Abstract-In order to observe the relationship between the protein carboxylmethylation and pancreatic secretion, we examined the effect of secretagogues on the activities of protein methylase II[S-adenosyl-L-methionine:protein carboxyl O-methyl transferase, EC 2.1.1.24] and SAM synthetase [ATP:L-methonone S-adenosyltransferase EC 2.5.1.6] in pancreatic fragment of chickens. In addition, S-adenosylmethionine concentration was measured by HPLC in chicken pancreatic tissues to test the relationship between SAM-synthetase and protein methylation. The results were as follows 1) The time course changes of amylase activity in the absence and presence of CCK-PZ(4 ng) was rising steadily throughout 60 min. incubation period. In the presence of both CCK-PZ(4 ng) and secretin(2CU), the release of amylase was increased when compared with unstimulated control. 2) The CCK-PZ(4 ng) and secretin(2CU) did not affect the activity of SAM synthetase. But sod. nitroprusside, at a concentration that inhibits the stimulation of amylase release, decreased the activity of SAM synthetase. 3) The activity of protein methylase II was not changed in secretagogues-stimulated group. 4) SAM concentration in chicken pancreatic tissue was not changed in secretagogues-stimulated group.
