Objective: This study was conducted to isolate the cellulolytic microorganism from the rumen of Holstein steers and characterize endoglucanase gene (Cel5A) from the isolated microorganism. Methods: To isolate anaerobic microbes having endoglucanase, rumen fluid was obtained from Holstein steers fed roughage diet. The isolated anaerobic bacteria had 98% similarity with Eubacterium cellulosolvens (E. cellulosolvens) Ce2 (Accession number: AB163733). The Cel5A from isolated E. cellulolsovens sp. was cloned using the published genome sequence and expressed through the Escherichia coli BL21. Results: The maximum activity of recombinant Cel5A (rCel5A) was observed at 50°C and pH 4.0. The enzyme was constant at the temperature range of 20°C to 40°C but also, at the pH range of 3 to 9. The metal ions including Ca2+, K+, Ni2+, Mg2+, and Fe2+ increased the endoglucanase activity but the addition of Mn2+, Cu2+, and Zn2+ decreased. The Km and Vmax value of rCel5A were 14.05 mg/mL and 45.66 μmol/min/mg. Turnover number, Kcat and catalytic efficiency, Kcat/Km values of rCel5A was 96.69 (s–1) and 6.88 (mL/mg/s), respectively. Conclusion: Our results indicated that rCel5A of E. cellulosolvens isolated from Holstein steers had a broad pH range with high stability under various conditions, which might be one of the beneficial characteristics of this enzyme for possible industrial application.

To investigate the relative nutritive value of sorghum to corn for growing chicken, a total of 300 female Single Comb White Leghorn (SCWL) chicks (3 day old) were fed the diets in which corn portion (60% of diets) were replaced by sorghum grain in various combinations, ie, 60% corn (control), 45% corn + 15% sorghum, 30% corn + 30% sorghum, 15% corn + 45% sorghum and 60% sorghum. Birds were fed starter (0-6 weeks), grower (7-14 weeks) and pullet diets (15-20 weeks) with the age of chicks. Growing performance of four sorghum fed groups were slightly poorer than control group, but there was no consistent tendency among treatments as the sorghum level increased. Body weight of each treatment at 20 weeks of age were 1537 (60% corn), 1467 (45% corn + 15% sorghum), 1466 (30%p corn + 30% sorghum), 1468 (15% corn + 45% sorghum), and 1469 (60% sorghum). Nutrients utilizabilities showed no significant differences among treatments.

To examine the relative nutritive value of sorghum to corn for laying hen, 20 weeks old 240 Single Comb White Leghorn (SCWL) starting layer were employed and randomely assigned to each of 5 experimental diets which were prepared by the substitution of sorghum for corn portion (60%) of the basal diet with various ratios, ie, 60% corn (control), 45% corn + 15% sorghum, 30% corn + 30% sorghum, 15% corn + 45% sorghum and 60% sorghum. Five groups of starting layers employed in this study has been pretreated with the same manner as laying period during the growing period. Hens were fed phase I (21-42 weeks) and phase II diet (43-62 weeks) during the laying period, and laying performances were examined. Egg production rate tended to decrease with the increase of sorghum level. However, only 60% sorghum group showed significantly (p$lt;0.01) low egg production (75.28%) as compared to that of control group (79.42%). In average egg weight and feed efficiency, there was no significant (p$gt;0.05) difference among treatments. Feed consumption tended to decrease with increased sorghum level with no significant (p$gt;0.05) differences among treatments. Egg yolk color index was significantly (p$lt;0.01) affected by the substitution level of sorghum. There were no significant differences among treatments in dry matter, crude protein, crude fat, and total carbohydrate utilizabilities. However, sorghum 60% group showed a little lower crude protein utilizability compared with other dietary groups.

This study was carried out to explore the accuracy of Near Infrared Reflectance Spectroscopy (NIRS) for the prediction of digestibility and energy value of corn silages. The spectral data were regressed against a range of digestibility and energy parameters using modified partial least squares(MPLS) multivariate analysis in conjunction with first and second order derivatization, with scatter correction procedure(SNV-Detrend) to reduce the effect of extraneous noise. Calibration models for NIRS measurements gave multivariate correlation coefficients of determination(R²) and standard errors of cross validation of 0.92(SECV 1.73), 0.91(SECV 1.13) and 0.93(SECV 1.74) for in vitro dry matter digestibility(IVDMD), in vitro true digestibility(IVTD), and cellulase dry matter digestibility(CDMD), respectively. The standard error of prediction(SEP) and the multiple correlation coefficient of validation(R²v) on the validation set(n = 39) was used in comparing the prediction accuracy. The SEP value was 0.80(TDN), 0.01(NEL), and 0.01(ME). The relative ability of NIRS to predict digestibility and energy value was very good for CDMD, total digestible nutrients(TDN), net energy for lactation(NEL) and metabolizable energy(ME). This paper shows the potential of NIRS to predict the digestibility and energy value of corn silage as a routine method in feeding programmes and for giving advice to farmers.
착유우 (搾乳牛) 의 볏짚사료위주 사육시 양조부산물 (釀造副産物) 과 Beet Pulp 첨가에 관한 연구

The current studies were conducted to determine the effect of feeding value of brewers grain(BG) and beet pulp(BP) for lactating cows compared to corn silage and hay with rice straw. Twelve lactating cows were alloted into 3 × 3 Latin square method to determine the effect of feeding BG and BP with rice straw on lactating performance. Whole tract nutritive digestion, ruminal dry matter and protein degradation, and protein retention rate were also examined with sheep. The results obtained from present experiments are as follows; 1. Daily milk yield of cows fed corn silage-hay (CS-H), rice straw-beet pulp-brewers grain (RS-BP-BG) and rice straw-beet pulp (RS-BP) was 18.12, 19.21 and 18.32㎏, respectively. Cows fed RS-BP-BG had the highest milk yield(P$lt;0.05). Milk fat and liveweight were not affected by diets. 2. No significant differences on ruminal C₂ and C₄ concentrations were observed among experimental feeds, but C₃ concentration in CS-H, RS-BP-BG and RS-BP were 13.60, 37.18 and 31.82 mM/L(P$lt;0.05). Sheep fed CS-H had the highest ruminal P (6.2), while those fed RS-BP showed the lowest ruminal P (5.7) (P$lt; 0.05). 3. Percent dry matter disappearances of experimental diets containing CS-H, RS-BP-BG and RS-BP in the rumen were 66.7, 63.3 and 69.2%, and the protein disappearances from nylon bags incubated for 24 hrs were 66.7, 61.3 and 67.1%, respectively. 4. Whole tract dry matter, protein, NDF and ADF digestion was influenced by dietry treatment with highest digestibility in sheep fed RS-BP (P$lt;0.05), while no significant differences on digestibilities of ether extract and crude ash were observed among diets. 5. Sheep fed CS-H had the highest total N intake, and RS-BP had the highest N retention (35.86%) (P$lt; 0.05). 6. Total viable counts of bacteria in the rumen of sheep fed CS-H, RS-BP-BG and RS-BP were 3.63×10^9, 3.91×10^9 and 4.05×10^9/㎖, and cellulolytic bacteria in rumen content was 0.3×10^7, 1.5×10^7 and 1.1×10^7/㎖. 7. Gross income was highest for cows fed RS-BP-BG. In conclusion, the lactating performance was improved when cows were fed RS-BP-BG as a replacement of CS-H

The purpose of this study was to compare kinematic factors according to skill levels of women's Yurchenko motion in new horse vaulting. For this study, 6 national female-gymnasts participated in this experiment. The four digital video cameras(Sony, PD-150) were used to record motions. Kwon3D 3.0 was used to process data and analyze for factors. In conclusion, a high CoG of skilled group in comparison with non-skilled group was produced by a horizontal forward velocity from board contact phase and a vertical velocity at taking off horse vault. Therefore, Increasing the height of leap generated an easy and composed aerial rotation and that performed a safe landing motion.
