        Demography and mass-rearing of Carposina sasakii Matsumura (Lepidoptera: Carposinidae) reared on Golden Delicious and Red Fuji apples in the laboratory

        Carposina sasakii Matsumura (Lepidoptera: Carposinidae), is one of the most serious fruit-boring pests in over ten species of fruit trees, and is especially damaging apples in the northern of China. The application of new planting systems, i.e., high-density and dwarfing rootstock orchard systems with mixed apple varieties, makes it im portant to study the fitness of C. sasakii on these apple varieties to gain fundamental knowledge for use in pest management involving this insect. In this study, life table data of C. sasakii were collected using Golden Delicious and Red Fuji apples as hosts. The egg-larva duration of male C. sasakii reared on Golden Delicious apples (22.81 d) was significantly shorter than that reared on Red Fuji apples (24.27 d). The egg-larva mortality in Golden Delicious apples (59.00%) was lower than that in Red Fuji apples (72.49%). The mortality of the pupal stage, however, was higher in Golden Delicious (10.51%) than in Red Fuji (0%). The total oviposition period (TPOP) on Golden Delicious apples (32.94 d) was significantly shorter than in individuals reared on Red Fuji apples (34.19 d). The intrinsic rate of increase (r = 0.0581 d −1 ), net reproductive rate (R 0 = 7.57 offspring), and finite rate of increase (λ = 1.0598 d −1 ) were all higher on Golden Delicious than those on Red Fuji. When the net re productive rate (R 0 ) was used, the harvest rate of pupae was higher (0.8678) when reared on Golden Delicious apples than when reared on Red Fuji apples (0.8398). When a large cohort size (n = 200) was used for effective bootstrap sample, the P E values for C. sasakii reared on Golden Delicious apples and Red Fuji apples were both almost equal to 1. For C. sasakii culturing purposes, Golden Delicious apples would be more productive than Red Fuji.

        By means of the model equivalence theory, this paper proposes a model equivalence based least squares iterative algorithm for estimating the parameters of stochastic dynamical systems with ARMA noise. The proposed algorithm reduces the number of the unknown noise terms in the information vector and can give more accurate parameter estimates compared with the generalized extended least squares algorithm. The validity of the proposed method is evaluated through a numerical example.

        MiR-494-3p Upregulation Exacerbates Cerebral Ischemia Injury by Targeting Bhlhe40

        Purpose: Cerebral ischemia is related to insufficient blood supply and is characterized by abnormal reactive oxygen species(ROS) production and cell apoptosis. Previous studies have revealed a key role for basic helix-loop-helix family member e40 (Bhlhe40)in oxidative stress and cell apoptosis. This study aimed to investigate the roles of miR-494-3p in cerebral ischemia/reperfusion(I/R) injury. Materials and Methods: A mouse middle cerebral artery occlusion (MCAO/R) model was established to mimic cerebral ischemiain vivo. Brain infarct area was assessed using triphenyl tetrazolium chloride staining. Oxygen-glucose deprivation/reoxygenation(OGD/R) operation was adopted to mimic neuronal injury in vitro. Cell apoptosis was analyzed by flow cytometry. Therelationship between miR-494-3p and Bhlhe40 was validated by luciferase reporter and RNA immunoprecipitation assays. Results: Bhlhe40 expression was downregulated both in MCAO/R animal models and OGD/R-induced SH-SY5Y cells. Bhlhe40overexpression inhibited cell apoptosis and reduced ROS production in SH-SY5Y cells after OGD/R treatment. MiR-494-3p wasverified to bind to Bhlhe40 and negatively regulate Bhlhe40 expression. Additionally, cell apoptosis and ROS production in OGD/R-treated SH-SY5Y cells were accelerated by miR-494-3p overexpression. Rescue experiments suggested that Bhlhe40 could reversethe effects of miR-494-3p overexpression on ROS production and cell apoptosis. Conclusion: MiR-494-3p exacerbates brain injury and neuronal injury by regulating Bhlhe40 after I/R.

        Study of compensatory growth based on different nutrition conditions of Bombyx mori

        Organisms achieve compensatory growth after a period of nutrient restriction followed by recovering the nutrient status. However, the underlying mechanisms associated with such growth acceleration remain unclear. The silkworm Bombyx mori is a lepidopteran model insect. This study aimed to investigate the physiological characteristics and the underlying mechanisms in B. mori fed on mulberry leaves (MG), artificial diet (AG), and artificial diet + mulberry leaves (AG-MG), respectively. Silkworms in AG-MG which fed on artificial diet from 1st to 3rd instars followed by feeding on mulberry leaves from 4th to 5th instars exhibited a higher weight gain rate than that in MG and AG, indicating that compensatory growth occurred as a result of the switch in the silkworm food regime. Trypsin and lipase activities of silkworms in AG-MG were shown to be up-regulated at 72 h after changing food. Digital gene expression profiling (DGE) analysis revealed that genes related to metabolism and development in silkworm midguts were differentially expressed. The quantitative real-time PCR (qRT-PCR) re sults showed that the expression levels of IIS/PI3K-AKT pathway genes including INR, IRS, AKT, PI3K60 and PI3K110 of silkworms in AG were down-regulated compared with that in MG at 0 h. Whereas AKT, PI3K and PI3K60 of silkworms were significantly increased by 1.68-, 1.49-, 1.67-fold, respectively, at 72 h after switched to mulberry leaves than the same instar fed on artificial diet. PDK’s expression of silkworms in AG was higher than that in other two groups at each timepoint. Compared with MG and AG, PTEN and IRS were down-regulated at 48 and 72 h in AG-MG. Collectively, these results indicate that compensatory growth in B. mori is regulated by IIS/ PI3K-AKT signaling pathway.

        A regression for estimating metabolizable glucose in diets of weaned piglets for optimal growth performance

        Objective: Two experiments were conducted to provide a new approach for evaluating feed nutritional value by metabolizable glucose (MG) in piglet diets with different levels of starch and crude fiber. In Exp 1, a regression equation for MG was generated. In Exp 2, the equation was verified, and the optimal growth performance of piglets under appropriate MG levels was tested. Methods: In Exp 1, 20 weaned piglets (7.74±0.81 kg body weight [BW]) were randomly assigned to 1 of 4 treatments, including the basal diet containing different levels of MG (starch, 25.80%, 31.67%, 45.71%, 49.36%; crude fiber, 1.23%, 1.35%, 1.80%, 1.51%). The piglets were implanted with an ileal fistula, cannulation of the carotid artery, portal vein, and mesenteric artery. The chyme from the ileum fistula and blood samples were collected. In Exp 2, 30 weaned piglets (8.96±0.50 kg BW) were randomly assigned to 1 of 5 treatments, including the experimental diets with different levels of MG (37.6, 132.5, 300.0, 354.3, and 412.5 g/kg). The piglets' BW, and feed consumption were recorded to calculate growth performance during the 28-d experiment. Results: In Exp 1, the MG levels in 4 diets were 239.62, 280.68, 400.79, and 454.35 g/kg. The regression equation for the MG levels and dietary nutrients was: Y (MG) = 12.13×X<sub>1</sub> (starch)+23.18×X<sub>2</sub> (crude fiber)-196.44 (R<sup>2</sup> = 0.9989, p = 0.033). In Exp 2, treatments with 132.5 and 300.0 g/kg MG significantly (p<0.05) increased average daily gain and feed conversion efficiency of weaned piglets, increased digestibility of crude fat, and had no effect on digestibility of crude protein compared to 300.0 to 412.5 g/kg MG. Conclusion: The pig model combining the ileum fistula and cannulation of blood vessels was successfully used to determine the dietary MG levels. The recommended MG level in weaned pig diets is 132.5 to 300.0 g/kg.

        Composite hydrogels filled with inclusion complexes made from β-cyclodextrins with poly(propylene glycol) bisamine

        A novel kind of composite hydrogel lled with inclusion complexes (ICs) made fromb-cyclodextrins (b-CDs) with amine-terminatedpoly(propylene glycol) (PPG-BA) was prepared by copolymerization of methacryloyl end-capped poly(L-lactide)-poly(ethylene glycol)-poly-(L-lactide) macromer (DMPEGL) with acrylic acid (AA) in water under UV irradiation. Owing to the hydrogen bonding interactionof COOH in AA on the network backbones with NH2 and OH in ICs embedded in the crosslinked network, the composite hydrogelsmechanical properties being signicantly improved. The mass loss test conducted in water indicates that they degrade by following a dualdegradation mechanism, that is, not onlyb-CDs slipping from the polymer axles and then escaping out of the hydrogel network, but alsothe bulk hydrogel network hydrolyzing itself to lose the weight. These composite hydrogels show potential to be used as biomaterials fortissue engineering scaolds and carriers of drugs with controlled release.

        Lactate potentiates angiogenesis and neurogenesis in experimental intracerebral hemorrhage

        Lactate accumulation has been observed in the brain with intracerebral hemorrhage (ICH). However, the outcome of lactate accumulation has not been well characterized. Here, we report that lactate accumulation contributes to angiogenesis and neurogenesis in ICH. In the first set of the experiment, a rat model of ICH was induced by injecting collagenase into the brain. The effects of lactate accumulation on the neurological function, apoptosis, and numbers of newborn endothelial cells and neurons, as well as the proliferation-associated signaling pathway, were evaluated in the rat brain. In the second set, exogenous L-lactate was infused into intact rat brains so that its effects could be further assessed. Following ICH, lactate accumulated around the hematoma; the numbers of PCNA+/vWF+ nuclei and PCNA+/DCX+ cells were significantly increased compared with the numbers in the Sham group. Moreover, ICH induced translocation of nuclear factor-kappa B (NF-κB) p65 into the nucleus, resulting in a notable upregulation of VEGF and bFGF mRNAs and proteins compared with the levels in the Sham controls. Administration of a lactate dehydrogenase inhibitor dramatically inhibited these effects, decreased the vascular density, and aggravated neurological severity scores and apoptosis after ICH. After exogenous L-lactate infusion, the numbers of PCNA+/vWF+ nuclei and PCNA+/DCX+ cells were strikingly increased compared with the numbers in the Sham controls. In addition, lactate facilitated NF-κB translocation to induce increased transcription of VEGF and bFGF. Co-infusion with an NF-κB inhibitor significantly inhibited these effects. These data suggest that lactate potentiates angiogenesis and neurogenesis by activating the NF-κB signaling pathway following ICH.
