The common γ-chain (γc) plays a central role in signaling by IL-2 and other γc-dependent cytokines. Here we report that activated T cells produce an alternatively spliced form of γc mRNA that results in protein expression and secretion of the γc extracellular domain. The soluble form of γc (sγc) is present in serum and directly binds to IL-2Rβ and IL-7Rα proteins on T cells to inhibit cytokine signaling and promote inflammation. sγc suppressed IL-7 signaling to impair naive T cell survival during homeostasis and exacerbated Th17-cell-mediated inflammation by inhibiting IL-2 signaling upon T cell activation. Reciprocally, the severity of Th17-cell-mediated inflammatory diseases was markedly diminished in mice lacking sγc. Thus, sγc expression is a naturally occurring immunomodulator that regulates γc cytokine signaling and controls T cell activation and differentiation.
Effect of Vitamin E on Lipid Parameters in Ovariectomized Rats

The risk of cardiovascular disease drastically increases at the onset of menopause, in part, because of rise inblood cholesterol and unfavorable changes in lipid profile. This study was designed to investigate the dose-dependent effectsof vitamin E supplementation on lipid parameters in ovariectomized (ovx) rats. Sixty 12-month-old female Sprague-Dawleyrats were either sham-operated (sham; one group) or ovx (four groups). All rats were maintained on a semipurified casein-based diet (AIN-93M; 75 IU vitamin E/kg of diet) for a period of 120 days. Thereafter, ovx rats were placed on one of fourdoses of vitamin E treatment (75, 300, 525, or 750 IU vitamin E/kg of diet), while the sham group was continued on 75 IUvitamin E/kg of diet for 100 days. Ovariectomy tended to increase (by 24%, P. 0.1) serum nonhigh-density lipoprotein(HDL) cholesterol and decrease (by 14%, P. 0.1) HDL cholesterol. Vitamin E did not have any significant effects on serumlipid parameters. Liver total lipids were notably increased (P. .001) in ovx animals, and supplementation with vitamin E at525 IU/kg of diet was able to significantly reduce liver total lipids by 13%. Additionally, ovariectomy caused an increase inserum glucose and liver C18:1fatty acid concentrations along with decreases in C18:0, C20:4, and C22:6fatty acid concentra-tions. These alterations on liver fatty acid profiles were unaffected by vitamin E. The findings of this study suggest that vit-amin E supplementation moderately improves lipid parameters in ovarian hormone-deficient rats.
Oleanane-type saponins from Glochidion glomerulatum and their cytotoxic activities

Eight oleanane-type saponins, glomerulosides A-H, were isolated from leaves of Glochidion glomerulatum. All isolated compounds were evaluated for cytotoxic activity on four human cancer cell lines, A-549, HT-29, OVCAR, and MCF-7. Glomerulosides C and E, which contain a benzoyloxy group at C-22, showed significant cytotoxic activities against the A-549, HT-29, and OVCAR cancer cell lines with IC<SUB>50</SUB> values ranging from 5.9 to 9.8μM. Glomeruloside A showed cytotoxicity on HT-29 and OVCAR cell lines with IC<SUB>50</SUB> values of 7.3 and 6.6μM, respectively. Moreover, glomeruloside B exhibited significant activity on A-549 and HT-29 cancer cell lines with IC<SUB>50</SUB> values of 9.7 and 7.5μM. In contrast, glomerulosides F-H, lacking a benzoyloxy group, showed only moderate cytotoxic activity.

Synchrotron-radiation based photoemission spectroscopy (SRPES) was used to examine the changes in the TiO<SUB>2</SUB> surface structures as a result of high-energy e-beam exposure (under atmospheric pressure). The influence of the e-beam pre-treatment on the structure of the post-deposited Pt films on TiO<SUB>2</SUB> was also investigated. High-energy e-beam treatment increased concentrations of OH species and C-O bonds on the surface. The interface reaction between Ti-oxide and Pt was reduced by the e-beam treatment. Consequently, different Pt film surface morphologies could be found on TiO<SUB>2</SUB> with and without e-beam treatment.

A general method for fabricating transient plasma structures with high-intensity laser pulses is developed to gain fine control over laser-plasma interactions. These structures have been used as programmable photonic devices in the development of laser-wakefield accelerators, soft X-ray lasers and plasma nonlinear optics driven by multi-terawatt laser pulses. Plasma ramps are used to control electron injection in laser-wakefield accelerators, plasma waveguides are used to enhance the efficiency of soft X-ray lasers by orders of magnitude and periodic plasma structures are used to achieve quasi-phase matching in relativistic harmonic generation. By scanning the interaction length with the same plasma-fabrication method, tomographic measurements are carried out to resolve the injection/acceleration process in laser-wakefield accelerators and amplification processes in X-ray lasers and relativistic harmonic generation. A theoretical analysis and a computer simulation are also carried out to provide insightful pictures of these processes. These research works show that by controlling plasma structures with optical fabrication methods, laser-plasma interaction can be engineered to expand and enrich the frontier of high-field physics.

In order to understand the effect of duck hepatitis B virus (DHBV) on the economic performance of ducks, Three groups (DHBV congenitally infected, experimentally infected and DHBV negative) Brown Tsaiya ducks (Anas platyrhyncha) were used for experimental animals. Artificial insemination and pedigree hatching were applied in the propagation of ducklings, and the efficiency of vertical transmission and experimental infection was analyzed through the detection of DHBV DNA in the sera of 8-week-old offspring. The observation of the records of the first year indicated that the persistent infection had no significant effects on the performance of ducks, except the egg number of survival ducks up to 40 week of age. Thus DHBV infection did not appear to give ill effects to the economic performance of ducks in first laying year. A higher infection rate (85.3%) was obtained in congenital transmission than that (75.5%) of experimental infection. Both modes of infection did not reach 100% infectious rate, although some ducks developed transient viraemia in a tracing of DHBV DNA for 24 weeks to 11 challenged ducklings.

An ongoing pandemic of coronavirus disease 2019 (COVID-19) is now the greatest threat to global public health. Herbal medicines and their derived natural products have drawn much attention in the treatment of COVID-19, but the detailed mechanisms by which natural products inhibit SARS-CoV-2 have not been elucidated. Here, we show that platycodin D (PD), a triterpenoid saponin abundant in Platycodon grandiflorum (PG), a dietary and medicinal herb commonly used in East Asia, effectively blocks the two main SARS-CoV-2 infection routes via lysosome- and transmembrane protease serine 2 (TMPRSS2)-driven entry. Mechanistically, PD prevents host entry of SARS-CoV-2 by redistributing membrane cholesterol to prevent membrane fusion, which can be reinstated by treatment with a PD-encapsulating agent. Furthermore, the inhibitory effects of PD are recapitulated by the pharmacological inhibition or gene silencing of NPC1 , which is mutated in patients with Niemann–Pick type C (NPC) displaying disrupted membrane cholesterol distribution. Finally, readily available local foods or herbal medicines containing PG root show similar inhibitory effects against SARS-CoV-2 infection. Our study proposes that PD is a potent natural product for preventing or treating COVID-19 and that briefly disrupting the distribution of membrane cholesterol is a potential novel therapeutic strategy for SARS-CoV-2 infection.
