        Genetic Diversity and Population Structure Analysis of Ever-Bearing and June-Bearing Strawberry Cultivars Using SSR Markers

        This study was carried out to select simple sequence repeat markers (SSR), to identify a DNAprofile database, and to analyze the population structure of 28 strawberry cultivars (Fragaria ×ananassa Duch.) including 22 cultivars of Ever-bearing and 6 of June-bearing strawberry typesusing SSR markers. We used 10 SSR markers to analyze the genetic diversity and populationstructure of 28 strawberry cultivars. A total 135 alleles were detected with an average of 13.5 perlocus. The genetic diversity (1-D) of 28 strawberry cultivars ranged from 0.53 (EMFn226) to 0.89(FxaHGA02P13), with an average of 0.79. Indices of evenness ranged from 0.57 to 0.83 (average0.71). Analysis of the distribution of 28 strawberry genetic groups with different K values wasperformed. With K = 2, it was confirmed that the 28 strawberry genetic groups were divided intotwo. These 10 SSR makers will be useful to identify and differentiate between Ever-bearing andJune-bearing strawberry cultivar.

        국내 감자 품종 판별을 위한 다중 초위성체 마커 세트 개발

        국내 감자품종들의 품종간 유연관계를 분석하고 품종구분을 위한 DNA 표지인자를 개발하기 위하여 SSR(simple sequence repeats) 분석 및 다중초위성체 마커세트(multiplex-SSR set)를 개발하였다. 기존에 보고된 92개의 SSR 마커를 디자인 하고 이들을 이용하여 국내에서 육성된 24개 감자 품종에 대해 유전적 다양성을 분석하였다. 92개의 SSR 마커 중 PIC(polymorphism information contents) 값이 높은 14개의 SSR 마커를 선발하였고 PIC 값은 SSR 마커별로 0.48에서 0.89로 나타났고, 평균 값은 0.79였다. PSSR-29의 PIC 값은 0.48로 가장 낮은 값을 나타내었으며 PSSR-191에서 0.89로 가장 높은 값을 보였다. 선발된 14개의 SSR 마커를 이용하여 UPGMA 집괴분석 결과 24개의 감자 품종 중 21개의 품종이 2개의 집단으로 구분 할 수 있었으며 Ⅰ 집단과 Ⅱ 집단에는 각각 16개, 5개의 품종들이 군집되었으나 3개의 품종은 군집되지 않았다. 선발된 14개의 SSR 마커를 이용한 결과 24개의 품종에서 총 121개의 대립인자가 확인되었으며 각 마커별 대립인자는 3개에서 34개까지 확인되었고 평균 10.8개로 나타났다. 선발된 SSR 마커 중에서 PSSR-17, PSSR-24, PSSR-29 마커를 조합하여 다중초위성체 마커세트(multiplex-SSR set)를 개발하였다. 다중초위성체 마커세트는 한번의 PCR 반응과 PAGE 분석 만으로 본 연구에서 사용된 국내 24개의 감자 품종을 구분할 수 있었며 PIC 값은 0.95로 나타났다. To analyze the genetic relationships among Korean potato cultivars and to develop cultivar identification method using DNA markers, we carried out genotyping using simple sequence repeats (SSR) analysis and developed multiplex-SSR set. Initially, we designed 92 SSR primer combinations reported previously and applied them to twenty four Korean potato cultivars. Among the 92 SSR markers, we selected 14 SSR markers based on polymorphism information contents (PIC) values. PIC values of the selected 14 markers ranged from 0.48 to 0.89 with an average of 0.76. PIC value of PSSR-29 was the lowest with 0.48 and PSSR-191 was the highest with 0.89. UPGMA clustering analysis based on genetic distances using 14 SSR markers classified 21 potato cultivars into 2 clusters. Cluster I and II included 16 and 5 cultivars, respectively. And 3 cultivars were not classified into major cluster group I and II. These 14 SSR markers generated a total of 121 alleles and the average number of alleles per SSR marker was 10.8 with a range from 3 to 34. Among the selected markers, we combined three SSR markers, PSSR-17, PSSR-24 and PSSR-24, as a multiplex-SSR set. This multiplex-SSR set used in the study can distinguish all the cultivars with one time PCR and PAGE (Polyacrylamide gel electrophoresis) analysis and PIC value of multiplex-SSR set was 0.95.

        감자 풋마름병 저항성 유전자원 선발 및 RAPD를 이용한 유전적 다양성 분석

        본 논문은 감자 풋마름병에 대한 저항성 계통을 선발하고 이들에 대한 유전적 다양성 평가를 위하여 RAPD 분석을 실시하였다. 감자 풋마름병에 대한 저항성 검정 방법은 양액재배 방식을 이용하였으며 고령지농업연구센터에서 보유한 감자 유전자원 440계통을 대상으로 풋마름병원균 race 1과 3에 저항성 계통을 선발하였다. 1차 선발에는 race 1 또는 race 3에 저항성인 계통을 선발하여 모두 72개의 계통을 선발하였으며, 2차 선발은 1차 선발 계통을 재평가하여 race 1, 3에 모두 저항성인 계통을 선발하였다. 2회에 걸쳐 병저항성 평가를 통하여 AG14075 등 총 20계통을 선발하였다. 선발된 20개의 계통간의 유전적 다양성을 알아보기 위해 URP 프라이머를 이용하여 RAPD 분석을 실시하였다. 11개의 URP 프라이머를 이용하였을 때 각각의 primer에서 5개에서 7개의 다형성 DNA 밴드를 발견할 수 있었다. RAPD분석 결과 유전적 유사도 지수는 0.56에서 0.82의 범위에 속하였다. 선발된 계통은 유전적 유사도 지수 0.56에서 2그룹으로 나눠졌다. 제 1그룹은 AG14075을 포함한 4계통이 포함되었고 제 2그룹은 AG14252을 포함한 16계통이 포함되었다. 제 2그룹에 속한 16계통은 유전적 유사도 지수 0.59에서 2개의 하위 그룹으로 나뉘어졌으며, 첫 번째 그룹은 AG34326을 포함한7계통이, 두 번째 그룹은 AG14252을 포함한 9계통이 포함되었다. 본 연구 결과를 통해 선발된 풋마름병 저항성 유전자원은 차후 육성모본으로 이용될 수 있을 것이며 RAPD 분석을 이용한 유전적 유연관계 분석 결과는 새로운 저항성 품종 육성을 위한 교배조합 작성에 이용될 수 있을 것을 판단된다. This study was carried out to select potato (Solanum tuberosum) clones resistant to Bacterial wilt (BW) disease (Ralstonia solanacearum) and evaluate genetic diversity with RAPD (Randomly amplified polymorphic DNA). A total of 440 clones collected and maintained at Highland Agriculture Research Center were tested in the hydroponic culture system with R. solanacearum race 1 and 3. After 40 days in dipping in hydroponic culture system, the resistance was evaluated as the range from 0 (resistance) to 4 (susceptible). Seventy-two clones were selected in the first screening as a resistant to race 1 or 3 in 2007, and the selected lines were tested again as the same procedure above. After the second screening, a total of 20 lines were selected as resistance to BW in 2008. For the evaluation of genetic diversity of the selected 20 clones, RAPD analysis was carried out with potato URP primer sets. From the 11 URP primers, 5 to 7 polymorphic DNA bands were amplified in selected resistant clones with each primer. With RAPD analysis, the genetic similarity was shown from 0.56 to 0.82. The selected clones were separated into two distinct groups at the genetic similarity value point of 0.56. Four clones including AG14252 were integrated into the first group, and the others, 16 clones, were grouped in the second group. In the second group, the two sub-groups showed genetic similarity value of 0.59. Seven clones including AG34326 and nine clones were separated into the first and second sub-groups, respectively. The results have revealed that bacterial wilt resistance test using hydroponic culture is favorable for the selection of BW resistant potato clones, and that RAPD analysis is useful for the identification of genetic similarity. The selected potato clones could be used as parent clones in BW resistance breeding program of potatoes.

        칩가공 품질이 우수하고 역병에 강한 감자 품종 '만강'

        Potato late blight caused by the fungus Phytophthora infestans is one of the most important diseases worldwide. Developing potatoeswith resistance to late blight through breeding programs is a key research interest in Korea. ‘Mangang’ was bred for late blight resistancethrough a breeding program at the Highland Agriculture Research Institute of National Institute of Crop Science. The population derived fromthe cross between ‘Atlantic’ as the female parent, which has excellent chipping quality and ‘CIP393371.159’ as the male parent, a late blightresistantclone introduced from International Potato Center (CIP), was selected. In ‘Mangang’, the plant maturity occurs late and the growth habitis of the erect type. The tuber shape is round and eye-depth is medium. The skin color of ‘Mangang’ is light yellow; however, there arepink spots around the eyes of the tuber, and the flesh color is white. Leaf color is green and flower color is light-purple. ‘Mangang’ exhibitsresistance to early blight (Alternaria solani) and leaf roll virus (PLRV). In addition, it has moderate resistance to common scab (Streptomycesscabies). The response to late blight was evaluated in a non-agrochemical treated field for five years between 2012 and 2016; ‘Mangang’exhibited resistance to late blight. The mean AUDPC (Area Under Disease Progress Curve) values of ‘Mangang’ and ‘Atlantic’ were 165.5 and795.8, respectively. Total tuber yield from the non-agrochemical application trial at three regions during the five years was 40.3 ton/ha; thiswas 40.4% higher than that from ‘Atlantic’. The dry matter content of ‘Mangang’ was 21.3%, which was slightly higher than that of ‘Atlantic’. The chip color was 66.5, and the chip processing quality of ‘Mangang’ was at a level similar to that of ‘Atlantic’ (Registration No. 9312).

        Vitamin C Content of Potato Clones from Korean Breeding Lines and Compositional Changes during Growth and after Storage

        Vitamin C is the main vitamin source in potato tubers, and its antioxidant activity is known to benefit human health. To improve the vitamin C content and understand its accumulation patterns, we selected 10 advanced clones with high vitamin C content from 268 clones. The levels of the selected clones ranged from 32.3 to 38.9mg·100 g-1 fresh weight. Among them, four double-cropping clones with short dormancy were selected for further analysis. The vitamin C content of the potato tubers from six potato clones, including two control cultivars, ‘Dejima’and ‘Chubaek’, was determined on specific days after planting (DAP) and during development. Developmental stages were categorized by weight: I (5-10 g), II (30-40 g), III (70-80 g), and IV (> 120 g). Vitamin C content was highest 90 DAP and dropped slowly by 100 and 110 DAP. Until 90 DAP, we found that developmental stages I and II showed higher contents of vitamin C; however, there were small differences by developmental stage (I to IV) by 100 and 110 DAP. In conclusion, vitamin C content was higher in younger tubers than in mature tubers, and its content was highest 90 DAP in the selected clones. During storage, vitamin C content significantly declined in all clones. The decrease was highest in H06035-4 and H06085-2 compared to ‘Chubaek’ and ‘Dejima’, and the differences between selected clones and control varieties narrowed after 4 months of storage. When we consider the agronomic characteristics and consistency of high vitamin C content, H06035-4 was selected as a high vitamin C content promising line.
