarker experiments showed that the binding sites of Scuand Scue mainly located within the sub-domain IIA ofBSA. Furthermore, molecular docking studies indicatedthat one BSA could bind three Scue, while one BSA couldcarry only two Scu. All these results clearly indicated theinteractions of Scu and Scue with BSA, which will lay thefoundation for further research to determine the pharmacologyand pharmacodynamics of Scu and Scue for treatingischemic cerebrovascular disease.

Background: A role for autophagy, a conserved cellular response to stress, has recently been demonstrated in human cancers. Heme oxygenase-1 (HO-1) is a stress-inducible enzyme with anti-apoptotic property. In this study, we investigated the association between isoliquiritigenin (ISQ) and autophagy in HCT-116 cells. In addition, we determined whether the anti-apoptotic effects are mediated by HO-1-dependent autophagy. Methods: The protein expression of HO-1, LC3B-II, ATG5, ATG7, Benclin-1, PARP were analyzed by western blot analysis. Autophagy was assessed by acridine orange staining, immunofl uorescence (LC3 puncta). Knock-down of HO-1 expression was achieved with siRNA. Results: ISQ signifi cant induced LC3B-II, Beclin-1, ATG5, ATG7, PARP cleavage, expression compared to control values. Treatment with bafi lomycin A1 (BaF), autophagy inhibitor, significantly enhanced ISQ-induced accumulation of the autophagosomal marker LC3B-II and PARP cleavage. ISQ induce autophagy which inhibits against ISQ-induced apoptosis in HCT-116 cells. Furthermore, HO-1 knockdown by siRNA abolished the effects of anti-apoptotic and decreased LC3-II/LC3-I ratio in HCT-116 cells. The protective role of autophagy was dependent on HO-1 in HCT-116. Conclusions: Our results demonstrated that ISQ induce autophagy which is mediated by induction of HO-1 expression. ISQ-induced autophagy followed by apoptosis in HCT-116 cells.

Shield tunnel lining is mainly composed of prefabricated segment, accurate description of longitudinal seam joint mechanics is important for design and analysis of shield tunnel segmental linings, however, due to its stress state is complex and internal force changes show the characteristics of nonlinear, there is no more rapid and accurate calculation model to analysis the mechanics. In the view of the problem of the entity unit simulating bolt bending with low computing speed, the model of shield tunnel longitudinal seam that using the nonlinear spring to simulate bending bolt joint is proposed. Combined with longitudinal seam joint test, analysis the entity model and spring bolt model, the study concluded that: 1) the calculation results of spring bolt model are closer to the test results than entity unit model; and 2) calculation efficiency of spring bolt model bolt up 1 times than the physical model. Based on the spring bolt model, further study the relationship between viscous coefficient, friction coefficient and calculation efficiency, and analyzes the regularity of deformation and damage of shield tunnel connection.

Natural ester is a suitable substitute for mineral oil and has been widely used in AC transformer in many countries. In order to further application of natural ester in direct current (DC) equipment, it is needed to investigate its long term insulation property under DC condition. In this paper, a thermal ageing experiment was conducted for both mineral oil-paper and natural ester-paper insulation. The DC breakdown and space charge characteristics of insulation paper impregnated with natural ester and mineral oil was compared. Results show that the resistivity of the paper immersed in natural ester and mineral oil both increase as the ageing goes on. While insulation paper impregnated with natural ester has higher resistivity and DC breakdown voltage than the paper impregnated with mineral oil. The DC breakdown voltage for the oil impregnated insulation paper being DC pre-stressing is higher than that without pre-stressing. The average DC breakdown field strength difference between the test with pre-stressing and without pre-stressing clearly shows that there is an apparent enhancement effect for the homo-charge injection on the DC breakdown.
