        Identification of MicroRNAs as potential biomarkers for detecting ischemic stroke

        Background: Increasing epidemic of ischemic stroke (IS) makes it urgent to understand the pathogenesis and regulatory mechanism, previous studies have described microRNAs (miRNAs) is part of the brain's response to ischemia. Objective: The aim of this study was to screen potential biomarkers for the prediction and novel treatment of IS. Methods: Differentially expressed miRNAs were screened from three newly diagnosed IS patients and three controls by RNA sequencing technology. Furthermore, target prediction databases were then used to analysis the target genes of different expressed miRNAs, and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway database were used to identify the functions and the main biochemical and signal pathways of differentially expressed target genes. Results: Our results revealed that 27 miRNAs were differentially expressed in IS, among which, hsa-miR-659-5p was the most highly increased and was first found to be associated with IS. In addition, KEGG pathway analyses showed that differentially expressed miRNAs were mainly significantly enriched in lysosome pathway, cytokine-cytokine receptor interaction pathway, spliceosome pathway, base excision repair pathway. Conclusions: miRNAs were involved in IS pathogenesis, and hsa-miR-659-5p, hsa-miR-151a-3p and hsa-miR-29c-5p as the three highest |log2FoldChange| regulation in this study, which may be the biomarkers of IS and need further study.

        Simulated altitude exercise training damages small intestinal mucosa barrier in the rats

        <P>This study investigated the effect of simulated altitude training on the changes of small intestinal mucosa barrier, bacterial overgrowth and inflammatory response in the small intestine of rat. Male 8-week-old Sprague-Dawley rats were randomly divided into four groups: normal oxygen sedentary group (n=30), normal oxygen exercise group (n=30), low oxygen sedentary group (n=30) and low oxygen exercise group (n=30). Exercise training was on a treadmill for 1 hr per day on days 3, 6, and 9 in the hypoxia condition. Hematological profiles, hematolxylin and eosin staining, fluorescence <I>in situ</I> hybridization, reverse transcription-polymerase chain reaction and Western blot were used to analyze the effect of simulated altitude training on the amount of bacteria, and expression of mRNA and protein. Simulated exercise training significantly increased red blood cells and hematocrit. The small intestinal mucosa barrier was significantly injured by the simulated altitude exercise training. Comparatively more bacterial growth was evident in the small intestine by the simulated altitude exercise training. mRNA levels of interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α) and protein expression of nuclear factor-kappa B (NF-κB) were significantly elevated by simulated altitude exercise training. These results suggest that the simulated altitude exercise training may impair the small intestinal mucosa barrier via elevation of bacterial growth and inflammatory cytokines (IL-6, TNF-α) and the up-regulation of NF-κB in the rats.</P>

        An Efficient Model Based on Smoothed ℓ<sub>0</sub> Norm for Sparse Signal Reconstruction

        Compressed sensing (CS) is a new theory. With regard to the sparse signal, an exact reconstruction can be obtained with sufficient CS measurements. Nevertheless, in practical applications, the transform coefficients of many signals usually have weak sparsity and suffer from a variety of noise disturbances. What's worse, most existing classical algorithms are not able to effectively solve this issue. So we proposed an efficient algorithm based on smoothed ℓ<sub>0</sub> norm for sparse signal reconstruction. The direct ℓ<sub>0</sub> norm problem is NP hard, but it is unrealistic to directly solve the ℓ<sub>0</sub> norm problem for the reconstruction of the sparse signal. To select a suitable sequence of smoothed function and solve the ℓ<sub>0</sub> norm optimization problem effectively, we come up with a generalized approximate function model as the objective function to calculate the original signal. The proposed model preserves sharper edges, which is better than any other existing norm based algorithm. As a result, following this model, extensive simulations show that the proposed algorithm is superior to the similar algorithms used for solving the same problem.

      • Research on China's Steel Demand Using Combined Forecast

        This research conducts an error analysis between the forecasting value and the actual value of steel demand of 2010 in China, which is based on the analysis of forecasting methods and their results of Chinese steel demand in the existing studies, and then forecasts China’s steel demand in 2015 by making use of a combined forecasting method. The combined forecasting method includes two stages. In the first stage, with reference to actual steel demand of China in 2010, a threshold is set and some forecasting results are selected according to the error between the forecasting value and actual value of steel demand of each method. In the second stage, weights of corresponding selected methods are determined which are based on the error. And the final demand of China’s steel in 2015 is forecasted through the combined forecasting method.

        A retrospective study on the efficacy of Roxadustat in peritoneal dialysis patients with erythropoietin hyporesponsiveness

        Background/Aims: Roxadustat, an oral medication for treating renal anemia, is a hypoxia-inducible factor prolyl hydroxylase inhibitor used for regulating iron metabolism and promoting erythropoiesis. To investigate the efficacy and safety of roxadustat in patients undergoing peritoneal dialysis (PD) with erythropoietin hyporesponsiveness. Methods: Single-center, retrospective study, 81 PD patients (with erythropoietin hyporesponsiveness) were divided into the roxadustat group (n = 61) and erythropoiesis-stimulating agents (ESAs) group (n = 20). Hemoglobin (Hb), total cholesterol, intact parathyroid hormone (iPTH), brain natriuretic peptide (BNP), related indicators of cardiac function and high-sensitivity C-reactive protein (hs-CRP) were collected. Additionally, adverse events were also recorded. The follow-up period was 16 weeks. Results: The two groups exhibited similar baseline demographic and clinical characteristics. At baseline, the roxadustat group had a mean Hb level of 89.8 ± 18.9 g/L, while the ESAs group had a mean Hb level of 95.2 ± 16.0 g/L. By week 16, the Hb levels had increased to 118 ± 19.8 g/L (p < 0.05) in the roxadustat group and 101 ± 19.3 g/L (p > 0.05) in the ESAs group. The efficacy of roxadustat in improving anemia was not influenced by baseline levels of hs-CRP and iPTH. Cholesterol was decreased in the roxadustat group without statin use. An increase in left ventricular ejection fraction and stabilization of BNP were observed in the roxadustat group. Conclusions: For PD patients with erythropoietin hyporesponsiveness, roxadustat can significantly improve renal anemia. The efficacy of roxadustat in improving renal anemia was not affected by baseline levels of hs-CRP0 and iPTH.

        Efficient fluorescent white organic light-emitting devices with a reduced efficiency roll-off based on a blue ambipolar fluorescent emitter

        White organic light-emitting devices (WOLEDs) with fluorescent donor-acceptor-substituted spirobifluorene compounds (red 2-diphenylamino-7-(2,2-dicyanovinyl)-9,90-spirobifluorene and blue 2- diphenylamino-7-(2,2-diphenylvinyl)-9,90-spirobifluorene) have been fabricated. The optimized WOLEDs shows a maximum current efficiency 5.9 cd/A and very low efficiency roll-off. From the brightness at maximum current efficiency to high brightness of 10000 cd/m2, the current efficiency rolloff is only 0.4%. It can be attributed to the ambipolar blue fluorescent emitter with voltage-independnet mobility which makes the device having a broader charge recombination zone and balance of carrier transport.

        Damping for Wind Turbine Electrically Excited Synchronous Generators

        The electrically excited synchronous generator (EESG) is applied in wind turbine systems recently. In an EESG control system, electrical torque is affected by stator flux and rotor current. So the control system is more complicated than that of the permanent-magnet synchronous generator (PMSG). Thus, the higher demanding of the control system is required especially in case of wind turbine mechanical resonance. In this paper, the mechanism of rotor speed resonant phenomenon is introduced from the viewpoint of mechanics firstly, and the characteristics of an effective damping torque are illustrated through system eigenvalues analysis. Considering the variables are tightly coupled, the four-order small signal equation for torque is derived considering stator and rotor control systems with regulators, and the bode plot of the closed loop transfer function is analyzed. According to the four-order mathematical equation, the stator flux, stator current, and electrical torque responses are derived by torque reference step and ramp in MATLAB from a pure mathematical deduction, which is identical with the responses in PSCAD/EMTDC simulation results. At last, the simulation studies are carried out in PSCAD software package to verify the resonant damping control strategy used in the EESG wind turbine system.

      • Real-time prediction of dynamic irregularity and acceleration of HSR bridges using modified LSGAN and in-service train

        Dynamic irregularity and acceleration of bridges subjected to high-speed trains provide crucial information for comprehensive evaluation of the health state of under-track structures. This paper proposes a novel approach for real-time estimation of vertical track dynamic irregularity and bridge acceleration using deep generative adversarial network (GAN) and vibration data from in-service train. The vehicle-body and bogie acceleration responses are correlated with the two target variables by modeling train-bridge interaction (TBI) through least squares generative adversarial network (LSGAN). To realize supervised learning required in the present task, the conventional LSGAN is modified by implementing new loss function and linear activation function. The proposed approach can offer pointwise and accurate estimates of track dynamic irregularity and bridge acceleration, allowing frequent inspection of high-speed railway (HSR) bridges in an economical way. Thanks to its applicability in scenarios of high noise level and critical resonance condition, the proposed approach has a promising prospect in engineering applications.
