        5 ' 5 ' - dithiobis ( 2 - nitrobenzoic acid ) 에 의한 메탈로사이오닌 cystein 기의 분자 특징

        In the previous study of the reaction of the cysteinyl groups of metallothionein (MT) with 5,5`-dithiobis(2-nitrobenzoic acid) (DTNB) (Korean Biochem. J. (1984) 17, 288-298), it was found that the DTNB titrable cysteinyl groups were markedly affected either by the presence of metal or by the changes of protein conformation. In order to clarify these phenomena, we prepared the a fragment (4-metal cluster) of MT-I and examined the DTNB reaction. The ratio of 1.7 was obtained as the DTNB titrable cysteinyl groups to the bound metal ions. Thus these data support the notion that only one type of cysteinyl groups may interact with DTNB, mainly simply coordinated cysteinyl groups. Furthermore, we investigated the effects of var ous disulfides on the DTNB reaction rates to explore the micro environment around the cysteinyl groups. Disulfides tested were cystamine, 2-hydroxyethyl disulfide, dimethyl disulfide, and 3,3`-dithiodipropanoic acid. The DTNB reaction rate of MT-I was enhanced by cystamine and 2-hydroxyethyl disulfide, but that of the α fragment was inhibited by both disulfides. Others had no effects on the DTNB reaction of MT. Therefore, the DTNB titrable cysteinyl groups of MT seem to be located in a hydrophilic and negatively charged microenvironment.

J. (1984) 17, 288-289), DTNB에 의하여 적정되는 cysteinyl기의 수는 금속의 존재나 메탈로사이오닌의 구조에 영향을 받는다. 이것을 더 연구하기 위해 메탈로사이오닌의 $\alpha$ fragment를 분리하여 DTNB와 반응시켰다. DTNB로 적정가능한 cysteinyl기의 수와 금속이온수의 비는 1.7이었다. 이것은 메탈로사이오닌 내의 cysteinyl키 중 simply coordinate된 것만이 DTNB와 반응함을 뒷받침 해준다. 더 나아가 cysteinyl 거의 분자내 환경을 알기 위해 disulfide 의 영향을 살펴보았다. Cystamine, 2,2'-dihydroxyethyl disulfide, 3,3'-dithiodipropanoic acid, dimethyl disulfide 동의 disulfide가 사용 되었다. 메탈로사이오닌-I의 경우에는 cystamine과 2,2'-hydroxyethyl disulfide에 의하여 반응속도가 증가하였으나 $\alpha$ fragment의 경우에는 감소하였다. 다른 두 disulfide는 반응속도에 영향을 마치지 않았다. 따라서 DTNB와 반응하는 메탈로사이오닌의 cysteinyl기는 그 미세환경이 음성전하를 띠고 친수성의 위치에 처해 있다고 추정된다.

        샵 창업 시 중요사항에 따른 피부관리샵 운영 특성 및 직업만족도

        본 연구는 샵 창업 시 중요사항에 따른 피부관리샵 운영 특성 및 직업만족도에 대한 차이를 살펴보고, 보다 나은 소규모 피부관리샵 운영을 위한 기초자료를 제공하고자 하였다. 연구대상은 현재 전국 각 지역에서 소규모 피부관리샵을 운영중인 원장들을 대상으로 2016년 1월~ 3월의 기간을 두고 설문조사를 실시하였다. 연구결과, 샵 창업 시 교통수단 및 주차편리성을 중요사항으로 생각한 운영자들이 오랜 근무년수, 샵 운영기간, 높은 월 고정수입, 최고수익의 비율이 각각 가장 높았다. 직업만족도(F=3.223, p<.05)에서는 창업 시 중요사항으로 유동인구를 중요하게 생각하는 운영자가 가장 높았고 (M=4.11), 투자비용을 중요하게 생각하는 운영자의 직업만족도가 가장 낮았다(M=3.41). 세부적으로 직업 재선택 의향(F=2.960, p<.05)에서는 교통수단 및 주차편리성(M=3.95)을 삽 창업 시 중요하게 생각하는 운영자가 가장 높았다. 따라서 샵 창업 시 중요사항에 따른 안정적인 운영을 위해서는 유동인구가 많고, 교통수단 및 주차 편리성이 높은 위치파악과 선정으로 고객의 접근성과 직업만족도를 높이는 것이 필요할 것이다. 또한 후속 연구에서는 운영특성에 따른 마켓팅 전략에 관한 지속적인 노력과 연구가 수반되어야 할 것으로 사료된다. The purpose of this study was to investigate the differences in the characteristics of the aesthetic salon operation and job satisfaction according to important considerations in opening aesthetic salons as well as to provide basic data for improving the operation of the salons. Toward this aim, a questionnaire survey was conducted for owners of small aesthetic salons across South Korea from January- March, 2016. Results were as follows: Among those who identified means of transportation and parking convenience as key considerations in launching aesthetic salon businesses, length of service, length of business operation, monthly fixed revenue and proportion of the highest profit were highest. Job satisfaction (F=3.223, p<.05) was highest (M=4.11) among those that responded `floating population` as a primary component in launching aesthetic salon businesses, and lowest among owners who mentioned `cost of investment` (M=3.41). Specifically, job mobility (F=2.960, p<.05) was the highest among respondents who selected means of transportation and parking convenience (M=3.95) as key criteria in launching aesthetic salon business. For the stable operation of an aesthetic salon, a location with high floating population, good transportation, parking convenience, high levels of customer access, and job satisfaction are critical factors. In addition, future studies should examine marketing strategies by operating characteristics.

        Cadastral non-coincidence question has been regarded as the biggest issue in Korean cadastral system. In retrospect, cadastral or land registration has been a byproduct of Japanese imperialistic invasion, facilitating tax collection and resource exploitation. Therefore, over several decades, cadastral non-coincidence lots have been widespread in Korean society. This study analyzes cadastral non-coincidence issues from the social, economic, and administrative dimensions. In addition, it suggests couples of policy alternatives including enactment of special law, supply of liquidation refund, and national cadastral resurvey.

        유동층 생물반응기를 이용한 우유폐수처리에 관한 연구

        This study was carried to investigate the milk-wastewater treatment in batch and continuous fluidized-bed bioreactor containing active microorganism, Aspergillus niger KCTC 1231. The reactor was operated at 30℃. The COD removal efficiency and MLSS were measured to determine the performance of the fluidized-bed bioreactor by cell concentration, aeration rate, and organic loading rate, respectively. At conditions of 60 ㎖ cell concentration and 2.0LPM aeration rate, milk-wastewater showed the COD removal efficiency of 81.51% and MLSS of 43,562 mg/ℓ, Also the same conditions, at adding 80 ㎖ chitosan beads, the COD removal efficiency and MLSS were about 85.79% and 62,482.5 mg/ℓ, respectively. In case of organic loading rate 3.88 kgCOD/㎥·day, free cell and adding chitosan beads showed about 80∼85% and 85∼90% COD removal efficiency, respectively. It was showed that milk-wastewater treatment of fluidized-bed bioreactor with chitosan beads was about 5% more efficient.

        남부지역 시설원예산물의 가공품개발 연구 : 오이지의 품질변화에 미치는 키토산의 효과

        This study was investigated the characteristics of chitosans(red crab, fungal) prepared from red crab shell and Absidia coerulea IFO 5301 and the effects of chitosans on the quality of cucumber pickles during fermentation. The molecular weight of red crab and fungal chitosan were 6.5 × 10⁵ and 2.75×10⁵ Daltons, respectively. The degree of deacetylation, 76% and 88%, respectively. From the I.R. spectra, the absorption bands at 1655 cm⁻¹(arnide I), 1550 cm⁻¹(arnide II) of red crab and reference chitosan became very weak, similarly. And at solid state ¹³C-NMR spectra, C=O(carbonyl carbon) signals were absent, whereas CH₃(methyl carbon) signals were residues. Chemical shift of ¹³C-NMR spectra of red crab, fungal and reference chitosans were in good agreement with slight experimental deviation. The cucumber pickles were fermented at 25°C for 7days in 10% brine. The rate of change in pH and total acidity of cucumber pickle liquids with 10% brine(control) was rapidly while with 20 mg chitosan was slowly. The turbidity of cucumber pickle liquid with 20 mg chitosan was lower than that of the control. These results showed applicability of greenhouse cucumber as a cucumber pickle and qualities of cucumber pickles by added chitosans were improved.
