They cause a sort of anti-toxic response or confusion of the immune system, and ultimately bring various types of diseases. This syndrome is related "Poison in Excrement" or "Internal Damage(內傷發□)" and Leaky Gut Syndrome, so I will study on the relationship between them. Method: Study on the relationship between Dae-Jang-Jung-gyeok and Leaky Gut Syndrome. Result: Korean doctors in ancient times called this syndrome "Poison in Excrement" or "Internal Damage(內傷發□)" and treated it with "Dae-Jang-Jung-Gyeok(大腸正格)". Leaky Gut Syndrome is a sort of clinical lesion, which allows foreign and harmful toxins in and results in a disorder of the immune system due to the leaking intestinal mucosa. Conclusion: Based upon the analyses, Dae-Jang-Jung-Gyeok and Leaky Gut Syndrome are closely related. Therefore diseases caused by the Leaky Gut Syndrome might be expected to be cured by Dae-Jang-Jung-Gyeok. Particularly the use of treatments for self-immune diseases and allergic diseases such as atopyic dermatitis and articular rheumatism should be expected to reveal a new path of treatment for other disorders such as Leaky Gut Syndrome. Effect of Highly Pressurized Hydrogen Gas Charging on the Hydrogen Embrittlement of API X70 Steel

During the use of API X70 steel as a pipeline structural material for the transportation of natural gas,hydrogen embrittlement can occur due to the hydrogen contained in natural gas. The aim of this study is toinvestigate the effects of the hydrogen content under high-pressure hydrogen gas conditions on the hydrogenembrittlement of air-cooled API X70 steel. The air-cooled API X70 steel was manufactured by hot rollingand was then air-cooled to room temperature. Tensile test specimens were held for 0 h, 1000 h, and 2000 hwithin a pressure vessel filled with 100% hydrogen gas at a gas pressure of 10 MPa, with the tensile teststhen performed at room temperature. The microstructure of the API X70 steel consists of coarse polygonalferrite, coarse pearlite, and fine acicular ferrite. The yield and tensile strength increased and elongationdecreased considerably after a holding time of 2000 h compared to those of 0 h and 1000 h within the pressurevessel. The morphology of the fracture surface changed from ductile to brittle upon hydrogen gas charging. Secondary cracks were observed in both of the hydrogen-gas-charged specimens. No external cracks wereformed on the surface of the tensile-tested specimen with a 0 h holding time; however, many external crackswere observed on the specimen surface subjected to hydrogen gas charging.

In 2016, the World Health Organization revised the diagnostic criteria for myeloproliferative neoplasms (MPNs) based on the discovery of disease-driving genetic aberrations and extensive analysis of the clinical characteristics of patients with MPNs. Recent studies have suggested that additional somatic mutations have a clinical impact on the prognosis of patients harboring these genetic abnormalities. Treatment strategies have also advanced with the introduction of JAK inhibitors, one of which has been approved for the treatment of patients with myelofibrosis and those with hydroxyurea-resistant or intolerant polycythemia vera. Recently developed drugs aim to elicit hematologic responses, as well as symptomatic and molecular responses, and the response criteria were refined accordingly. Based on these changes, we have revised the guidelines and present the diagnosis, treatment, and risk stratification of MPNs encountered in Korea.
Development of Plastid InDel Markers to Discriminate Lemons from Other Citrus Groups

Lemon (Citrus limon), an interspecific hybrid between sour orange and citron, has been widely used as a rootstock along with trifoliate orange. Though lemons are superior to trifoliate orange in terms of their high seed germination rate throughout the year, one of the obstacles to using lemons as rootstocks is the lack of reliable, lemon-specific molecular markers to discriminate buds of the micro-grafted scion from those of the lemon rootstock. In order to obtain lemon-specific molecular markers, we compared the whole-plastid genomes available from four citrus species (lemon, pummelo, sweet orange, and mandarin) and developed seven plastid insertion/deletion (InDel) markers. The plastid InDel markers were applied to 46 citrus accessions that included lemons (17 accessions), grapefruit, mandarin, pummelo, sour orange, orange, papeda, tangor, and tangelo groups. The resulting dendrogram revealed that the citrus accessions used in this analysis could be distinctly classified into seven clusters. Lemons formed a separate cluster and had identical allele sizes for each InDel locus among all accessions investigated. This set of InDel markers could be a useful molecular tool for the rapid and clear discrimination of micro-grafted scions and lemon rootstocks during the production of virus-free citrus trees. The plastid InDel markers with maternal inheritance features can also be used to analyze the phylogenetic origin of various citrus cultivars including lemons.

<P><B>Abstract</B></P> <P>Bioassay-guided fractionation of the methanolic extract from the roots of <I>Cynanchum atratum</I> has resulted in the isolation of three new pregnane glycosides (<B>1</B>–<B>3</B>) along with four known compounds (<B>4</B>–<B>7</B>). Their structures were identified by analysis of the spectroscopic data including extensive 2D NMR. All of the isolates were evaluated for their potential to inhibit the melanin production in α-melanocyte stimulating hormone (α-MSH)-activated B16 melanoma cells. Of these, compounds <B>4</B>–<B>7</B> dose-dependently inhibited the melanin production with the IC<SUB>50</SUB> values ranging from 4 μM to 33 μM.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

Sensitive detection of dengue virus NS1 by highly stable affibody-functionalized gold nanoparticles

<P>Infection with dengue virus (DENV) is a serious health issue that causes severe dengue fever and occasionally lethal complications, such as dengue hemorrhagic fever. Therefore, rapid and sensitive DENV detection is important to reduce morbidity and mortality. Here, we screened dengue NS1-specific affibodies and developed highly stable anti-NS1 affibody-functionalized AuNPs to improve the detection sensitivity of the NS1 antigen in ELISA. First, we screened NS1 antigen-specific affibody molecules (ZNS112, ZNS116, and ZNS146) from an affibody phage library. The affibodies were then expressed and purified from <I>Escherichia coli.</I> Among them, the ZNS112 affibody showed the highest equilibrium binding constant (<I>K</I>d) of 1 μM and was sufficiently stable at high temperatures. This affibody was functionalized on AuNPs measuring 20 nm in diameter, which were used as carriers to achieve amplification of the signal in ELISA. The developed anti-NS1 affibody-functionalized AuNPs ((ZNS112)2-AuNP) showed good properties, such as easy synthesis, a high number of affibody conjugations on the AuNPs, and excellent stability under harsh conditions with high salt concentrations and temperature. In addition, this nanoparticle-based enhanced ELISA resulted in a 14.2-fold signal amplification performance for dengue NS1 detection in comparison with the affibody-based ELISA. This novel and sensitive method using (ZNS112)2-AuNPs may have applications in the detection of DENV in infected patients at an early stage and for the detection of other pathogens in clinical diagnostics.</P>
