틸라피아의 해수순치에 (海水馴致) 관한 생리학적 연구 Ⅴ. 미세구조적 변화

This study was taken to examine the ultrastructural changes of the gill and kidney of female tilapia (Oreochromis niloticus) living in 0‰, 10‰, 20‰, and 30‰salt concentrations, respectively, by scanning and transmission electron microscope. The ultrastructure by scanning electron microscope (SEM) indicated that the gill secondary lamella of tilapia exposed to seawater, were characterized by rough convoluted surfaces during the adaptation. Transmission electron microscopy(TEM) indicated that mitochondria in chloride cells exposed to seawater, were both large and elongate and contained well-developed cristae. TEM also showed the increased chloride cells exposed to seawater. The presence of two mitochondria-rich cell types may regard to their pos-sible role in the hypoosmoregulatory changes during seawater-adaptation. The kidney tissues in freshwater-adapted tilapia contained most of small, circular mitochondria, and mitochondria had less electron-dense cy-toplasm. The ultrastructure by TEM indicated that the kidney tissues of tilapia exposed to seawater, were characterized by the increased mitochondria during the adaptation. TEM also indicated that mitochondria in kidney tissues exposed to seawater, were both spherical and elongate and contained well-developed cristae. The kidney tissues in seawater-adapted fish contained most of large, elliptic(elongate) mitochondria and mitochondria had more electron-dense cytoplasm.
도입온수어종인 틸라피아의 albumin 과 globulin , transferrin 의 분리 추출에 관한 연구

This study were taken to isolate albumin, globulin and transferrin fractions from the serum of tilapia spp., Tilapia mossambica, by physico-chemical analyses such as the classical salting-out, dialysis, electrophoretic method, ion-exchange chromatography and gel filtration chromatography. The results obtained in these experiments were summarized as follows: 1. The albumin fraction is composed of several components possessing relatively higher electrophoretic mobilities and lower molecular sizes than the globulin components. 2. The molecular weight of albumin component was 89,000 and it possessed a BPB-binding ability. 3. None of the albumin components was found to fit completely the definition of mammalian serum albumin. 4. The molecular weight of the globulin components was more than 295,000. 5. The correlation coefficient of the albumin /globulin ratio by BCG and blood analyzer was +0.599. 6. The molecular weight of transferrin component was about 70,000. 7. Tilapia transferrin fractions were found to be polymorphic. 8. The male transferrin showed two strong bands, while the female transferrin showed a single weak band. 9. one of the two bands of male transferrin showed the same mobility with band of female transferrin. 10. If band above-mentioned had significant effect(p$lt;0.01) upon sex discrimination(about 97%, 31 out of 32 individuals).
한국산 미꾸리에 관한 육종 번식학적 연구 1 . 미성숙과 성숙 단계에서의 한국산 미꾸리의 호르몬 농도 변화

This study was undertaken to understand the changes of hormones levels in Korean loach(Misgurnus anguillicaudatus) measured by ELIA at approximately March and May/`90. Sexual maturation was studied on 100 Korean loath females and 50 males. The results obtained in this study were as follows; 1. Serum LH, FSH, and estradiol-17β levels showed all significant increase in the female loath in March, and the highest peak in April in association with initiation of spawning, and abrupt decrease in Ma,, coinciding with the rapid numerical increase of mature oocytes. 2. In May, coinciding with the histologically mature stage, prolactin and thyroxine levels of the male were all higher than those of the female. 3. Correlation coefficients of serum LH with FSH and estradiol-17β were +0.979 and +0.829. respectively, in the female loath in May. 4. Correlation coefficients of serum prolactin with T₄ and T₃ were +0.765 and +0.99 in the female loath in May. 5. Correlation coefficient of serum LH with FSH was +0.959 in the male loath in May. 6. It is concluded from these observations that LH. FSH and estradiol-17β played an important part in sexual maturation of Korean loach(Misgurnus anguillicaudatus).
한국산 미꾸리에 관한 육종 번식학적 연구 2 . 미성숙과 성숙 단계에서의 한국산 미꾸리의 조직학적 변화

This study was undertaken to understand the changes of development stages of ovary, testis and pituitary of Korean loach(Misgurnus anguillicaudatus) investigated by histological techniques from March to May/`90. Sexual maturation was studied on 100 Korean loach females and 50 males. The results obtained in this study were as follows: l. Gonadosomatic indices(GSI) increased abruptly from April to May. and showed the highest peak in May, coinciding with the numerical drastic increase of mature oocytes in female, and mature spermatozoa and spermatozoa in male, respectively. 2. The ratio of early perinucleolus oocytes, Stage l, was 13.67% and of ripe oocytes was 42.9% in May. 3. The vitellogenic phase began as these late perinucleolus oocytes became transformed into early maturing oocytes through the accumulation of yolk. 4. In pituitary gland in mature loach, ventral lobe occupied major area of gland. Ventral lobe was relatively larger than in sexually immature individuals. Ventral lobe consisted of irregular-shaped masses and columns of uniformly staining cells. Ventral lobe appeared to be more basophilic than acidophilic.
