Catechol-O-methyltransferase (COMT; E. C. 2.1.1.6) from rat liver was purified and characterized. The purification steps involved acid precipitation, ammmonium sulfate fractionation, Sephadex G-100 gel filtration chromatography, and DEAF-cellulose ion-exchange chromatography. COMT was purified 147-fold with 23% recovery. On the SDS-PAGE, the final preparation showed one major band and a few minor bands. The K_m value of COMT for p-nitrocatechol was 3.8 μM and that for SAM was 283 μM. Magnesium ion was required for the optimal enzyme activity and the pH optimum was 7.8 with phosphate buffer. Maximum activity was exerted at 50℃. High ionic strength resulted in inactivation of the enzyme. The molecular weight was estimated as 25,700 by SDS-PAGE. When COMT was treated with a sulfhydryl group modifying reagent, iodoacetic acid, the inhibition of the enzyme activity was observed.

In order to investigate the effect of zinc on the structure and mechanical properties of the silver base dental amalgam, the experiment was performed by setting the zinc content from 0 to 8% in Ag-Sn system alloy for dental application. From this experiment following results were obtained ; 1. γphase in the pre-alloy structure decreased as zinc content increased, whereas free Sn and Ag-Zn phase increased as zinc content increased. 2. Unreacted γphase in the amalgam structure decreased as zinc content increased, whereas γ₂phase increased as zinc content increased. 3. Hardness of γphase in the pre-alloy increased as zinc content increased. 4. Hardness and compressive strength of amalgam alloy containning zinc up to 3.7% increased as znic content increased, and then dereased with much more zinc content. 5. Hardness and compressive strength of amalgam alloy increased with aging.

In order to investigate the effect of mercury-alloy ratio, particle size, particle shape and adding elements on triturations, photographic observation of trituration process and microstructual observation of amalgam at low temperature have been observed, and also wettability and temperature measurement of the capsule during trituration carried out. The results obtained from this study were as follows. 1. Trituration of prealloy and mercury in amalgametor was mainly influenced by wettability of prealloy particle. 2. The time required for trituration of prealloy particles decreased with increasing mercury-alloy ratio and particle size. 3. According to microstructural observation at low temperature, Ag-Hg and Sn-Hg phase appeared in early stages of amalgam.

본 연구는 중고생들의 자원봉사활동과 리더십과의 관계를 규명하는 데에 목적이 있다. 이를 위하여 본 연구는 문헌 연구와 조사연구를 병행하였다. 조사대상은 경기도 소재 고등학교와 서울시 소재 중학교 재학생 402명이었고 자료는 SPSS WIN 12.0(Statistical Package for Social Science)를 이용하여 인구사회학적 특성, 중고생자원봉사활동과 리더십과의 관계를 살펴보았다. 연구 결과, 중고생자원봉사활동 총시간, 자원봉사활동 태도, 자원봉사활동에 대한 만족도가 리더십을 향상시키는데 영향을 미치지 못하였다. 이러한 결과는 본 연구 대상자의 자원봉사활동 시간이 자원봉사활동을 집중적으로 하기에는 비교적 짧은 1회 1~2시간이었다는 점, 자원봉사활동 태도와 만족도가 긍정보다는 부정적인 사례가 더 많았던 것과 관련이 있을 것으로 보인다.따라서 본 연구의 연구 결과를 토대로 볼 때 단순히 청소년 자원봉사활동만으로는 리더십이 높아지지는 않는다는 것이다. 청소년 자원봉사활동이 리더십을 향상시키기 위해서는 보다 철저한 자원봉사활동의 관리가 필요할 것으로 판단된다. The Study is for the purpose of look into the relationship between teenagers' voluntary service experi-ence and leadership. To this end, the present study studied literature and conducted a research. the study subjects were high school students from Gyeonggi-do as well as junior high school students from Seoul. 438 students in all. From the collected questionnaires, 36 deemed inappropriate for using as data were discarded with a total 402 questionnaires used for the analysis. SPSS WIN 12.0(Statistical Package for Social Science) was used to analyze the data, in order to find out social-demographic characteristics, the relationship between teen-agers' voluntary service experience and leadership life skill.In result of this study, total hours, attitude, satisfaction degree of teenagers' voluntary service don't affect to upgrade leadership life skill. This result are caused by the below to fact. First, this research project's vol-untary service is only 1~2 hours which is very short of concentrating in voluntary service. Two, the attitude and satisfaction degree of voluntary service has more negative case that positive case. So, this research show that a simple teenagers' voluntary service doesn't upgrade leadership life skill. We conclude volun-tary have to be managed more thoroughly to increase leadership life skill by teenagers' voluntary service.

본 연구에서는 내수압용 동합금과 최적의 주조조건을 규명하고저 다음과 같은 내용으로 실험하였다. 주형재료는 자연사 합상사 및 Resin Coated sand의 3종에 대하여 검토하였고 주조방안에 있어서는 탕구비가 1:0.7:0.5(가압탕구계)와 1:2:1(비가압탕구계)의 두 경우에 대하여 검토하는 동시에 Screen을 탕구계에 설치하여 그의 영향에 대해서도 검토하였다. 그리고 용해조건 및 주입온도에 있어서는 용해지금의 장입내용, 용해분위기, 탈개스처리 및 주입온도를 각각 달리하여 응고속도, 내수압, 화학성분 및 기계적성질 등을 비교 검토하였다. 본 연구결과에 의하면 주형재료로서는 합성사 생형이 가장 우수하였고 주조방안은 주입구와 탕도의 접합부에 Screen을 설치한 비가압 탕구계가 훨씬 좋았으며 용해분위기는 다량의 Scrap을 사용할 경우는 산화성 분위기가 환원성분위기 보다 좋은 결과를 가져왔다. 이는 Allen & Hewitt의 산소-수소 평형곡선으로서 뒷바침된다. 그리고 장입하기 전에 반드시 자력선별에 의하여 철분을 제거하는 것이 바람직하였고 본 실험에서의 주입온도는 1,150℃가 가장 양호하였다. The present work is concerned with the effects of molding material, gating system, melting condition & pouring temperature on the soundness of Cu-alloy castings. These effects were evaluated by the measurements of solidification rate, leakage, chemical composition and mechanical properties. Because the soundness of castins not only depend on solidification rate, chemical composition and mechanical properties, but also depend on leakage of hydraulic pressure. The castins were poured in three different mold; natural bonded sand, synthetic sand, and resin coated sand. A 5㎏ melt of each alloy was made by degassing with Cu-P alloy in cryptol furnace under various conditions to determine the effects mentioned above. On the basis of these experimental results, favorable condition of casting process was proposed as follows. 1. Synthetic sand mold was more effective than the others. 2. Unpressurized gating system having screen at juncture between the runner and ingate would be more desirable in order to avoid the turbulence and non-metallic inclusion such as dross and sand. 3. When scraps were only charged degassing was desirable after melting under oxidizing condition. This reason may be rationalized by equilibrium curve of oxygen and hydrogen proposed by Allen & Hewitt. 4. The optimum pouring temperature was found to be 1150℃.
