Because many materials for skin application can induce the skin irritation, we should evaluate the primary skin irritation test. Recently, several alternative in vitro methods to Draize skin irritation test using rabbits have been developed. The most promising alternative method for skin irritation is the using of reconstituted human skin equivalent model(RhE). In this study, we introduce the development status of RhE model, principle and procedure of test method, and validation method. Further studies are in progress to improve the test protocols and the validation studies for KeraSkinTM developed by Korea in order to be certified internationally.
Safety Evaluation and Anti-wrinkle Effects of Retinoids on Skin

Retinoids have many beneficial effects on dermatological applications. But, retinoids cause skin irritation. In this study, the safety of retinoids was clarified via both primary skin irritation test in rabbits and sensitization study using an integrated model for the differentiation of chemical-induced allergic and irritant skin reaction (IMDS), an alternative method to sensitization test. The effects of retinoids on the change of ultraviolet A (UVA)-induced matrix metalloproteinase-1 (MMP-1) in human skin fibroblasts and the modulation of type-1 pN collagen synthesis in hairless mice were examined to clarify the anti-wrinkle effects. Alltrans retinol (t-ROL) and its derivative, all-trans retinoic acid (t-RA), showed mild skin irritation but did not induce the sensitization. t-ROL and t-RA exerted anti-wrinkle effects by inhibiting the UVA-induced MMP-1 in human skin fibroblasts and increasing the type-1 pN collagen synthesis in hairless mice. These findings suggest that retinoids do not induce the allergy, and show anti-wrinkle effects by decreasing MMP-1 activation and increasing collagen synthesis.
항산화력이 우수한 Fructose 1,6-diphosphate(FDP)를 피부적용제로 응용하기 위한 안전성 평가

Fructose 1,6-diphosphate (FDP), a glycolytic metabolite, is reported to ameliorate inflammation and inhibit the nitric oxide production in murine macrophages stimulated with endotoxin. It is also reported that FDP has cytoprotective effects against hypoxia of ischemia/reperfusion injury in brain and heart, and may play a protective role in ultraviolet B (UVB, 280~320nm)-injured keratinocyte by attenuating prostaglandin (PG)-E₂ production and cyclooxygenase (COX)-2 expression, which are possibly through blocking the intracellular reactive oxygen species (ROS) accumulation. Therefore FDP is considered to act as a potent antioxidant especially in the skin. We conducted the several safety tests (single-does toxicity, primary skin irritation test, eye irritation test, skin sensitization test, phototoxicity test, photosensitization test and human patch test) to see if FDP is safe incase used for the skin application, Our data obtained hitherto suggest that FDP is very safe if applied to the skin.

The concentrations of hydroxide, carbonate and oxalate for Y(III), Ba(II) and Cu(II) ions in an aqueous solution have been theoretically calcuated with respect to pH and their solubility diagram could be obtained. The optimum pH ofr oxalate coprecipitates at room temperature was estimated as<4, which was not influenced by carbonates and hydroxides in H2O solvent. The yield is dependent on the concentration of added oxalic acid, but the concentration of oxalic acid was fixed as 0.1M in this calculation for simplicity.
미성숙 감(Diospyros kaki Thunb. Sangju-Dungsi) 에탄올 추출물의 항알레르기 생리활성 연구

This study was conducted to verify the availability of immature persimmon ethanol extract (IPEE) as a natural and functional ingredient in protecting inflammation and allergy of skin based on the mechanism. The major content analysis, antioxidant activities, anti-allergic activity, anti-inflammatory effect, and safety related to irritation of IPEE were evaluated. The gallic acid content per 10 mg/mL of IPEE was 0.522% (5.22 mg/g). The total polyphenol and flavonoid contents were 428.3 mg/g and 31.1 mg/g, respectively. In ABTS+ activity, DPPH ability and SOD-like activity, it showed a concentration-dependent increase, which indicated IPEE has excellent antioxidant activities. As for the anti-allergy test in RBL-2H3 cells, the IPEE showed a decrease in β-hexosaminidase secretion as the concentration increases, and IPEE tended to decrease IL-4 secretion in all RBL-2H3 cells compared to the IgE + HSA group. IPEE showed good anti-inflammatory effect in RAW 264.7 cells by decrease of NO production and inflammation cytokines (TNF-α and IL-6). Also IPEE showed non-irritant in BCOP assay. By the results of this study, the IPEE containing high tannins, had good antioxidant, anti-allergic, and anti-inflammatory effects, which indicated that the immature persimmon is considered to be a useful for the development of related functional ingredients.
Antitumor effect of a newly synthesized celecoxib derivative encapsulated in liposome

A new cyclooxygenase-2 inhibitor (code: PCX-3)was synthesized as a sodium salt form of celecoxib, a nonsteroidal anti-inflammatory drug (NSAID), and tested for its anticancer activity using human colon adenocarcinoma cells (HT-29) in vitro. Anti-proliferative effect of HT-29cells by PCX-3 in DPPC/Chol liposomes was more effective than the free PCX-3 by 2-folds (IC30 = 125 lM vs. 227.5 lM). The same liposomal formulation of PCX-3 also showed a 2-fold increased effect than the free one both in DNA fragmentation and caspase activity of HT-29 cells at 19-743 lM and 37–371 lM ranges, respectively, suggesting apoptosis-based anti-proliferative effect. Down regulation of prostaglandin E2 level of HT-29 cells by the treatment of liposomal PCX-3 was more profound than its free form at 0.001–0.002 lM range. These data suggest that the liposomal formulation of this newly synthesized PCX-3 could be re-visited as a new anticancer or chemopreventive agent in the future.
