<P>53BP1 (p53-binding protein 1) is a conserved nuclear protein that is phosphorylated in response to DNA damage and rapidly recruited to the site of DNA double strand breaks, demonstrating its role in the early events to DNA damage and repair of damaged DNA. In this study, we used the yeast two-hybrid system to identify proteins that interact with 53BP1. Identification and characterization of 53BP1 protein interactions may help to further elucidate the function and regulation of 53BP1. We identified protein phosphatase 5 (PP5), a serine/threonine phosphatase that has been implicated in multiple cellular function, as a 53BP1-binding protein. This interaction further confirmed that 53BP1 interacts with PP5 in PP5-overexpressing U2OS cells, after radiomimetic agent neocarzinostatin (NCS) treatment. 53BP1 dephosphorylation at Ser-25 and Ser-1778 was accelerated in PP5-overexpressing U2OS cells following NCS treatment, and its dephosphorylation was correlated with reduced phospho-53BP1 foci formation. In contrast, the overexpression of PP5 had no effect on NCS-activated BRCA1-Ser-1524 phosphorylation. Additionally, PP5 down-regulation inhibited the dephosphorylation of 53BP1 on Ser-1778 and the disappearance of phospho-53BP1 foci following NCS treatment. Moreover, non-homologous end-joining activity was reduced in PP5-overexpressing U2OS cells. These findings indicate that PP5 plays an important role in the regulation of 53BP1 phosphorylation and activity in vivo.</P>
Ape1/Ref-1 Stimulates GDNF/GFRՁ1-mediated Downstream Signaling and Neuroblastoma Proliferation

We previously reported that glial cell line-derived neurotropic factor (GDNF) receptor Ձ1 (GFRՁ1) is a direct target of apurinic/apyrimidinic endonuclease 1 (Ape1/Ref-1). In the present study, we further analyzed the physiological roles of Ape1/Ref-1-induced GFRՁ1 expression in Neuro2a mouse neuroblastoma cells. Ape1/Ref-1 expression caused the clustering of GFRՁ1 immunoreactivity in lipid rafts in response to GDNF. We also found that Ret, a downstream target of GFRՁ1, was functionally activated by GDNF in Ape1/Ref-1-expressing cells. Moreover, GDNF promoted the proliferation of Ape1/Ref-1-expressing Neuro2a cells. Furthermore, GFRՁ1-specific RNA experiments demonstrated that the downregulation of GFRՁ1 by siRNA in Ape1/Ref-1-expressing cells impaired the ability of GDNF to phosphorylate Akt and PLCՃ-1 and to stimulate cellular proliferation. These results show an association between Ape1/Ref-1 and GDNF/GFRՁ signaling, and suggest a potential molecular mechanism for the involvement of Ape1/Ref-1 in neuronal proliferation.
SETDB1 genomic DNA 를 표적하는 TALEN construct 제작 및 분석

TALEN은 특정유전자를 표적 하여 knock-out 시킬 수 있는 새로운 개념의 유전자 클로닝 방법이다. TALEN 플라스미드에는 DNA binding 도메인과 Fok1 절단효소 기능이 융합되어 있기 때문에, genomic DNA 의 어느 부위라도 결합할 수 있고, 표적 염기서열을 절단하여 유전자 돌연변이를 유도할 수 있다. 본 연구에서 우리는 SETDB1 HMTase 유전자의 단백질 개시코돈과 프로모터 -25 upstream 부위를 표적 하는 두 종의 TALEN constructs 를 제작하였다. 이를 위하여 두 단계의 클로닝이 진행되었다. 첫 번째는 모듈벡터에서 pFUS배열벡터로 표적서열을 옮겨 콜로니 PCR을 통해 smear밴드와 Esp1 제한 효소를 이용하여 약 1 kb의 insert가 들어 있음을 확인하였다. 두 번째는 배열 벡터로부터 TALEN 발현벡터로 옮기는 과정을 진행하였으며, 염기서열분석을 통해 확인하였다. 그 결과 최초의 고안된 모듈벡터 서열들이 약 100 bp 간격으로 배열되어 있음을 확인하였다. 제작된 TALEN-DBEX2 construct는 transfection을 통해 SETDB1의 발현이 사라지는 것을 확인하였고, T7E1 분석을 통하여 표적부위에서 돌연변이가 발생하였음을 추정할 수 있었다. 한편, TALEN-DBPR25 transfection을 통하여서도 SETDB1의 발현이 감소하는 현상을 확인 하였다. DBEX2, DBPR25를 이입시킨 HeLa 세포에서 세포 형태가 길어지는 현상을 관찰할 수 있었다. 그러므로 단백질 개시코돈 또는 -25 upstream을 표적 하는 TALEN knock-out 방법은 SETDB1 유전자의 기능연구에 매우 유용하다고 사료된다. TALEN is a newly developed gene engineering method to knock out specific genes. It contains a DNA binding domain and a Fok1 nuclease domain in the TALEN plasmid. Therefore, the engineered TALEN construct can bind to any region of genomic DNA and cut the target nucleotide, thereby inducing mutation. In this study, we constructed two TALEN constructs targeted to a protein initiation codon (DBEX2) or the 25th upstream region (DBPR25) to enable mRNA synthesis of SETDB1 HMTase. We performed the TALEN cloning in two steps. The first step was from module vectors to pFUS array vectors. We confirmed successful cloning with a colony PCR experiment and Esp31 restriction enzyme digestion, which resulted in a smear band and a 1 Kb insert band, respectively The second step of the cloning was from a pFUS array vector to a mammalian TALEN expression vector. The engineered TALEN construct was sequenced with specific primers in an expression vector. As expected, a specific array from the module vectors was shown in the sequencing analysis. The specific module sequences were regularly arrayed in every 100 bp, and SETDB1 expression totally disappeared in the TALEN-DBEX2 transfection. PCR amplification targeting of DBEX2 was performed, and the PCR product was digested with a T7E1 restriction enzyme. The expression of SETDB1 was down-regulated in the TALEN-DBPR25 transfection. Morphological changes were also observed in the two TALEN constructs with transfected HeLa cells. These results suggest that the engineered TALEN constructs in two strategic approaches are very useful to knock-out of the SETDB1 gene and to study gene function.

산업혁명 이후 대기 중의 온실가스 중 하나인 이산화탄소가 증가하면서 전세계적으로 지구온난화가 화두되고 있다. 이산화탄소 저감기술인 CCS(Carbon Capture and Storage) 기술은 이산화탄소를 포집 후 저장하는 기술로 저장의 한계에 따라 CCU(Carbon Capture and Utilization) 기술이 각광받고 있다. CCU는 이산화탄소를 이온화 하여 무기 혹은 유기재료로 전환이 가능하다. 폐기물의 금속이온을 이용하여 화학적으로 무기탄산화가 가능하며, 폐기물과 이산화탄소를 동시에 처리할 수 있다는 이점이 있다. 본 연구에서는 최종산물을 탄산칼슘을 생성하는 것이며, 탄산칼슘의 수율을 증대시키기 위해서는 폐기물 내의 칼슘 이온의 추출효율을 올리는 것이 중요하다. 하지만 용매를 이용할 경우 칼슘이온 외에 다른 이온들도 같이 추출되면서 탄산칼슘의 순도를 떨어뜨린다. 이에 최종생성물의 순도를 높이기 위하여 칼슘이온만 선택적으로 추출하기 위해 실험을 진행하였다. 실험에 사용된 폐기물은 탈황석고와 폐시멘트를 이용하였으며, 칼슘이온을 선택적으로 추출한 샘플을 이용하여 탄산화반응을 통하여 탄산칼슘을 생성하였다. 생성된 탄산칼슘은 XRD (X-ray diffraction analyzer(Ultima Ⅳ))와 FE-SEM(Field emission scanning electron microscope, JEOL-7800)를 통하여 결정구조를 분석하였다.
암모니아 화합물을 이용한 폐기물 처리 및 이산화탄소 저감의 다양한 방법 소개

온실가스인 이산화탄소는 다른 온실가스에 비해 Global Warming Potential(GWP)가 가장 낮지만 배출량이 전체 온실가스 중 88 %의 비중을 차지하고 있다. 많은 국가에서 기후변화에 관심을 가지고 이산화탄소 저감에 대한 연구개발이 활발히 일어나고 있다. 본 연구에서는 암모늄 화합물을 이용하여 이산화탄소를 포집하고 산업폐기물의 금속이온을 이용하여 무기재료인 탄산칼슘을 생성하는 다양한 방법을 소개한다. 탄산칼슘 생성을 위해 칼슘이온이 포함된 탈황석고, 폐시멘트를 이용하였다. 결과에서 이산화탄소 포집 성능 및 최종생성물의 결정구조를 확인하였으며, 이산화탄소 loading 값 는 약 2.0의 값을 가진다. X-Ray Diffraction, Scanning Elec-tron Microscope의 분석을 통하여 탄산칼슘이 생성되었음을 확인하였으며, 결정구조는 Vaterite가 생성됨을 확인할 수 있다. 효과적인 공정을 위하여, 생성물을 생성한 후 용액을 회수하여 재이용할 수 있어 연속적인 공정이 가능하다. 회수된 용액의 재이용의 가능성을 보기위하여 이산화탄소를 재흡수 시키면서 같은 공정을 2 cycle씩 진행하여, 연속적인 공정의 잠재성을 확인하였다.
