        미토콘드리아 크리스테에 존재하는 cytochrome-c-oxidase의 단백질 소단위 분포에 관한 연구

        The topology of the enzyme has been investigated by biochemical studies including chemical labeling and cross linking. Thirteen subunits(polypeptides) of the cytochrome-c-oxidase have localistic characteristics of existing in the matrix side or cytoplasmic side in the mitochondria. In order to observe the distribution of the enzyme subunit on the mitochondria membrane, immunogold-labeling methods were employed. Antibody was obtained from the serum of immunized rabbit with enzyme subunit antigen which was obtained from cytochrome-c-oxidase of the beef heart muscle mitochondria. Beef heart muscle tissue as a tissue antigen was stained with immunized rabbit IgG and protein A gold complex. Electron microscopy has identified the existance of cytochrome-c-oxidase subunit $Mt_I,\;Mt_{II}\;and\;Mt_{III}$ on the membrane of cristae and outer chamber of mitochondria and the subunit $C_{IV}$ on the membrane of cristae and matrix of mitochondria. Particularly, the subunit $C_{IV}$ was also observed to exist in the sarcoplasm of muscle tissue.

        폐흡충에 대한 면역항체의 면역반응 활성에 관한 연구

        This study was performed to investigate the activity of antibodies in the tissues of Paragonimus westermani at the different developmental stages. Enzyme-linked immunosorbent assay (ELISA) and Immunelectron microscopy (IEM) were applied, using the dog sera infected with metacercariae isolated from Cambaroides similis. These dog sera were obtained from 3rd to 96th week after infection by bleeding. The supernatants of homogenated worms for worm antigen were used. The worm tissues were embedded in Lowicryl HM 20 medium, treated with infected serum and protein A gold complex (particle size; 12 nm) and observed by electron microscope. In the pattern of antibody levels by ELISA test in all developemental worm antigens, the activity of antibody was very weak in the 3rd week, but strong in the worm antibody from 4th to 20th week after infection. Its activity was maintained even till 96th week. The antibody level of the L2th week worm antigen was higher than those of the 20th and 48th week worm antigens. Generally, many gold particles were observed on the secretory granules and the epithelial lamellae. Thus, it was concluded that the antigenic materials in the developmental worm tissues were especially concentrated on the secretory granules in the parenchymal tissues and the epithelial lamellae in the lumen of the caecum.

        고들빼기의 고콜레스테롤혈증 개선효과

        Effect of various fractions prepared from the methanol extract of Ixeris sonchifolia and flavonoids isolated from the extract on hypercholesterolemic mice were examined. When various fractions $(CHCl_3,\;Interphase,\;EtOAc,\;n-BuOH\;and\;H_2O)$were administered to mice, the Interphase material produced a significant activity. Total cholesterol level in the Interphase material-treated group given doses of 25 and 50mg was decreased significantly by as much as 16% and 30%, respectively. Among the isolated flavonoids, cynaroside caused a significant hypocholesterolemic activity.

        The Pharmaco - chemical Study on the Plant of Ixeris spp. - 1. Anti - hypercholesterolemic Effect of Ixeris sonchifolia

        고들빼기의 부위별 메탄올 엑스가 고콜레스테롤 생쥐의 혈청 콜레스테롤농도에 미치는 영향을 살펴보았다. 고콜레스테롤 생쥐는 식이에 1% 콜레스테롤과 0.5% 답즙산을 첨가하므로서 유도하였으며, 잎의 메탄올 엑스를 투여한 군은 대조군에 비해 혈청 콜레스테롤 농도를 현저히 감소시켰으나 뿌리의 경우에는 효과가 나타나지 않았다 . 또한, 잎의 메탄올 엑스는 고콜레스테롤 흰쥐에 대해서도 100㎎/㎏ 투여 용량에서 혈청 콜레스테롤 농도를 현저히 감소 시켰으며 동맥경화성 지표를 개선시켰다. 반면에, 정상흰쥐에 대해서는 효과가 나타나지 않았다. 이상과 같은 결과로 잎의 메탄올 엑스는 과잉의 콜레스테롤을 섭취하였을때 체내대사 이용도를 증가시키므로서 혈청 콜레스테롤 농도를 감소시킨다고 생각된다. The methanol extracts from different parts of Ixeris sonchifolia (Compositae) were evaluated for their total cholesterol lowering effect in mice. Mice were rendered hypercholesterolemic with 1% cholesterol and 0.5% cholic acid. Significant lowering in serum cholesterol was observed in mice with the methanol extract from leaves (MeOH-LF), whereas the methanol extract from roots (MeOH-RT) was devoid of this effect. In rats with cholesterol-induced hyperlipidemia MeOH-LF in a dose of 100㎎/㎏ body weight caused significant decrease of total cholesterol, and the atherogenic index was also improved. On the other hand, total cholesterol in rats fed a stock diet was not affected by administration of the MeOH-LF. Thus, it is suggested that this MeOH-LF probably may increase the metabolic utilization only when fed with excess cholesterol.

        Isolation of Adenosine and Free Amino Acid Composition from the Leaves of Allium tuberosum

        부추 잎으로부터 purine nucleoside인 adenosine을 분리하고 그 구조를 기기적인 분석방법에 의하여 동정하였으며 또한 유리 아미노산 관련 화합물들의 조성과 상대함량을 표준품과 동일 조건하에서 아미노산 자동 분석 기기로 비교 검토하였다. 가장 함량이 많은 유리 아미노산들은 alanine, glutamic acid, aspartic acid 그리고 valine이었다. From the leaves of Allium tuberosum (Liliaceae), the purine nucleoside, adenosine was isolated and its structure was characterized on the basis of spectral data. Besides this nucleoside, the composition and relative content of free amino acids and related compounds, compared to standards determined under identical conditions was also investigated using automatic amino acid analyzer. Major free amino acids were alanine, glutamic acid, aspartic acid and valine.

        기름종개 C. sinensis와 왕종개 C. longicorpus가 함께 서식하는 낙동강상류의 여러 수역에서 이들사이의 잡종이라고 추측되는 개체들이 높은 빈도 (43%)로 출현하였는데 그들은 모두 암컷이었다. 이들 잡종 개체들은 체측에 특이한 구름무늬 반문을 가지면서 형태적으로는 기름종개와 왕종개의 중간형질을 보여주고 그들의 염색체수는 2배체형인 49개이거나 3배형인 73개이었다. 이들은 기름종개와 왕종개 사이에 잡종에서 유래된 후 이 종과는 독립된 단성복합집단(unisexual complex) 이라고 생각한다.
