        Saccharomyces cerevisiae Cmr1 Protein Preferentially Binds to UV-Damaged DNA In Vitro

        DNA metabolic processes such as DNA replication, recombination, and repair are fundamentally important for the maintenance of genome integrity and cell viability. Although a large number of proteins involved in these pathways have been extensively studied, many proteins still remain to be identified. In this study, we isolated DNA-binding proteins from Saccharomyces cerevisiae using DNA-cellulose columns. By analyzing the proteins using mass spectrometry, an uncharacterized protein, Cmr1/YDL156W, was identified. Cmr1 showed sequence homology to human Damaged-DNA binding protein 2 in its C-terminal WD40 repeats. Consistent with this finding, the purified recombinant Cmr1 protein was found to be intrinsically associated with DNA-binding activity and exhibited higher affinity to UV-damaged DNA substrates. Chromatin isolation experiments revealed that Cmr1 localized in both the chromatin and supernatant fractions, and the level of Cmr1 in the chromatin fraction increased when yeast cells were irradiated with UV. These results suggest that Cmr1 may be involved in DNA-damage responses in yeast.

        The logical thought in the later Moists' Thoughts is called the Moists' Logical Analysis. Their Logical Thought is composed of six books. The Books of the Moist Cannon I and II, the Books of Explanation of the Moist Cannon I and II, Ta-chu and Hsiao-chu. These books contains simple brief, and short sentences. The majority of which define or explain important terms, thus making it difficult to grasp the thought as a whole. The purpose of this paper is to recompose their logics from the standpoint of the traditional logic what they call Pien contains not only logical but also ethical and political contents. Therefore, Pien includes all the value evaluations such as True-False and Good-Bad judgments. Discussions of Pien are conducted, broadly viewed, on the rasis of three analyses-an analysis of terminology which analyzes the names of the facts, analysis of proposition which analyses propositions manifecting ideas, and an analysis of inference which clarifies reasons by way of explanation. The name is a concept, and is correspondingly the term. It is classified, variously according to different methods, as concepts of genus and species or universal and particular concepts, However, in the Moists' Logical Analysis, it is classified simply as the highest concept, genus concept and the singular concept. A concept has two aspects-connotative and donotative-, and the Moists' Logical Analysis Stresses connotation rather than denotation and has been an impediment to the development of formal logic. The analysis analyses variously the corresfondence between the subject and the predicate in the proposition. It fails to derive AEIO by way of the Quantity and the Quality of the proposition. The reason for this is that the Chinese language has no formal device to differentiate the singular and the plural. The quantity of a Sentence in determined by its context, and is not formally manifested, thus making it ambiguous. Inference in the Moists' Analysis has various forms because the Moists, stresses connotation rather than denotation. Therefore, discussion of Inference may involve explanation of instances whose number is too large for this paper and I defer it to a later paper.

      • 흰쥐 적출 대동맥에서의 알파-1 아드레날린성 수용체 탈감작 현상

        목적 : 흰쥐의 적출 대동맥 절편에서 알파-1 아드레날린성 수용체의 탈감작에 따른 수축력 변화를 확인하고 수축력 변화와 관련된 요인들과의 관계를 검정하기 위해 실시되었다. 대상 및 방법 : Sprague-Dawley rat의 대동맥을 적출하여 알파-1 아드레날린성 수용체 선택적 효현제인 phenylephrine으로 탈감작시킨 뒤 수축력과 이완력의 변화를 확인하였다. 결과 : Phenylephrine에 의한 수축력은 phenylephrine 탈감작에 의해 감소되었다. EDRF와 관련이 있는 이완제인 acetylcholine과 histamine에 의한 이완반응은 phenylephrine 처치로서 변화를 볼 수 없었으나 sodium nitroprusside에 의한 이완은 항진되었다. 탈감작현상은 LNMMA와 methylene blue처치로 인해 영향을 받지 아니하였다. KCl에 의한 수축은 phenylephrine 탈감작으로 억제되었으나 phorbol ester에 의한 수축은 phenylephrine 탈감작으로 인해 영향을 받지 아니하였다. 결론 : phenylephrine에 의해 나타나는 알파-1 수용체 탈감작에 따른 수축력 감소 현상은 이완 작용의 변화와는 관련이 없으며 수축기전의 변화, 특히 calcium에 대한 반응성의 변화에 기인함을 시사하였다. We evaluated alpha 1 adrenergic receptor desensitization and decreased contractility in isolated rat aorta. 4 hour exposure to 10^-5M of phenylephrine (phenylephrine exposure) decreased phenylephrine(10^-10 to 10^-5M)-induced contractility. Phenylephrine exposure had no effect on the relaxation by NO releasing agents, such as acetylcholine(10^-9 to 10^-5M) and histamine(10^-8 to 10^-4M), but enhanced nitroprusside(10^-10 to 10^-6M)-induced relaxation. Phenylephrine-induced desensitization was not altered by the presence of methylene blue and N-monomethyl-L-arginine. After phenylephrine exposure KCl-induced contractility was decreased but phorbol ester-induced contractility was not changed. From these results we can assume that prolonged phenylephrine exposure alter calcium-dependent contraction mechanism and involve in alpha 1 adrenergic receptor desensitization-mediated decrease in contractility.

        The moist literature consisted of 86 volumes of 6 groups during the period of the Han Dynasy, of which 53 books of 5 groups remain at the present time, Of these, Six books the books of Moist Canon (Ⅰ,Ⅱ), books of explanation of the Moist Canon(Ⅰ,Ⅱ), Ta-chu(大取), Hsiao-chu(小取)-are called the treatises of the Moist arguments, and considered to be the literature representative of the Later Moist thought. This article is concerned with this literatuer. It is a general opinion that the Chinese Philosopy is devoid of epistemology. But although in-depth studies on problems of logic and knowledge were conducted chiefly by the School of Names and Moists of 4∼3 centries B.C., it is regrettable that what remains of their contents to this day is only fragmentary and not in detail. I interpret the Moist propositions as claiming that perceptive capacity and reasoning capacity are needed in obtaining knowledge. It is again regrettable that the propositions, although constituting an outstanding view that clarified the two kinds of knowledge, one obtainable through perception and the other through reasoning, have faded into oblivion together with their claimant, the Moist. As to the idea of loving human beings, the Confucians idea is basically the same as the Moist's. The confucians claimed that to exercise Jen(仁) originates from human nature and to act in accordance with human nature is goodness. However, the Moists took a Utilitarian position that Chien-Ai(兼愛) is needed to bring forth benefits to the society. Moreover, they also took a Hedonistic position that, in distinguishing goodness and evil, what is beneficial is goodness, while what is harmful is evil, and that a benefit is that which pleases to obtain while a harm is that which displeases to obtain. This Moist thought, whose similar idea arose as the Utilitarianism in modern England, was born too long ahead of the time and as a result I has been forgotten about. For this reason it has been revived as an object of scholarly interests in modern days.

        Hrq1 Functions Independently of Sgs1 to Preserve Genome Integrity in Saccharomyces cerevisiae

        Maintenance of genome stability in eukaryotes involves a number of conserved proteins, including RecQ helicases, which play multiple roles at various steps in homologous recombination and DNA repair pathways. Sgs1 has been described as the only RecQ helicase in lower eukaryotes. However, recent studies revealed the presence of a second RecQ helicase, Hrq1, which is most homologous to human RECQL4. Here we show that hrq1Δ mutation resulted in increased mitotic recombination and spontaneous mutation in Saccharomyces cerevisiae, and sgs1Δ mutation had additive effects on the phenotypes of hrq1Δ. We also observed that the hrq1Δ mutant was sensitive to 4-nitroquinoline 1-oxide and cisplatin, which was not complemented by overexpression of Sgs1. In addition, the hrq1Δ sgs1Δ double mutant displayed synthetic growth defect as well as a shortened chronological life span compared with the respective single mutants. Analysis of the type of age-dependent Canr mutations revealed that only point mutations were found in hrq1Δ, whereas significant numbers of gross deletion mutations were found in sgs1Δ. Our results suggest that Hrq1 is involved in recombination and DNA repair pathways in S. cerevisiae independent of Sgs1.

        Hrq1 Facilitates Nucleotide Excision Repair of DNA Damage Induced by 4-Nitroquinoline-1-Oxide and Cisplatin in Saccharomyces cerevisiae

        Hrq1 helicase is a novel member of the RecQ family. Amongthe five human RecQ helicases, Hrq1 is most homologous toRECQL4 and is conserved in fungal genomes. Recent geneticand biochemical studies have shown that it is a functionalgene, involved in the maintenance of genome stability. Tobetter define the roles of Hrq1 in yeast cells, we investigatedgenetic interactions between HRQ1 and several DNA repairgenes. Based on DNA damage sensitivities induced by 4-nitroquinoline-1-oxide (4-NQO) or cisplatin, RAD4 was foundto be epistatic to HRQ1. On the other hand, mutant strainsdefective in either homologous recombination (HR) or postreplicationrepair (PRR) became more sensitive by additionaldeletion of HRQ1, indicating that HRQ1 functions in theRAD4-dependent nucleotide excision repair (NER) pathwayindependent of HR or PRR. In support of this, yeast twohybridanalysis showed that Hrq1 interacted with Rad4,which was enhanced by DNA damage. Overexpression ofHrq1K318A helicase-deficient protein rendered mutant cellsmore sensitive to 4-NQO and cisplatin, suggesting thathelicase activity is required for the proper function of Hrq1in NER.
