        Marein Prevented LPS-Induced Osteoclastogenesis by Regulating the NF-κB Pathway In Vitro

        Many bone diseases such as osteolysis, osteomyelitis, and septic arthritis are caused by gramnegative bacterial infection, and lipopolysaccharide (LPS), a bacterial product, plays an essential role in this process. Drugs that inhibit LPS-induced osteoclastogenesis are urgently needed to prevent bone destruction in infective bone diseases. Marein, a major bioactive compound of Coreopsis tinctoria, possesses anti-oxidative, anti-inflammatory, anti-hypertensive, antihyperlipidemic, and anti-diabetic effects. In this study, we measured the effect of marein on RAW264.7 cells by CCK-8 assay and used TRAP staining to determine osteoclastogenesis. The levels of osteoclast-related genes and NF-κB-related proteins were then analyzed by western blot, and the levels of pro-inflammatory cytokines were quantified by ELISA. Our results showed that marein inhibited LPS-induced osteoclast formation by osteoclast precursor RAW264.7 cells. The effect of marein was related to its inhibitory function on expressions of pro-inflammatory cytokines and osteoclast-related genes containing RANK, TRAF6, MMP-9, CK, and CAII. Additionally, marein leads to markedly inhibited NF-κB signaling pathway activation in LPS-induced RAW264.7 cells. Concurrently, when the NF-κB signaling pathway was inhibited, osteoclast formation and proinflammatory cytokine expression were decreased. Collectively, marein could inhibit LPS-induced osteoclast formation in RAW264.7 cells via regulating the NF-κB signaling pathway. Our data demonstrate that marein might be a potential drug for bacteria-induced bone destruction disease. Our findings provide new insights into LPS-induced bone disease.

        IMC-based Design for Teleoperation Systems with Time Delays

        The presence of time delays in communication introduces a limitation to the stability of bilateral teleoperation systems. In this study, we address the design of Internal Model Control (IMC)-based controllers for teleoperation systems with arbitrary time delays. The two degrees of freedom IMC structure is utilized for the master and the slave respectively and thus the passivity-assumption on the external forces is not required. The stability of the overall system is analyzed by regarding the whole system as a multivariable system. A performance-oriented controller design process which guarantees the delayed tracking between the master and the slave is also given. Finally, an example is given to validate the effectiveness of the proposed method.

      • Scutellaria Extract Decreases the Proportion of Side Population Cells in a Myeloma Cell Line by Down-regulating the Expression of ABCG2 Protein

        Background and Aims: Scutellaria is one of the most popular traditional Chinese herbal remedies against various human diseases, including cancer. In this study, we examined the active effects of Scutellaria extract and its main flavonoid constituents on the proportion of side population cells within human multiple myeloma cell line RPMI8226 in vitro and explored the potential molecular mechanisms involved. Materials and Methods: The contents of flavonoids in ethanolic extract of Scutellaria baicalensis Georgi were determined using high performance liquid chromatography. The antiproliferative effect of the ethanolic extract on RPMI-8226 was determined by CCK assay. Apoptosis was measured by annexin combining with propidium iodide in a flow cytometer. Cell cycle analysis was performed by propidium iodide staining in combination with flow cytometry analysis. Hoechst 33342 exclusion assay was used for the identification of side population within RPMI8226 cells. The expression of ABCG2 protein was assessed by Western blotting assay. Results: The content of major flavonoids constitutents of Scutellaria extract was baicalin (10.2%), wogonoside (2.50%), baicalein (2.29%), and wogonin (0.99%), respectively. The crude Scutellaria extract did not show significant anti-proliferative effect, apoptosis induction and cell cycle arrest in RPMI-8226 within the concentrations of $1-75{\mu}g/mL$. However, the ethanolic extract, baicalein, wogonin and baicalin reduced the side population cells in RPMI-8226, and data showed that baicalein and wogonin had stronger inhibitory effects. Correspondingly, they also exhibited significant effects on decreasing the expression level of ABCG2 protein in RPMI-8226 in vitro. Conclusions: Our results for the first time demonstrated a novel mechanism of action for Scutellaria extract and its main active flavonoids, namely targeting SP cells by modulating the expression of ABCG2 protein. This study provides an insight for new therapeutic strategies targeting cancer stem cells of multiple myeloma.

        Ground type-I collagen—a focused study on its fibrillogenesis behavior and bioactivity in vitro

        Take bovine tendon type-I collagen as the raw material, we pretreated it by freezing and then grinding the macromolecule at a low temperature (4–10 °C). Effects of the grinding process on the molecular structure, water solubility, fibrillogenesis behavior in vitro and biological activity of collagen were systematically prospected. Sodium dodecyl sulfate–polyacrylamide gel electrophoresis (SDS–PAGE) results revealed that the collagen had typical structural characteristics of type-I collagen; moreover, grinding treatment gave rise to partially degraded collagen molecules which took on continuous bands with lower molecular weights different from α1 and α2. More strikingly, the degradation degree positively corresponded to the grinding duration. Circular dichroism (CD) analysis manifested that with extended grinding, intensities of both the positive and negative peaks for collagen samples shrunk. Interestingly, grinding also contributed to improved water solubility of collagen. The water solubility of the sample ground for 20 min (COL20) was 1.91 mg/mL, which is about 6 times higher than that of the natural collagen sample (0.28 mg/mL). Amid the study concerning the fibrillogenesis behavior, turbidity and rheological data collaboratively demonstrated that with prolonged grinding time, the assembly rate and degree of the sample plunged. Assembled gel from the ground collagen with finer fibrils was also observed in scanning electron microscope (SEM), which further embraced the conclusion aforementioned in fibrillogenesis kinetics. Cell experiments corroborated that the grinding process can be harnessed to manipulate the cell proliferation promoting ability of collagen. Simultaneously, further dissection of COL20 with the best biological activity clarified that the water-soluble part and water-insoluble part of COL20 had distinct aptitudes as to promoting cell proliferation as well as inhibiting the growth of liver cancer cells.

        Creep Properties of a Nickel-Based Single Crystal Superalloy with Low Density

        A novel nickel-based single crystal superalloy with low density had been designed and the creep deformation mechanismsof the experiment alloy under two conditions (1050 °C/200 MPa and 1100 °C/137 MPa) had been systematically discussed. The topological inversion had been observed after fracture under the above two conditions. The effects of temperature andstress on creep life of the experiment alloy had been found no significant difference under the two conditions. It was indicatedthat the small decrease in temperature exerted limited influence on the creep behaviors of single crystal superalloys in therange of high temperature.

      • Unequal Error Protection in Image Transmission Based on LDPC Codes

        This paper investigates the performance of image transmission with unequal error protection (UEP) schemes based on irregular Low-Density Parity-Check (LDPC) codes. Firstly, the UEP is achieved by mapping different bits of the image bytes to different positions of LDPC codes, i.e. the more important bits in the image are mapped to the variable node with higher degrees in irregular LDPC codes, after the channel coding, we map the information bits into a power efficient QAM constellation and map the parity check bits into a spectral efficiency 16QAM constellation. Simulation results show that the UEP scheme is effective.

        Study on the Thermal Transient Response of TSV Considering the Effect of Electronic-Thermal Coupling

        The transmission performance of TSV considering the effect of electronic-thermal coupling is an new challenge in three dimension integrated circuit. This paper presents the thermal equivalent circuit (TEC) model of the TSV, and discussed the thermal equivalent parameters for TSV. Si layer is equivalent to transmission line according to its thermal characteristic. Thermal transient response (TTR) of TSV considering electronic-thermal coupling effects are proposed, iteration flow electronic-thermal coupling for TSV is analyzed. Furthermore, the influences of TTR are investigated with the non-coupling and considering coupling for TSV. Finally, the relationship among temperature, thickness of $SiO_2$, radius of via and frequency of excitation source are addressed, which are verified by the simulation.

        The effect of chlorfenapyr exposure on Solenopsis invicta Buren (Hymenoptera: Formicidae)

        Red imported fire ant (RIFA), Solenopsis invicta Buren, pose a serious risk to the agriculture, environment, and public health in areas invaded by this species. Chemical insecticides have been widely used to prevent and control this plaguing pest. To discover a novel and cost-effective compound for controlling fire ants, we examined the toxic effect of chlorfenapyr on fire ants. While chlorfenapyr exhibited an insufficient horizontal toxicity transfer and knockdown effect to fire ants, feeding with chlorfenapyr solution increased fire ant workers’ gathering abilities while decreasing their walking, climbing, and adhesion ability. The mortalities of the small, medium, and large-size workers were 84.17%, 98.75%, and 82.08%, respectively, after 72 h of treatment with 25, 50, and 200 μg/ml chlorfenapyr. Additionally, females and males exhibited more higher tolerance than workers to chlorfenapyr; meanwhile, females showed a higher resistance than males. That is, the mortality of males was 100% after 120 h of treatment with 6.25 μg/ml chlorfenapyr. In contrast, the mortality of females was 79.05% after 144 h of treatment with 100 μg/ml chlorfenapyr. In addition, the chlorfenapyr bait exhibited significant toxicity on fire ants. Treated with 0.01% chlorfenapyr bait showed a better toxic effect than 0.025% during the period of 2 d-7 d. However, the weight of ant corpses was 1.32 and 1.23 g at 12 d of treatment with 0.025% and 0.01% chlorfenapyr bait, respectively. The present research showed that chlorfenapyr appeared to be an effectively potential toxic compound for developing bait to manage S. invicta.

        Rebalancing SMAD7/SMAD3 Signaling Reduces Adhesion Formation during Flexor Tendon Healing

        Transforming growth factor-β is a key factor in regulating adhesion formation during tendon healing. We investigated the effectiveness of SMAD family members, SMAD7 and SMAD3, in the TGF-β/Smad signaling during flexor tendon repair. Mouse flexor toe deep tendon rupture anastomosis models were made. On days 3, 7, 14, 21, and 28, the expressions of smad7 and smad3 in flexor tendon tissues were detected by RT-qPCR and western blot. Furthermore, postoperative intraperitoneal injections of SMAD7 agonists or SMAD3 antagonists were given. The degree of tendon healing was evaluated by adhesion testing and biomechanical experiments. Hematoxylin and eosin (HE) staining was used to observe the pathological changes. Immunohistochemistry was used to evaluate the expressions of collagen III, SMAD3, and SMAD7. The mRNA levels of matrix metalloproteinases, Mmp2 and Mmp9, and scleraxis (SCX) in flexor tendon tissue were detected by RT-qPCR. Smad3 expression increased and Smad7 expression decreased in flexor tendon tissue after injury. In addition, the SMAD7 agonist blocked SMAD3 phosphorylation. SMAD7 agonist and SMAD3 antagonist both improved adhesion formation during flexor tendon healing, and decreased the expressions of collagen III, Mmp9, and SCX, while increasing Mmp2 expression. This study provides a possible theoretical basis for the SMAD7-SMAD3 signal cascade during flexor tendon adhesion healing.
