Enterobacter agglomerans CBNU45로부터 분리된 제한효소 EagBI 의 특성

토양에서 분리된 Enterobacter agglomerans CBNU45는 type II 제한효소인 EagBI을 생산하고 있음을 발견하였다. EagBI을 DEAE-cellulose, phosphocellulose P11, hydroxylapatite column chromatography를 거쳐 부분 정제하여 그 특성을 알아보았다. EagBI은 여섯개의 염기배열 5’-CGAT${\downarrow}$CG-3’을 인식하고 T 와 C 사이를 절단하여 두개의 염기가 3’-말단쪽으로 돌출된 cohesive end를 형성하였다. EagBI의 반응 최적조건은 10mM Tris-HCl(pH 7.8), 6~10mM $MgCl_2$, 37${\circ}C$이었으며 NaCl이 없는 반응 완충용액에서 가장 좋은 활성을 보였다. EagBI은 $dam^-$와 $dam^+$ 메칠화 DNA도 절단할 수 있으며 65${\circ}C$에서 10분 동안 열처리하였을 때 효소의 활성을 상실하였다. 따라서 EagBI은 PvuI의 isoschizomer이나 NaCl 요구성과 열안정성에서 PvuI보다 편리한 제한효소로 확인되었다. EagBI is a type II restriction endonuclease from Enterobacter agglomerans strain CBNU45 isolated from soil. EagBI was partially purified by DEAE-cellulose, phosphocellulose P11 and hydroxylapatite column chromatography. EagBI recognizes and cleaves the sequence 5'-CGAT${\downarrow}$CG-3' and generates 2-base 3'-protruding cohesive ends. The optimal reaction conditions of EagBI are 10 mM Tris-HCl (pH 7.8), 6-10 mM $MgCl_2$, at 37 ${\circ}C$. The enzyme is maximally active in the absence of NaCl, able to cleave both $dam^-$ and $dam^+$ DNAs, and sensitive to heat treatment (at 65 ${\circ}C$ for 10 min). Therefore, although EagBI is an isoschizomer of PvuI, it is more useful than PvuI in respect of the NaCl requirement and heat-stability.

Nano-sized gadolinium-doped ceria (GDC)/nickel particle-dispersed $La_ySr_{1-y}Ti_{1-x}Fe_xO_3$ (LSFTO)-based composite solid oxide fuel cell anodes were fabricated by an infiltration method and the effects of the GDC/Ni nanoparticles on the anode polarization resistance and cell performance were investigated in terms of the infiltration time and nickel content. The anodic polarization resistance of the LSFTO anode was significantly enhanced by GDC and/or Ni infiltration and it decreased with increasing infiltration time and Ni content, respectively. It is believed that the observed phenomena are associated with enhancement of the ionic conductivity and catalytic activity in the nanocomposite anodes by the addition of GDC and Ni. Power densities of cells with the LSFTO and LSFTO-GDC/Ni nanocomposite anodes were 150 and $300mW/cm^2$ at $800^{\circ}C$, respectively.

The incidence of systemic lupus erythematosus (SLE) is known to be affected by sex hormone. Patients with Klinefelter's syndrome were reported to have abnormal sex hormonal metabolism and their chronic estrogenic stimulation seems to affect the pathogenesis of SLE. Therefore, association of SLE and Klinefelter's syndrome has been considered as a clue of the effect of sex hormone on SLE. We report the first case of Klinefelter's syndrome in a patient with SLE in Korea and discuss the association of SLE with Klinefelter's syndrome.(Korean J Med 59:331-334, 2000)
