Construction of Targeting Vectors to Disrupt PAT1 Gene in Arabidopsis Trp1-100

To develop the gene targeting system by homologous recombination in Arabidopsis thaliana, we constructed two targeting vectors and showed the reliability of the scheme which is based on genetic complementation of phosphoribosylanthranilate transferase (PAT1) gene. The PAT1 gene, which is essential for tryptophan biosynthesis, was selected as a target gene because the loss of function leads to fluorescence phenotype due to the accumulation of anthranilic acid derivatives. pHS113 contains PAT1 gene surrounding 5' and 3' flanking portions, but the most coding region of the PAT1 gene is replaced by the neomycin phosphotransferase gene (NPTⅡ). pHS117 consists of 1.1 kb internal fragment of genomic PAT1 gene following with NPTⅡ gene. In this targeting strategy, Arabidopsis PAT1 gene can be disrupted by single-step of transformation experiment.

Arabidopsis trp1 mutant plants, deficient in phosphoribosyI anthranilate transferase (PAT) activity, accumulate anthranilate compounds, which render them blue fluorescence. The visible phenotype of trp1 makes the PAT gene an excellent reporter gene in the mutant. In order to develop a system for the homologous recombination using the phenotypic characteristic of trp1-100, we established optimum conditions for the isolation and regenera tion of protoplast from auxin-conditioned, trp1-100 root cultures. Trvptophan had to be supplemented in the germination medium for the efficient cell division and subsequent plant regeneration. When 10 uM tryptophan was added to the germination medium, we obtained the highest yield of protoplasts ($3{\times}10^6 cells/g$) and the best viability (92%). Thirty percent of root protoplast derived from meristematic cells underwent cell division within 5 days in callus-induction medium. Regenerated rosette leaves (2-3 mm) were transferred to rooting medium and finally acclimated to the soil for flowering.

In connection with biodiversity that has recently become the topic, competition to secure biological genetic resources is being heightened all over the world. Korea also has a variety of efforts to secure and preserve plant genetic resources, and has recognized the importance of the function and role of arboretums. Arboretums in Korea, however, have the problems in terms of quality due to the rapid increase. One of the problems is shortages in experts who are essential for the proper functioning of arboretums. To solve the problem, the State introduced Gardener Training Program certification system and started to train gardeners. However, gardeners who have actually employed at arboretums are less than 20% and thus the problem of shortages in experts still exists. In this context, this study examined 44 registered arboretums in Korea to find out the current situation of experts; analyzed arboretum experts and the program certification system in view of relevant laws; and investigated the current situation of training programs being operated in three Gardener Training Program certification organizations. Finally, this study conducted a survey of 68 gardeners who completed the training program at Chollipo Arboretum and tried to suggest a plan for securing experts from the gardener training programs. The plan for utilizing the trained gardeners as experts in arboretums is as follows: First, legal standards for the employment of experts in arboretums should be strengthened. Second, it is necessary to evaluate training programs, in order to raise the reliability of arboretum experts` expertise. Third, official validity that can be honored in all arboretums should be granted to a certificate of program completion. Finally, networks of gardeners who completed the programs should be formed through follow-up management of them.

본 연구에서는 다변화되어 가는 현대사회 시민들의 의식수준을 조사하고 환경보존이 재해방지에 미치는 영향에 대해 본 고찰을 통하여 분석하여 보았다. 첫째 , 환경문제 의식수준은 높고 의식은 매스미디어에서 가장 많이 얻는다. 둘째, 시민들은 오늘의 광주시 환경상태를 생각하며 그 대책이 시급하다고 생각하고 있다. 셋째 , 환경보전 교육이 절실하게 필요하고 가장 시급한 문제는 수질 오염을 지향하고 있으며 환경파괴 없는 경제성장을 요구하고 있다. 넷째, 시민들은 중소도시에서 살고 싶고 미래에 대한 견해는 비교적 높은 참여의식을 갖고 있다. 다른 지역으로 이주에 대한 문제는 중소도시를 원하고 있어 문화생활을 누리면서도 공해가 없는 곳을 원하고 있는 것으로 평가되며 환경문제는 앞으로 광주시에 증가할 것이라는 비관적 예상을 보여주고 있고 환경보전은 비교적 높은 참여의식을 보여주고 있으나 실제로는 적극적이 아닌 것으로 평가 예측된다. A method is described for the determination of dibutyltin (DBT) in sediment by isotope dilution using liquid chromatography inductively-coupled plasma/mass spectrometry (LC-ICP/MS). To achieve the highest accuracy and precision, special attentions are paid in optimization and evaluation of overall processes of the analysis including extraction of analytes, characterization of the standards used for calibration and LC-ICP/MS conditions. An approach for characterization of natural abundance DBT standard has been developed by combining inductively-coupled plasma/optical emission spectrometry (ICP/OES) and LC-ICP/MS for the total Sn assay and the analysis of Sn species present as impurities, respectively. An excellent LC condition for separation of organotin species was found, which is suitable for simultaneous DBT and tributyltin (TBT) analysis as well as impurity analysis of DBT standards. Microwave extraction condition was also optimized for high efficiency while preventing species transformation. The present method determines the amount contents of DBT in sediments with expanded uncertainty of less than 5% and its result shows high degree of equivalence with reference values of an international inter-comparison and a certified reference material (CRM) within stated uncertainties.
