        Ribosomal Crystallography: Peptide Bond Formation, Chaperone Assistance and Antibiotics Activity

        The peptidyl transferase center (PTC) is located in a protein free environment, thus confirming that the ribosome is a ribozyme. This arched void has dimensions suitable for accommodating the 3′ends of the A-and the P-site tRNAs, and is situated within a universal sizable symmetry-related region that connects all ribosomal functional centers involved in amino-acid polymerization. The linkage between the elaborate PTC architecture and the A-site tRNA position revealed that the Ato P-site passage of the tRNA 3′end is performed by a rotatory motion, which leads to stereochemistry suitable for peptide bond formation and for substrate mediated catalysis, thus suggesting that the PTC evolved by genefusion. Adjacent to the PTC is the entrance of the protein exit tunnel, shown to play active roles in sequencespecific gating of nascent chains and in responding to cellular signals. This tunnel also provides a site that may be exploited for local co-translational folding and seems to assist in nascent chain trafficking into the hydrophobic space formed by the first bacterial chaperone, the trigger factor. Many antibiotics target ribosomes. Although the ribosome is highly conserved, subtle sequence and/or conformational variations enable drug selectivity, thus facilitating clinical usage. Comparisons of high-resolution structures of complexes of antibiotics bound to ribosomes from eubacteria resembling pathogens, to an archaeon that shares properties with eukaryotes and to its mutant that allows antibiotics binding, demonstrated the unambiguous difference between mere binding and therapeutical effectiveness. The observed variability in antibiotics inhibitory modes, accompanied by the elucidation of the structural basis to antibiotics mechanism justifies expectations for structural based improved properties of existing compounds as well as for the development of novel drugs.

      • Assessment of damages on a RC building after a big fire

        This paper presents a case study about the damages on the structural elements of a cast in place reinforced concrete (RC) building after a big fire which was able to be controlled after six hours. The fire broke off at the $2^{nd}$ basement floor of the building, which has five basements, one ground, and two normal floors. As a result of intensely stocked ignitable materials, it spread out to the all of the upstairs. In visual inspection, most of the typical fire damages were observed (such as spalling, net-like cracks, crumbled plasters, bared or visible reinforcement). Also, failures of the $2^{nd}$ basement columns were encountered. It has been concluded that the severity failures of the columns at the $2^{nd}$ basement caused utterly deformation of the building, which is responsible for the massive damages on the beam-column connections. All of the observed damages were categorized related to the types and presented separated regarding the floors. Besides to the visual inspection, the numerical analysis was run to verify the observed damaged on the building for columns, beams, and the connection regions. It is concluded from the study that several parameters such as duration of the fire, level of the temperature influence on the damages to the RC building. Also, it is highlighted by the study that if the damaged building is considered on the overall structural system, it is not able to satisfy the minimum service requirements neither gravity loads nor earthquake conditions.

        The Hebrew Biblical Bérit in Light of Ancient Near Eastern Covenants and Treaties

        A short historical survey of scholarly interpretations of the concept of bérit ברית in the Hebrew Bible is offered in this article as a background to understanding the urge for different interpretations offered by scholars in recent decades. The survey starts with a detailed discussion of the contribution of Mendenhall’s interpretations of the biblical covenant in light of legal treaties from the Ancient Near East during the second millennium BCE. Those mainly Hittite international treaties opened the door for a new interpretation of the biblical texts in light of legal rather than just religious relations between the God of Israel and his people. The second part of the article shows how the Hittite treaty-documents have led to new interpretations of the Hebrew Bible covenant, based on ancient Near Eastern legal-political as well as cultic material, and how it may still be used in current research, together with Mesopotamian-Akkadian documents, which were also brought into that discussion. The major dispute between scholars today is the attempt to pose an historical measurement on the possible connection between the Hittite legal documents titled in Hittite išḫiul- which are dated to the second half of the second millennium, and the Akkadian documents titled adê mainly of the first millennium BCE, with the Hebrew Bible texts drawn during the first millennium.

        Review : Quantitative Polymerase Chain Reaction for Microbial Growth Kinetics of Mixed Culture System

        Microbial growth kinetics is often used to optimize environmental processes owing to its relation to the breakdown of substrate (contaminants). However, the quantification of bacterial populations in the environment is difficult owing to the challenges of monitoring a specific bacterial population within a diverse microbial community. Conventional methods are unable to detect and quantify the growth of individual strains separately in the mixed culture reactor. This work describes a novel quantitative PCR (qPCR)-based genomic approach to quantify each species in mixed culture and interpret its growth kinetics in the mixed system. Batch experiments were performed for both single and dual cultures of Pseudomonas putida and Escherichia coli K12 to obtain Monod kinetic parameters (μmax and Ks). The growth curves and kinetics obtained by conventional methods (i.e., dry weight measurement and absorbance reading) were compared with that obtained by qPCR assay. We anticipate that the adoption of this qPCR-based genomic assay can contribute significantly to traditional microbial kinetics, modeling practice, and the operation of bioreactors, where handling of complex mixed cultures is required.

      • Comparison of 2-Dimensional and 3-Dimensional Conformal Treatment Plans in Gastric Cancer Radiotherapy

        Background: Postoperative chemoradiotherapy is accepted as standard treatment for stage IB-IV, M0 gastric cancer. Radiotherapy (RT) planning of gastric cancer is important because of the low radiation tolerance of surrounding critical organs. The purpose of this study was to compare the dosimetric aspects of 2-dimensional (2D) and 3-dimensional (3D) treatment plans, with the twin aims of evaluating the adequacy of 2D planning fields on coverage of planning target volume (PTV) and 3D conformal plans for both covering PTV and reducing the normal tissue doses. Materials and Methods: Thirty-six patients with stage II-IV gastric adenocarcinoma were treated with adjuvant chemoradiotherapy using 3DRT. For each patient, a second 2D treatment plan was generated. The two techniques were compared for target volume coverage and dose to normal tissues using dose volume histogram (DVH) analysis. Results: 3DRT provides more adequate coverage of the target volume. Comparative DVHs for the left kidney and spinal cord demonstrate lower radiation doses with the 3D technique. Conclusions: 3DRT produced better dose distributions and reduced radiation doses to left kidney and spinal cord compared to the 2D technique. For this reason it can be predicted that 3DRT will result in better tumor control and less normal tissue complications.

        Guidance and Counseling in Aotearoa, New Zealand, Secondary Schools: Revisiting the Issues

        The changes in education policies in Aotearoa, New Zealand, since the 1980s have significantly affected guidance and counseling in secondary schools, leading to variations in provision and effectiveness. Issues being faced by school guidance counselors were identified in “Guidance and Counselling in New Zealand Secondary Schools: Exploring the Issues,” which represented a call for an external government authority who understood guidance and counseling and their value in schools to create national guidelines and standards for this work (Crowe, 2006). This 2014 article revisits the issues and reports on the findings and recommendations of a 2013 evaluation of guidance and counseling in Aotearoa, New Zealand secondary schools. The government requested the evaluation in response to the need to provide good guidance and counseling for young people at risk of developing mental health issues. The results and recommendations from the evaluation have highlighted the issues being faced by guidance counselors, other guidance staff, and their schools. The effectiveness of guidance and counseling in schools in Aotearoa, New Zealand will depend upon the implementation of the recommendations made in the report to the Ministry of Education, and, in particular, the review of the guidance staffing formula and the establishment of guidelines/expectations for schools/wharekura regarding the provision of guidance and counseling.

        Validation of Self-administrated Questionnaire for Psychiatric Disorders in Patients with Functional Dyspepsia

        Introduction: Psychiatric comorbidity is common in patients with functional dyspepsia (FD) but a good screening tool for psychiatric disorders in gastrointestinal clinical practice is Lacking. Aims: 1) Evaluate the performance and optimal cut-off of 12-item General Health Questionnaire (GHQ-12) as a screening tool for psychiatric disorders in FD patients; 2) Compare health-related quality of Life (HRQoL) in FD patients with and without psychiatric comorbidities. Methods: Consecutive patients fulfilling Rome III criteria for FD without medical co-morbidities and gastroesophageal reflux disease were recruited in a gastroenterology clinic. The followings were conducted at 4 weeks after index oesophagogastroduodenoscopy: self-administrated questionnaires on socio-demographics, dyspeptic symptom severity (4-point Likert scale), GHQ-12, and 36-item short-form health survey (SF-36). Psychiatric disorders were diagnosed with Structured Clinical Interview for DSM-IV Axis I Disorders (SCID) by a trained psychiatrist, which served as reference standard. Results: 55 patients underwent psychiatrist-conducted interview and questionnaire assessment. 27 (49.1%) had current psychiatric disorders as determined by SCID (anxiety disorders: 38.2%, depressive disorders: 16.4%). Receiver operating characteristic curve analysis of GHQ-12 revealed an area under curve of 0.825 (95%CI: 0.698-0.914). Cut-off of GHQ-12 at 3 gave a sensitivity of 63.0% (95%CI = 42.4-80.6%) and specificity of 92.9% (95%CI = 76.5%-98.9%). Subjects with co-existing psychiatric disorders scored significantly Lower in multiple domains of SF-36 (mental component summary, general health, vitality and mental health). By multivariate Linear regression analysis, current psychiatric morbidities (Beta = -0.396, p = 0.002) and family history of psychiatric illness (Beta = -0.299, p = 0.015) were independent risk factors for poorer mental component summary in SF-36, while dyspepsia severity was the only independent risk factor for poorer physical component summary (Beta = -0.332, p = 0.027). Conclusions: Concomitant psychiatric disorders adversely affect HRQoL in FD patients. The use of GHQ-12 as a reliable screening tool for psychiatric disorders allows early intervention and may improve clinical outcomes of these patients.(J Neurogastroenterol Motil 2010;16:52-60)
