We regenerated a vascular media tissue by seeding smooth muscle cells on the inside of a hybrid vascular scaffold. The hybrid vascular scaffold consisted of two porous biodegradable poly(lactide-co-glycolide) (PLGA) thin layers on both the inner and the outer surfaces of the non-biodegradable expanded polytetrafluoroethylene (ePTFE) layer. The PLGA layers were designed to have porous structures for control of tissue regeneration by impregnating and then gas-foaming of ammonium bicarbonates as porogens of the biodegradable layers. To improve tissue regeneration, we coated gelatin polymers on inside the porous biodegradable PLGA surface. Media tissue regeneration was successfully achieved by replacing the PLGA layers and the tissues were observed by H&E staining and Sircol collagen assay. Tissue formation was dependent on cell culture period. While the two PLGA layers were expected to serve as biological layers of intima, media and adventitia of the blood vessel after their transformation into the engineered tissues, the non-biodegradable ePTFE in the middle of the hybrid scaffold did as a mechanically supporting layer.
MCF10A 인간유방상피세포에서 MKK6의 활성화에 의하여 유도되는 침윤성 및 이동성

Ras의 발현은 침습성과 종양의 재발을 포함한 유방암의 공격성의 지표로 알려져 왔다. 선행연구에서 우리 연구팀은 p38 MAPK 가 인간유방상피세포에서 H-ras와 N-ras에 의하여 다르게 조절되며, H-ras 특이적인 세포 침습성과 이동성의 표현형을 이끌어내는 핵심 신호전달 분자라는 것을 보였다(Cancer Res. 63, 5454-61,2003). 이번 연구에서, 우리는 "gain of function" 연구로서 MCF10 세포에서 전이성 잠재력을 유도하는데 있어서 p38 MAPK 기전의 역할을 좀더 알아보았다. p38 MAPK의 직접적인 상위 활성화 분자인 MAP kinase kinase (MKK)-6을 구조적으로 활성화되도록 발현시킨 MCF10A의 stable transfectants을 만들어 실험하였다 이 세포에 서는 세포의 침습과 이동이 MMP-2의 발현의 특이적으로 증가와 더불어 관찰되었다. 이는 MCP10A 세포에 있어서 전이 잠재성에 대한p38 MAPK 기전의 역할을 증명하는 것이다.

Ras expression has been suggested a marker for tumor aggressiveness of breast cancer, including the degrees of invasion and tumor recurrence. We previously showed that p38 MAPK is a key signaling molecule differentially regulated by H-ras and N-ras, leading to H-ras-specific cell invasive and migrative phenotypes in human breast epithelial cells (Cancer Res. :63, 5454-5461, 2003). In this study, we further investigated the role of p38 MAPK pathway in the induction of metastatic potential in MCF10A cells as a “gain of function” study. We established stable transfectants of MCF10A expressing constitutively activated mutant of MAP kinase kinase (MKK)-6, the direct upstream activator of p38 MAPK. We show the induction of invasion and cell migration with specific upregulation of MMP-2 in these cells, demonstrating the role of p38 MAPK pathway in the metastatic potential in MCF10A cells.[Supported by a grant(R04-2003-000-10063-0) from the Basic Research Program of the Korea Science & Engineering Foundation]

The most impotant parameter in predicting value of osteoporotic fracture is the level of BMD(bone mineral density). The formation of peak bone mass is important in the preventive aspect of osteoporotic fracture. Among the factors influencing the formation of peak bone mass, the hereditary influence is believed to be the most important. We studied the importance of hereditary influence in determing peak bone mass by comparing BMDs of mothers and their daughters in college, who are in the age of achieving peak bone mass. The BMDs of daughters were greater than those of mothers, and mean age of menarche in daughters was earlier than that of mothers. Inmothers, BMD of L2-L4 had significant positive correlatins with height, weight, and BMI, however femur neck had positive correlations with weight and BMI. In daughters, BMD of L2-L4 had significant correlatin with weight, total calorie intake and protein intake, but MBD of femur neck had correlation with weight, body fat and animal calcium intake. Daughters of osteopenic mothers might be advised to evaluate the bone mineral density at critical period for the formation of peak bone mass, to identify the risk factors and to encourage more calcium intake. In conclusion, the BMDs of daughters have strong positive correlation with those of mothers. Hereditary influence on peak bone mass can be strongly suggested, but common environmental factors cannot be excluded. (J Kor Endocrinol 8:66~71, 1993)
