        관중 메탄올추출물이 구강암세포주에 미치는 영향

        Dryopteris crassirhizoma is one of the naturally occurring substance wood ferns and is known forhaving anti-inflammatory, antiviral and anthelmintic activities. However, there is less report about its anticancer effectin human cancer cell lines. In the present study, the effect of methanol extract of dryopteris crassirhizoma (MEDC) onapoptosis in human oral cancer cell lines (MC3 and HN22 cells) was investigated. MEDC inhibited cell viability andinduced apoptosis. MEDC significantly increased Bak and truncated Bid proteins in MC3 cells and elevated only truncatedBid compared to the control while other Bcl-2 family proteins were not altered. MEDC has anticancer activityby inducing apoptotic cell death through the regulation of either Bak or Bid. These findings suggest that its extractpossibly may be used for treating oral cancer.

        SEM Findings on the Dentinal Tubules Changes by Application of the Desensitization Agents

        Objective: In order to observe and to compare several desensitization agents for dentinal tuble changes, Gluma, MS coat, 2% NaF, 10% SnF2 and SE bond were prepared. Methods: 5 extracted 3rd Molar were prepared, and devided with 6 pieces per a tooth for 30 specimens, classified them with 6 groups as group 1 as without any treatment, group 2 as applied with Gluma for 3 times, group 3 as MS coat for 3 times, group 4 as 2% NaF for 4 times, group 5 as 10% SnF2 and group 6 as SE bond for a time. SEM observation was done for all specimens for 2 portions per a sample and the number and degree of the obstacle of dentinal tubules were calculated. Results: It revealed the closing phenomina in all experimental group's samples were more in numbers of the degree than in control group (p<0.05), SE bond group was the most in closing the dentinal tubles and 10% SnF2 group was next, and MS coat, Gluma, and 2% NaF in sequence. There was no significant differences between in each experimental group by one way ANOVA test, with Tuckey or Sheffe test, otherwise there was a significant difference in statistical between in Gluma, 2% NaF and SE bond group by Dunkan test (p<0.05). Conclusion: All experimental group's agents can be used for desensitization agents in hypersensitive dentin clinically, and SE bond was the most recommended for severe hyper sensitive dental patient.

        구강암 세포주에서 실망초 추출물의 항암효능

        Objectives: Erigeron bonariensis is a type of Erigeron found throughout the tropical and subtropical areas as one of the perennial plants or pioneer plants. It is known to show detoxifying, antipyretic, and anticancer effects for various cancers. However, there are no reports on the anticancer effect of E. bonariensis on oral cancer cells. In the present study, we investigated the effects of the methanol extract of Erigeron bonariensis (MEEB) on the inhibition of cell growth and induction of apoptosis in mucoepidermoid carcinoma (MEC) cell lines, including the MC3 and YD15 oral cancer cells. Methods: MC3 Cells were treated by dimethyl sulfoxide (DMSO) or methanol extracts of 20 various natural products 20 μg/mL for 48 hours and cell viability were analyzed as Trypan blue exclusion assay. The effects of MEEB treatment on the cell viability of MC3 and YD15 cells, for 48 h, were analyzed by Trypan blue exclusion assay. The anticancer efficacy and apoptosis of oral cancer cell lines were analyzed by western blot analysis. The statistical significance of differences between groups was analyzed by Student’s two-tailed t-test. A value of P<0.05 compared to the vehicle control was considered statistically significant. Results: Among 20 different naturally derived products, MEEB significantly inhibited cell viability and increased cleaved poly [ADP-ribose] polymerase 1 (PARP) protein in the MC3 and YD15 cells in a concentration-dependent manner. Conclusions: These results suggest that MEEB can be used as a natural anticancer drug for the treatment of human oral cancer.

        Anticancer Effect of Vitamin K2 in MC3 Mucoepidermoid Carcinoma Cells

        Background. Vitamin K (VK) is a fat-soluble vitamin and is known to have anticancer activity in various cancer cell lines. However,there is no report on the anticancer effect of VK2 in mucoepidermoid carcinoma cells. Methods. The effects of VK2 on anti-proliferativeand apoptotic activity were recognized by the trypan blue exclusion assay, 4'-6-diamidino-2-phenylindole (DAPI) staining and Westernblot analysis. Results. The results showed that VK2 decreased cell viability and induced apoptotic programmed cell death in MC3 cellsevidenced by the cleavages of caspase3 and PARP. VK2 treatment clearly increased Bak and truncated Bid (t-Bid) compared withthe control treatment whereas it did not alter other Bcl-2 family members. Conclusions. Overall, our results suggest that VK2 can bea good apoptotic inducer accompanied by the increase in Bak and Bid protein. VK2 may be a potent target of anticancer drug candidatefor the treatment of oral cancer.

        Lycorine induces apoptosis by enhancing protein degradation of survivin in human oral cancer cell lines

        Lycorine, a natural alkaloid extracted from the Amaryllidaceae plant family, was reported to various physiological and pharmacological effects including anti-cancer activity. Nevertheless, there is no report of the anticancer effect of lycorine in oral cancer cells. The effects of lycorine on cell proliferation and apoptosis were examined through trypan blue exclusion assay, 4’-6-diamidino-2-phenylindole (DAPI) stain, Live/Dead assay, Western blot analysis and RT-PCR. Lycorine suppressed cell viability and induced apoptosis in MC3 and HSC-3 cell lines. Lycorine decreased survivin protein but did not affect its mRNA. It regulated survivin through accelerating protein degradation in a time-dependent manner although neither proteasome nor lysosome was not associated with lycorine-mediated protein degradation. Collectively, our results suggest that lycorine may be a potential therapeutic anti-cancer drug candidate for the treatment of human oral cancer.
