A rare cause of unexplained severe polyhydramnios: A case report of Bartter’s syndrome

Bartter’s syndrome (BS) is a severe tubulopathy causing renal salt- wasting, severe polyuria, dehydration, failure to thrive and secondary hyperaldosteronism. The incidence is about 1 in 1,000,000. It is an autosomal recessive disorder caused by inactivating mutations in genes that encode membrane proteins of the thick ascending limb of the loop of Henle. Diagnosis is usually made postnatally by a laboratory test or becomes evident when polyuria appears to the baby. Ultrasound findings are nonspecific without apparent fetal or placental abnormalities, thus it is difficult to suspect the disease. Here we report a case of 33-year-old nulliparous woman who was referred to our center at 31 weeks’ gestation because of severe polyhydramnios. The patient had underwent twice amnioreduction of 2L at 27 and 30 weeks of gestational age, respectively. At the time of visit, the amniotic fluid index was 44.12cm. Small stomach and enlarged liver were observed, thus esophageal atresia with tracheal esophageal fistula was suspected. Persistent right umbilical vein was also observed. Karyotyping showed no specific chromosomal abnormality. At our center, 3 amnioreductions were performed with a total fluid volume drainage of 8.4L (2.5L at 31weeks, 3L at 33weeks, and 2.9L at 35weeks of gestation). During the whole pregnancy period, total of 12.4 L of amnioreduction was done. After an uneventful antenatal period, a 2.21kg male baby was delivered by cesarean section at 36+1weeks of gestation. Laboratory test of the baby showed hyponatremia, hypokalemia and elevation of plasma renin and aldosterone. In addition, the baby showed a sign of polyuria. Therefore, with presumptive diagnosis of BS, genetic test was done and the result is pending. In summary, we report a rare case of prenatally diagnosed BS. When unexplained severe polyhydramnios is suspected, biochemical assessment of amniotic fluid may provide clue to the diagnosis. Our report may provide informative data for this rare disease.
cDNA Library Construction of Aphis gossypii Using Gateway Cloning System

Aphis gossypii was widely distributed throughout the tropical, subtropical and temperate zone. The chemical control of A. gossypii is becoming problem because it was rapidly appeared resistance expression to chemicals. We will attempt to resolve the this problem using RNAi technique. Besides, RNAi technology can be helpful to study the target genes of A. gossypii. In this study we produce cDNA library construction using gateway cloning system for selecting target gene in order to control of A. gossypii using RNAi. As a result, the 100-400bp of insert size, which is appropriate for RNAi was confirmed. Most of insert gene is associated with A. gossypii, after that insert sequence was compared with DNA databases and EST databases using NCBI blast search. Consequentially, A. gossypii of cDNA library with the titer of 3.15x105 clones were completed. And we will perform the LR recombination to transfer cDNA library into TRV2 (tobacco rattle virus) vector with att site. Then, after performing transformation using Agrobacterium tumefaciens (GV 2260), we inoculated to cucumber with A. tumefaciens. An insecticidal effect or a repellent activity against A. gossypii by changing behavior in transgenic cucumber plants were conformed. Also, the selecting target gene in order to control A. gossypii using RNAi may be provided.
Fabrication of dihydroxyflavone-conjugated hyaluronic acid nanogels for targeted antitumoral effect

<P><B>Abstract</B></P> <P>We prepared hyaluronic acid (HA)-based nanogels conjugated with dihydroxyflavone (DHF) and evaluated their cellular uptake and antitumoral efficiency. 4-(4,6-dimethoxy-1,3,5-triazin-2-yl)-4-methylmorpholinium chloride (DMTMM) was used as a conjugation agent for esterification between DHF and HA as well as crosslinking among HA. The conjugations were confirmed by nuclear magnetic resonance spectroscopy, UV/vis spectroscopy, and high-performance liquid chromatography. The size and Zeta-potential of the DHF/HA nanogels were reduced with an increase in the concentration of DMTMM due to the involvement of more HA molecules for the conjugation reactions. The DHF/HA nanogel with a smaller size was greatly taken up by two kinds of tumor cells (HeLa and HepG2), compared to NIH3T3. The cell viabilities were reduced to approximately 60% for HeLa and HepG2 cells after 48 h post treatment with DHF/HA nanogels.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Hyaluronic acid molecules are conjugated among themselves in nanogel form. </LI> <LI> Dihydroxyflavones are conjugated to hyaluronic acid nanogels. </LI> <LI> A low concentration of a conjugation agent leads to the nanogel size reduction. </LI> <LI> The nanogel specifically reduced the viability of tumor cells to approximately 60%. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>
Are children with asthma in South Korea also associated with vitamin D deficiency?

Recently, epidemiologic studies have shown that the lack of serum vitamin D levels may be associated with high asthma prevalence, but its effect is still controversial, depending on season, area, and food consumption. We aimed to examine the association of serum vitamin D levels with the prevalence of pediatric asthma in Korea. A total of 80 children (50 asthmatic children and 30 healthy controls) aged 6-14 years were participated in this study. Serum vitamin D levels were measured and compared between the two groups. Moreover, the relationship of serum vitamin D levels with results of pulmonary function test and environmental factors (lifestyle habits and residential factors) collected by a questionnaire survey were examined in asthmatic patients. Serum vitamin D levels in asthmatic children (16.63±4.20 ng/mL) were significantly lower than that in healthy controls (24.24±6.76 ng/mL) (p<0.05). Also, we found that the prevalence of asthma increase to 0.79-fold (odds ratio, 0.79; 95% confidence interval, 0.71 to 0.88; p<0.001) as serum vitamin D level is 1 ng/mL decreases. The increased time spent in outdoor could affect the increases of serum vitamin D levels significantly. However, no associations of serum vitamin D with pulmonary function and residential environmental factors (i.e., housing type, living floor, and indoor activity time) were observed. Our findings suggest that serum vitamin D levels were also associated with pediatric asthma in Korea. Moreover, management of serum vitamin D level in asthmatic children would be a promising approach for preventing exaggeration of their severity.

<P>Degradation on the current saturation of the output characteristics in amorphous indium–gallium–zinc–oxide (a-IGZO) thin-film transistors with the bottom-gate structure is investigated. As the drain-to-source voltage ( <TEX>${V}_{\textsf {DS}}$</TEX>) increases at a fixed gate-to-source voltage ( <TEX>${V}_{\textsf {GS}}$</TEX>), the current from drain to source ( <TEX>${I}_{\textsf {DS}}$</TEX>) becomes nonsaturated and increases due to the mobility enhancement and <TEX>${I}_{\textsf {DS}}$</TEX> also decreases again due to the electron trapping into the gate insulator and/or an interface, which occurs mainly at the channel edge near a drain. Analysis is validated by using the pulsed <TEX>${I}_{\textsf {DS}}$</TEX>– <TEX>${V}_{\textsf {DS}}$</TEX> measurement. Nonsaturated current is attributed to the Joule heating-assisted mobility enhancement and the thermally activated electron trapping, which is the reason why the nonsaturation becomes more prominent as the IGZO active thin film becomes thinner. Furthermore, it is found that the rate of the increased <TEX>${I}_{\textsf {DS}}$</TEX> per increasing <TEX>${V}_{\textsf {DS}}$</TEX> gets higher as either the channel width ( <TEX>${W}$</TEX>) increases or the channel length ( <TEX>${L}$</TEX>) decreases; whereas, the rate of the decreased <TEX>${I}_{\textsf {DS}}$</TEX> per increasing <TEX>${V}_{\textsf {DS}}$</TEX> becomes higher as the device size, i.e., <TEX>${W}\times {L}$</TEX>, increases. The former is well correlated with Joule heating while the latter with the self-heating-assisted electron trapping due to a total heat accumulated in an active layer.</P>
Detergent Free Fractionation of Nuclei from Mouse Embryonic Stem Cells

Mouse embryonic stem cells (ESCs), with their ability to self-renewal, and to differentiate into virtually all types, have become the major focus in contemporary cell-based therapy and regenerative medicine. Many studies about the signaling mechanism regulating self-renewal or differentiation have been performed in mouse ESCs. Pure nuclei fractionation is needed for the studies of gene regulation involved in self-renewal or differentiation of ESCs. Here we develop more efficient method for isolation of pure nuclei from mouse ESCs than the conventional method. We isolated more pure nuclei fraction with detergent-free buffer by freezing-thawing cycling. Mouse ESC nuclei isolated by our method were more pure than those by the conventional method. Oct4, a well-known stemness marker and nucleus protein, in the nuclei fraction, specifically bound to the oct4 binding site. Light electroscopy and electron microscopy showed the intactness of the isolated nuclei fraction. Flow cytometric analysis of the population of isolated nuclei revealed no signs of cell cycle specific losses of nuclei during isolation procedure. This contributes to studying nuclear signaling molecules involved in self-renewal and differentiation in mouse ESCs.
