Eukaryotic translation initiation factor 5A (eIF5A) is the only hypusine-containing protein, which is formed by deoxyhypusine synthase (DHS) and deoxyhypusine hydroxylase (DOHH). DOHH is a novel metalloenzyme with HEAT [named for human huntingtin (H), elongation factor 3 (E), a subunit of protein phosphatase 2A (A), and the target of rapamycin (T)]-repeat motifs. Inspite of much progress in determining the roles of iron-containing DOHH holoenzyme as an eIF5A hydroxylase, little is known about iron-free apoenzyme of DOHH under certain stress conditions. For this purpose, we compared cell growth in two yeast strains subjected to oxidative damage. Thus, the existence of more viable cells in the Saccharomyces cerevisiae BY4743 (parental yeast) strain than in the DOHH-null strain after H2O2 treatment indicates the importance of DOHH in protecting yeast cells against oxidative stress. To identify endogenous target proteins influenced by DOHH under oxidative damage, proteomic analysis was applied to the two yeast strains. Of these proteins, the oxidized form of peroxiredoxin I (PrxI) was concomitantly up-regulated in both strains under H2O2 treatment. Two-dimensional electrophoresis (DE) followed by immunoblot analysis shows that the recovery of the oxidized PrxI to the reduced enzyme under H2O2 treatment was found to be much slower in the DOHH-null strain than in the parental strain. Based on the results, we discovered a possible interaction between DOHH and PrxI by immunoprecipitation/immunoblotting in yeast under oxidative stress. Taken together, these results suggest that DOHH might be a candidate protein for protection of yeast cells against oxidative stress in conjunction with PrxI.

In this study, we present the results of performing the chloroplast genome analysis of the Hadong Cheonnyeon cha (HCNC), Camellia sinensis var. sinensis (CSS), and we achieved this by employing illumina sequencing technology. The total nucleotide sequence of 156 Gbp was sequenced and the size of the genome was measured by k-mer analysis, and was found to be about 3,109 Mbp. The sequence corresponding to the chloroplast was obtained, and the analysis was performed again to finally complete the chloroplast of 157,025 bp. The chloroplast had 126 protein coding genes with an average length of 2.4 kb and 40 non-coding RNA. It was confirmed that there were almost no structural differences from the 11 NCBI registries and there was only a difference in the nucleotide sequence. As a result, HCNC was clearly distinguished from assamica (GenBank JQ975030.1, MH394407.1 ∼ MH394410.1), and HCNC was most similar to KF562708.1. The GenBank registered KF562708.1 is one of the Chinese representative varieties knows as Longjing 43. Although the Chinese tea cultivars were registered in GenBank and they were compared with the HCNC, the Japanese cultivars were not registered in there. Further analysis of chloroplast genomes such as Yabukita, Hushun and Samidori were conducted to further investigate the relationship between the HCNC and Japanese tea cultivars. 차세대염기서열(NGS) 분석과 illumina sequencing 기술을 활용하여 하동 천년차나무의 엽록체 유전체 서열을 조사하고 NCBI에 등재된 차나무 11종의 염기서열을 비교하였다. 생산된 총 염기서열은 156 Gbp, GC 비율은 38.3%로 식물에서 전형적으로 나타나는 분포를 보였다. 등록된 11종의 서열에 mapping하여 확보한 최종 엽록체 서열은 157,025 bp를 확인하였다. 이 서열에 대한 유전자 예측결과 총 166개의 유전자를 확인하였고 이중 단백질 암호화 유전자의 평균 길이는 2.4 kb, 비암호화 유전자는 40개 정도였다. Circle map 구조는 다른 11개 서열과 거의 동일하였으며, 염기서열의 크기에서 근소한 차이를 보였다. Phylogenetic tree로 근연관계를 확인해본 결과 대엽종(assamica)인 JQ975030.1, MH394407.1 ∼ MH394410.1과 뚜렷한 차이를 보였으며, Longjing 43 (GenBank : KF562708.1)과 유사하였다. 중국 차나무 품종은 GenBank에 등록되어 있어 비교가 가능하였으나 일본 품종 중에 등록된 것은 없어 Yabukita, Hushun, Samidori 등의 엽록체 유전체 분석을 추가로 실시하여 하동천년차나무와 일본 차나무 품종의 근연관계를 추가로 연구하고자 한다.
The Impact of the Korean Wave, Power Structures, and Globalization

The purpose of this paper is to discuss the changing and expanding roles of popular culture of and on Asia (ns) in this media globalization era with a focus on Korean wave. This paper also addresses the continuities of power structures controlled by governments and global media industries. While governments try to expand their influences by recourse to popular culture, the media giants reinforce their power by co-opting ideas and incorporating talent from around the world. In other words, governments and global media industries are dedicated to exploiting the market-oriented media system rather than pursuing an equitable representation of Asians and Non-Asians alike and democratizing the media. This situation calls for civil society to discuss the publicness of media, which is easily disregarded in today’s increasingly interconnected, commercial media environment.

본 연구는 국내에서 육성된 차나무 품종별 홍차 제다 특성 비교에 필요한 기초 자료를 얻고자 수행되었다. 국내 육성 차나무 품종인 참녹, 금설, 비취설 3종을 하동지역의 전통 ‘햇볕 잭살’ 방식으로 홍차를 제다하였다. 총카테킨함량은 재래종 대비 비취설 3.04%, 금설 0.73%, 참녹 0.18% 높았으며, 카페인 함량은 비취설 1.54%, 금설1.36%, 참녹 1.16%, 품종별 테아플라빈 함량은 참녹 0.54% > 금설 0.52% > 비취설 0.38% 순서로 참녹이 상대적으로 빠르게 산화가 진행되었음을 알 수 있었다. 전체유리아미노산 함량은 금설이 가장 높았다. 8개 맛(신맛, 쓴맛, 쓴맛 후미, 떫은맛, 떫은맛 후미, 감칠맛, 감칠맛 후미, 짠맛)의 합산치는 비취설 10.6, 금설 2.1, 참녹 -1.7, 맛의평균치는 비취설 1.3 ± 1.8, 금설 0.3 ± 1.5, 참녹 -0.2 ± 1.9로 3개 품종 중 비취설이 맛이 풍부하였다. 향기성분은초콜릿 향과 꽃향은 금설보다 비취설이 우수하며, 청량함은 금설이 높은 경향을 보였다.

Emergence and early growth changes of stratification condition of tea (Camellia sinensis) seeds were investigatedin 7 treatments (control, pH 4, pH 10, 70% ethanol (EtOH), 10 mM H2O2, 100 mM H2O2, and physical shock (5.9 J)). Ethanoltreatment was toxic and did not induce emergence. The emergence rate was 36.7% in the control, 26.7% under pH 10, 46.7% underpH 4, 48.3% under physical shock, 51.7% under 10 mM H2O2, and 65.0% under 100 mM H2O2 treatments. It was higher byapproximately 178% in the H2O2 treatment as compared to the control. Plant height was 6.5 cm in the control, 6.6 cm under pH10, 7.6 cm under pH 4, 7.8 cm under physical shock, 8.3 cm under 10 mM H2O2, and 9.1 cm under 100 mM H2O2 treatments. Leaflength and leaf width were also higher under the H2O2 treatment. Therefore, hydrogen peroxide treatment induced emergence andincreased the uniformity of early growth. 본 연구는 차나무 종자의 교배육종에 위한 품종 육성의체계적인 기술확립을 위해 과산화수소 등 다양한 파종전처리를 통하여 차나무 종자의 출아율 및 유묘 소질을 알아보고자 수행하였다. 1. 무처리구의 출아율은 36.7%로 나타났고, 염기성 조건(pH10)에서는 출아율이 26.7%로 가장 낮은 발아율을 보인반면, 100 mM 과산화수소에서 가장 높은 65.0% 출아율을 보였다. 2. 출아율과 출아율지수는 처리에 따라 유의성이 나타났으며, 평균출아일수는 유의성이 나타나지 않았다. 3. 처리간의 평균출아일수는 과산화수소(10, 100 mM)와 물리적 충격 처리구에서 가장 짧은 46.5~47.0일이 소요되었으며, 출아율지수는 100 mM 과산화수소 처리구에서가장 높은 1.363 d-1m-1을 보였다. 4. 파종 60일 후에 묘 소질에서의 초장, 엽수, 엽장 및 엽폭 모두 조사에서 100 mM 과산화수소 처리구에서 9.1 cm, 4.4개 5.5 cm, 3.1 cm로 가장 크거나 많은 것으로 나타났다.

초국가적인 문화 전파와 인구 이동의 폭발적 증가로 인해 이제 우리의 이웃에서 동남 아시아 출신 외국인과 태국 혹은 베트남 음식점을 접하는 것은 그리 어렵지 않은 일 이며, 패밀리레스토랑 빕스의 샐러드바에서 인도네시아 음식 나시고렝은 인 기 메뉴가 되었 다. 소위 “동남아 현상”이라 일컬을 만한 이러한 새로운 환경 속에서 영화, 음 악 등 대중문화 가 동남아시아로부터 유입되고 있으며, 동남아를 주제로 한 혹은 동남아적 요 소를 첨가한 다 양한 장르의 대중문화물이 제작, 유통, 소비되고 있다. 최근 국내외적으로 선 풍적인 인기를 끌고 있는 걸그룹 2NE1의 멤버 산다라 박은 필리핀 교포 출신이며, 보이밴드 2PM의 닉쿤은 태국계 혈통이다. 2009년에는 동남아시아 출신 노동자가 주연을 맡은 영화 < 반두비>가 출시 되어 비평가들의 주목을 받은 바 있으며, 발리 혹은 싱가포르 등 동남아시아 를 극중 장면으로 삽입한 TV 드라마는 헤아릴 수조차 없을 정도다. 이에, 본 연구는 “동남아로 부터의 대중문화 현상”의 실체가 무엇인지, 이를 추동하는 요소에는 어떠한 것들이 있는지, 이 러한 새로운 현상의 의미는 무엇인지를 대중문화와 문화 흐름에 관한 이론적 체계 속에서 기 술, 분석, 평가 한다. It is remarkable that it has become commonplace to find people and cultural forms from Southeast Asia these days in Korea. In the realm of popular culture, more films and musical elements are flowing from Southeast Asia. For example, a K-pop singer Nichkun is Thai, a girl group 2NE1 member Sandara Park is from the Philippines, and a popular band Mocca is Indonesian. However, a so-called Southeast Asian Cultural Wave is not as big as the Korean Wave, and it is surmised that the former in Korea is only possible with the influences of the later in Southeast Asia. In addition, there are few research on Southeast Asian Cultural Wave as of now. This research is to describe, analyze and evaluate this new phenomenon in the Korean society.
국내육성 차 품종별 홍차(잭살차)의 성분 및 항산화 특성

Jaeksal black tea was made from the leaves of three Korean tea varieties, ‘Chamnok’, ‘Keumsull’, and ‘Beachwisull’. Among them, the tea components and antioxidant activities were analyzed to determine which variety is suitable for the black tea type. The black teas were prepared by the traditional Jaeksal fermentation method, where all steps are performed under sunshine. The contents of total phenolic compounds were highest in ‘Beachwisull’ at 171.9 mg GAE/g. In addition, the catechins and caffeine contents were much higher in ‘Beachwisull’ than in ‘Keumsull’ and ‘Chamnok’. The contents of theaflavins were higher in the black tea of 'Chamnok' and 'Keumsull' than that of 'Beachwisull'. The DPPH and ABTS radical-scavenging activities were highest in the 'Beachwisull'. A comparison of the cultivars revealed 'Beachwishull' black tea to have higher contents of total phenolic compounds, catechin, and caffeine, as well as higher antioxidant activity, whereas the theaflavin contents were higher in 'Chamnok'. Therefore, the 'Beachwisull' cultivar is most suitable for black tea characteristics.

Against the backdrop of cultural globalization and regionalization, a new transnational cultural phenomenon called Korean Wave has occurred in the past decade in Asia. This study observes the development of Korean Wave in Singapore and examines the ways in which Korean women diaspora enjoy homeland television dramas and make meanings while living in Singapore. It is found that Korean drama viewing provides women with cultural resources for them to tactically resist the male dominance in gender hierarchy. The Korean Wave phenomenon plays an interesting role in this semiotic juncture. Moving the cultural industries' position upwardly in Korean economy, it has allowed the expertise about Korean television drama to function as an agent of empowerment for women in familial power relations. It is, however, found that such an empowerment is limited in its political efficacy. Korean Wave has also heightened the ethnic pride among Korean women in their relations with other Asians. Articulated to these contexts, Korean Wave produces new meanings and serves new cultural interests for my subjects.

Nitrogen fertilizers are essential for productivity and quality improvement of all crops. However, excessive nitrogen can cause physiological disturbances. Therefore, depending upon the type of crop and cultivation method, controlling the amount of fertilizer is important. Typically, the proper amount of nitrogen fertilizer is determined by the organic matter content of the soil in which the plants (tea plants in our case) are grown. However, there is no difference in the amount of fertilizer applied during shading which leads to serious problems. The current study investigated the physiological activity of tea plants according to nitrogen concentration in oil cake during shading cultivation. The physiological activity expressed via photosynthetic ability showed that the photosynthetic ability remained steady without being significantly affected by nitrogen content up to 9 to 12% under non-shading conditions, whereas nitrogen excess disorder started to occur as the treatment period increased under shading conditions. After 3 weeks of treatment, the leaf chromaticity was the highest at 67.67 for 4.5% oil cake under shading condition, while the highest was 64.19 for 12% oil cake under the non-shading condition. In terms of inorganic nitrogen content in the soil, the NH4+-N content and the NO – -N content under shading were 1.3 times and 1.9 times higher, respectively than those for the non-shading condition. With increase in the nitrogen content of oil cake, the inorganic nitrogen (NO3– N, NH4+-N) also increased.The total free amino acid content was 614 mg·100g-1 at 18% nitrogen content when not shading, and 1,073 mg·100g-1 at 18% nitrogen content when shading was the highest. There was a 2.0-fold increase for shading (401 mg·100g-1) compared to non-shading (200 mg·100g-1) that was not treated with oil cake. Overall, the total free amino acid showed a tendency to increase as the nitrogen content increased, but both non-shading and shading showed a relatively decreased tendency for the straight oil cake (N 12%) treatment. Considering the growth and component content, the most effective nitrogen fertilizer was oil cake with 9% nitrogen content for non-shading cultivation and 4.5% nitrogen content for shading cultivation. 질소시비는 모든 작물에서 생산성 및 품질 향상에 반드시 필요하나 과잉시 생리장애가 발생할 수 있기 때문에 적정 시비량이있으며, 작물별, 토양별, 재배방법에 따라 조절이 필요하다. 차나무의 재배 토양의 유기물 함량에 따른 적정 질소시비량은 정해져 있으나 차광재배시 시비량에 차이를 두지 않으며 과잉에 따른 문제가 발생할 수 있다. 본 연구는 차광재배시 유박의 질소 농도에 따른 차나무 생리활성을 알아보았다. 광합성능력을 통한 생리활성은 비차광의 경우 질소 함량이 9–12%까지 큰 영향을받지 않고 꾸준한 광합성능력을 보이는 반면, 차광의 경우 처리기간이 길어질수록 질소과잉 장애가 발생하는 것으로 보였다. 처리 3주 후, 엽색도는 차광조건은 4.5% 유박에서 67.67로 가장 높았으며, 비차광조건은 12% 유박에서 64.19로 가장 높았다. 토양 내 무기질소 함량 변화는 차광조건이 비차광조건보다 암모늄태 질소는 1.3배, 질산태 질소는 1.9배 높은 함량을 보였다. 유박의 질소함량이 증가함에 따라 무기태질소(질산태, 암모늄태) 일반적으로 증가하는 경향을 보였다. 전체 유리아미노산 함량은 비차광시 질소함량 18%에서 614mg·100g-1, 차광시 질소함량 18%에서 1,073mg·100g-1으로 질소함량 18%에서 가장높았다. 유박을 처리하지 않은 비차광(200mg·100g-1) 대비 차광(402mg·100g-1)에서 2.0배 증가하였다. 전체적으로 질소함량이 높을수록 증가하는 경향을 보였으나 비차광, 차광 모두 속효성 유박인 질소함량 12% 처리구에서 상대적으로 감소하는경향을 보였다. 총 카테킨 함량은 차광 N-0에서 9.2%, 차광 N-4.5에서 9.17%로 가장 높았으며, 비차광 N-12에게 6.18%로 가장 낮았다. 생육 및 성분함량 등을 고려할 때 유묘 및 제한된 환경을 기준으로 비차광 재배시에는 질소함량 9%, 차광 재배시에는 질소함량 4.5% 유박을 시비하는 것이 가장 효율적 질소시비율로 사료된다.
