        Immunomodulatory effects of mixed Weissella cibaria JW15 with water extract of black soybean and burdock on Listeria monocytogenes infection in mice

        The objective of this study is to assess immunomodulatory effects of mixed Weissella (W.) cibaria JW15 strain with water extract of black soybean (Glycine max) and burdock (Arctium lappa) on Listeria (L.) monocytogenes infection in mice. Female 7-9 week old BALB/c mice were given a daily dose of 1 × 109 CFU of viable JW15 and JW15 mixed with black soybean (BS) and burdock (BD) in 200 μL PBS for 2 weeks. The nomal control group (NC) and positive control group (PC) were given 200 μL PBS. After 2 weeks, mice were infected with L. monocytogenes (1.0 × 105 CFU/mouse) via the tail vein. The NC was injected with 100 μL PBS without L. monocytogenes. After 2 days, mice were euthanized and their body weights were determined. In addition, their livers and spleens were weighed, and serum were analyzed for cytokine (Interleukin-1β (IL-1β) and Tumor necrosis factor-α (TNF-α)) production. The survival rate was monitored using 5 mice in each group in the same way above until the mice died. Two days after infection with L. monocytogenes, mean spleen weight per body weight (g/kg) of JW15 (5.4 ± 0.88 g/kg), JW15 + BS (6.0 ± 0.64 g/kg), and JW15 + BD (5.3 ± 0.38 g/kg) group were significantly lower than that of the PC (6.8 ± 0.57 g/kg). The level of IL-1β in the serum of JW15 + BD (113.6 ± 31.03 pg/mL) was significantly higher than that of the JW15 (67.9 ± 15.15 pg/mL). Collectively, combination W. cibaria JW15 and water extract of BD and BD have ability to induce synergistic immunomodulative effects and are suitable for consideration as a functional food for humans and functional feed additives for companion animals.

        고지방식을 섭취한 C57BL/6J 마우스에서 혼합유산균(Lactobacillus casei WK3, Enterococcus faecium WK5, Bifidobacterium longum WK9, Lactobacillus plantarum K-1)의 콜레스테롤 및 내장지방 저하 효과

        We investigated the serum cholesterol and visceral fat lowering effects of combined lactic acid bacteria (Lactobacillus casei WK3, Enterococcus faecium WK5, Bifidobacterium longum WK9, and Lactobacillus plantarum K-1) in high fat diet-fed mice. To investigate the hypocholesterolemic effects of lactic acid bacteria (LAB), six-week old male C57BL/6J high fat-diet fed mice were orally administered WK3 (2.0×109 CFU/mouse) alone or mixed with WK5, WK9 and K-1 (Combined LAB, 5.0×108 CFU/mouse) for 6 weeks. The normal diet control group and high fat-cholesterol diet control group (HFC) were given PBS instead of LAB. After 6 weeks, pathological and biochemical blood analyses were conducted. Upon blood lipid analysis, the levels of total cholesterol and low density lipoprotein cholesterol were significantly decreased in mice in the Combined LAB group (135.2±19.18 and 50.3±0.97 mg/dL, respectively) compared to the HFC group (154.3±15.60 and 64.1±0.73 mg/dL, respectively). However, treatments had no effects on the serum aspartate aminotransferase and alanine aminotransferase levels. In H&E stained liver sections, HFC mice showed accumulation of lipid droplets, but they were rare in livers of Combined LAB fed mice. The increased epididymal adipose tissue of high fat diet-fed mice was significantly reduced by Combined LAB supplementation. These results suggested that administration of Combined LAB had serum cholesterol and visceral fat lowering effects and could be useful probiotics for cholesterol and visceral fat lowering products. 본 연구에서는 혼합유산균의 기능성 탐색을 위한 연구의 일환으로 고지방식이 유도 마우스에 혼합유산균을 급이시켜 콜레스테롤 및 내장지방 저하 효과를 알아보았다. 실험군은 정상식이를 급이한 정상대조군, 고지방식이를 급이한 실험대조군(HFC), Lactobacillus casei WK3만 단독으로 투여한 단독유산균군(WK3), 4종 유산균을 급이한 혼합유산균 투여군(Combined LAB)으로 나누어 실험을 진행하였다. HFC군의 최종체중과 식이섭취량은 WK3군과 Combined LAB군보다 높았으나 유의적인 차이는 나타나지 않았다. 혈청 내 혈액화학을 분석한 결과 AST, ALT의 유산균 투여로 인한 유의적인 감소는 확인되지 않았으나 혈청 내 총콜레스테롤, LDL-콜레스테롤 함량, 동맥경화지수는 HFC군보다 Combined LAB군이 유의적으로 감소하였다. 간장과 부고환 지방조직의 장기 무게를 측정한 결과 WK3군과 Combined LAB군의 간장 무게가 HFC군보다 유의적으로 감소하였다. 또한 간장 조직을 병리학적으로 관찰한 결과 정상적인 간 조직에 비해 HFC군의 간 조직에서 간세포의 팽윤과 지방변성을 관찰할 수 있었다. 하지만 유산균을 투여한 WK3군과 Combined LAB군에서 간세포의 팽윤 및 지방변성이 현저하게 감소한 것을 확인할 수 있었다. 부고환 지방조직을 관찰한 결과 HFC에 비해 WK3와 Combined LAB군이 지방세포 크기를 유의적으로 감소시키는 것이 확인되었다. 따라서 4종 혼합 유산균은 고지방식이동물에서 혈중 총 콜레스테롤 및 LDL-콜레스테롤 함량을 감소시켜 지질 농도를 개선하는 효과를 보였으며, 또한 부고환 지방조직과 같은 내장지방세포의 비대화를 억제시키는 효과를 갖고 있는 것으로 확인되었다. 이러한 결과는 4종 혼합유산균이 내장지방의 축적과 지방간을 개선시킬 수 있을 것으로 평가되며, 고지혈증질환, 동맥경화 등과 같은 심혈관계 질환의 예방과 개선에 효과적일 것으로 생각된다.

        The immune enhancing effects and characteristics of Bifidobacterium longum and Bifidobacterium breve for the probiotic use in humans and animals

        The purpose of this study was to investigate probiotic characteristics and immune enhancing effects of Bifidobacterium (B.) longum KBB1-26 and BIF-4, B. breve KBB5-22 isolated from human intestine for probiotic use in humans and animals. We measured acid, bile and heat tolerance, antimicrobial activity against pathogenic bacteria, Escherichia (E.) coli, Salmonella (S.) Enteritidis, Staphylococcus (S.) aureus, and Listeria (L.) monocytogenes. Immune enhancing effects of B. longum and B. breve were investigated by measuring nitric oxide (NO), nuclear factor (NF–κb), interleukin-1ß (IL-1ß), interleukin-6 (IL-6), interleukin-12 (IL-12) and tumor necrosis factor- α (TNF- α) in RAW 264.7 cells or RAW BLUE cells. B. longum KBB1-26 was survived at pH 2.0. B. longum KBB1-26 and BIF-4, B. breve KBB5-22 also showed tolerance to 0.3% of oxgall bile salt. B. longum KBB1-26 was able to survive at 70 ℃ and 80 ℃ for 20 min. KBB1-26 showed the antimicrobial inhibition zone to pathogenic bacteria such as E. coli (12 mm), S. Enteritidis (14 mm), S. aureus (14 mm) and L. monocytogenes (41 mm). The production of NO (4.5 ± 0.00μM/mL) and IL-1β (39.7 ± 0.55pg/mL) of KBB1-26 significantly higher than BIF-4 and KBB5-22, respectively. In addition, KBB1-26 and KBB5-22 induce the production of high level of TNF-α and IL-6 in macrophages. Collectively, B. longum KBB1-26 have acid, bile, heat tolerance, antimicrobial activity and immune enhancing effects. These results suggest that KBB1-26 can be used as probiotics for humans and animals.

        Two Pediatric Cases of Successful Management of Postictal Transient Urinary Retention

        We report two cases of postictal urinary retention in pediatric patients with cognitive impairment. Two girls with intellectual disabilities, concomitant cerebral palsy (case 1) and Rett syndrome (case 2), developed urinary retention following seizures. Their caregivers brought them to the hospital with complaints of abdominal distension. After excluding neurological progression, they were referred to the rehabilitation clinic for the evaluation and management of postictal urinary retention. We followed two different approaches in each case to restore normal urination. While serial manual cystometrograms were performed in case 1, clean intermittent catheterization with a voiding diary was performed in case 2 until restoration of normal urination. Based on these pediatric cases of successfully managed postictal urinary retention, we suggest that more attention may be needed for children with cognitive impairment to diagnose and manage postictal urinary retention.

        일반논문 : 한국어 전전이음 /j/의 음절 내 위치

        이 논문은 한국어 전전이음 (on-glide) /j/가 음절 내에서 초성 자음에 위치하는지, 혹은 모음에 속하는지를 간단한 음절 구조 실험을 통해 알아보는 것을 목적으로 한다. 즉, /j/가 두음가설 (onset hypothesis)을 따르는지, 핵음가설(nucleus hypothesis)을 따르는지에 대해 알아보고자 한다. 이를 위해, 미국 위스콘신 지역 한국인 11명을 대상으로 음절 구조 인식 듣기 실험을 진행하였다. 예컨대, /간/(kan)과 /갼/(kjan) 사이에 영어 ‘cat’-‘bat’에서와 같은 라임 (rhyme)이 있는지 없는지를 살펴 라임이 있으면 두음 가설을, 라임이 없으면 핵음가설을 따른다고 보았다. 이 결과, 한국인들이 /j/를 음절 내에서 모음에 속한다고 생각하는 경향이 강하게 나타나, 핵음가설을 지지한다는 것을 알 수 있었다. 이는 한국인들의 음절 구조에 대한 전반적인 인식뿐만 아니라, /j/의 음절 내 위치를 보여주는 하나의 실험 논문으로서 그 의의가 있다.

        Weissella cibaria CMU 경구투여가 비글의 구취 저하에 미치는 효과

        This study assessed the effects of Weissella cibaria (W. cibaria) CMU on oral health in male and female beagles (n = 18) by measuring oral malodor and periodontal disease-related parameters (calculus, plaque, and gingivitis indices). Oral malodor and indicators of periodontal disease were assessed in five treatment groups: negative control (scaling and 0.24 mg of maltodextrin, n = 3), positive control (0.24 mg of maltodextrin, n = 3), and W. cibaria CMU groups (each n = 4) at low (CMU-L, $2{\times}10^7$ colony forming unit [CFU]), medium (CMU-M, $2{\times}10^8CFU$), and high (CMU-H, $2{\times}10^9CFU$) concentrations. After feeding with W. cibaria CMU for 6 weeks, total volatile sulfur compound concentrations in the CMU-L ($2.0{\pm}1.04ng/10mL$), CMU-M ($2.4{\pm}1.05ng/10mL$), and CMU-H ($2.6{\pm}1.33ng/10mL$) groups were significantly lower than in the positive control group ($3.2{\pm}1.65ng/10mL$). Also, CMU-L ($1.4{\pm}0.83ng/10mL$) and CMU-H ($1.9{\pm}1.14ng/10mL$) groups had methyl mercaptan levels lower than that in the positive control group ($2.4{\pm}1.21ng/10mL$) at week 2. The plaque index was significantly lower in the CMU-H group ($4.5{\pm}0.28$) than in the positive control group ($5.9{\pm}1.08$) at week 6. W. cibaria CMU could be useful as a novel oral hygiene probiotics for reducing volatile sulfur compounds production and inhibiting plaque growth in companion animals.

        개 생균제 사용에 적합한 유산균주의 선발 및 면역활성 평가

        This study was conducted to isolate lactic acid bacteria (LAB) from dog intestine and identify potential probiotic strains for canine use. One hundred and one LAB were isolated from feces of 20 healthy dogs. Acid, bile, and heat resistance along with adherence to Caco-2 cells and antimicrobial activity against pathogens were examined. To analyze immunomodulative effects, the production of nitric oxide (NO), TNF-${\alpha}$, and IL-$1{\beta}$ was measured using RAW 264.7 macrophages. Additionally, RAW BLUE cells were used to evaluate nuclear factor-${\kappa}B$ (NF-${\kappa}B$) generation. Ultimately, three strains were selected as canine probiotics and identified as Lactobacillus reuteri L10, Enterococcus faecium S33, and Bifidobacterium longum B3 by 16S rRNA sequence analysis. The L10 and S33 strains showed tolerance to pH 2.5 for 2 h, 1.0% Oxgall for 2 h, and $60^{\circ}C$ for 5 min. These strains also had strong antimicrobial activity against Escherichia coli KCTC 1682, Salmonella Enteritidis KCCM 12021, Staphylococcus aureus KCTC 1621, and Listeria monocytogenes KCTC 3569. All three strains exerted better immunomodulatory effects than Lactobacillus rhamnosus GG (LGG), a well-known commercial immunomodulatory strain, based on NO, NF-${\kappa}B$, IL-$1{\beta}$, and TNF-${\alpha}$ production. These results suggested that the three selected strains could serve as canine probiotics.
