        Optimization of Ascorbic Acid Extraction from Rugosa Rose (Rosa rugosa Thunb.) Fruit Using Response Surface Methodology and Validation of the Analytical Method

        In this study, ultrasound-assisted extraction was performed to extract ascorbic acid from rugosa rose (Rosa rugosa Thunb.) fruit. The optimal conditions were investigated by response surface methodology, using two variable including reaction time (16-44 min) and temperature (16-44°C). The ascorbic acid extraction was sensitive to the reaction time rather than the reaction temperature, and the optimal conditions for ascorbic acid extraction were 25°C and 30 min. Ascorbic acid and gallic acid in the rugosa rose fruit extract were completely separated by HPLC, with a resolution factor of over 1.5 between the two. The correlation coefficient of the ascorbic acid was 0.999 in a linearity test for 50-150 μg/mL concentration of extract. The limit of detection and limit of quantification values were 0.16 μg/mL and 29.89 μg/mL, respectively. The relative standard deviations (RSD) for repeatability and reproducibility were determined, and each RSD showed good precision at less than 5% (N=6).

        에탄올 생산 향상을 위한 발효저해물질 제거와 리그닌 유래 발효저해물질이 에탄올 발효에 미치는 영향

        본 연구에서는 옥살산 전처리 바이오매스의 액상가수분해산물에 포함된 발효저해물질을 전기투석과 XAD 수지 처리하여 제거한 후 에탄올을 생산하였다. 전기투석 과정에서 아세트산은 대부분 제거되었으며(95.6%), 비이온성 발효저해물질(Total phenolic compound: TPC, 5-hydroxymethyl furfural: HMF, furfural)은 XAD 수지 처리에 의해 효과적으로 제거되었다. 전기투석과 XAD 수지 처리된 액상가수분해산물로 발효를 수행한 결과 XAD 수지의 침지시간이 짧을수록 에탄올 생산이 향상되었다. 최대 에탄올 생산은 발효 72시간 후 6.16 g/ℓ로 전기투석 후 액상가수분해산물을 XAD-4 수지에서 5분 침지하였을 때 나타났다. 리그닌 유래 발효저해물질 중 syringaldehyde는 저농도(1 mM, 2 mM)에서 에탄올 생산을 향상시켰으며 5 mM에서는 발효에 부정적인 영향을 주었다. 리그닌 유래 발효저해물질의 시너지효과를 확인하고자 합성배지로 발효를 수행하였으며, Syringaldehyde (1 mM)와 ferulic acid (1 mM) 합성배지를 이용하여 발효를 수행한 결과 syringaldehyde보다 ferulic acid의 영향으로 에탄올 생산이 감소했다. In this study, ethanol was produced from a biomass hydrolysate that had been treated by electrodialysis (ED) and Amberlite XAD resin to remove fermentation inhibitors. Most of the acetic acid (95.6%) was removed during the ED process. Non-ionizable compounds such as total phenolic compounds, 5-hydroxymethyl furfural, and furfural were effectively removed by the XAD resin treatment. Ethanol production was improved when the ED-treated hydrolysate was treated with XAD-4 resin for a short reaction time. The highest ethanol production from ED-treated hydrolysate was 6.16 g/ℓ (after 72 h of fermentation) when the treatment with XAD-4 resin was for 5 min. Among the ligninderived fermentation inhibitors tested, syringaldehyde in low concentrations (1 and 2 mM) in the hydrolysate increased ethanol production, whereas a high concentration (5 mM) inhibited the ethanol production process. A synthetic medium containing syringaldehyde and ferulic acid was prepared to investigate the synergistic effect of inhibitors on ethanol fermentation. Ethanol production decreased in the mixture of 1 mM syringaldehyde and 1 mM ferulic acid, implying that the effect of ferulic acid on ethanol fermentation is comparable to that of syringaldehyde.

        Nutritional components of Ojuk leaf tea and Ojuk shoot tea prepared from the leaves and shoots of black bamboo (Phyllostachys nigra Munro) by tea manufacturing process were evaluated. In addition, the extraction yield of water soluble components from these teas in the general tea brewing condition (water extraction in 80oC for 10 min.) and the contents of polyphenol and flavonoid were compared with not only the dried raw materials, but also green tea and mate tea. Finally, offline and online scavenging activities against 2,2`-azino-bis(3-ethylbenzothiazoline- 6-sulphonic acid) (ABTS) and di(phenyl)-(2,4,6 trinitrophenyl)iminoazanium free radicals were investigated to evaluate the antioxidant activity and explore the components showing ABTS free radical scavenging activity from tea infusion. These results demonstrated that these teas from black bamboo contain various nutritional components and can be used as traditional tea beneficial to human health.

        학교의 규모가 확대될수록 그에 따르는 도면 뿐만 아니라 도면 작업도 엄청나게 능어난다. 이러한 어려운 작업으로 인해 많은 인력과 시간을 투자해야만 한다. 그래서 이러한 문제점을 해결하기 위한 방안으로 Compus 도면 관리 시스템을 설계하고 구현 하였다. 본 시스템은 평면도와 전경도의 여러가지 공사 정보를 관리하고, 여러 가지 공사 상황 및 도면을 수정할 수 있는 drawing tool을 제공한다. As the size of campus environments, being large school administrators have much to do with drawings and drawing-related works. This situation requires much investments in human efforts and time. We have designed and implemented the Campus-drawing Management System to solve this problem partially. This system will manage a complete view, a ground plan and much information of drawings. And this system provides drawing tools for modifying drawings and maintaining informations about plans.

        대학생을 위한 리더십향상 집단상담 구안 및 효과 연구

        현대사회에서의 리더십은 크게 셀프리더십, 팔로워십, 능력부여리더십, 서번트리더십으로 분류될 수 있다. 한국의 대학생은 향후 인생의 목표를 설정하고 계획하여 추진하고 더불어 공동체의 일원으로 서로 교류하고 성장 한다는 발달적 과제를 가지고 있다. 따라서 대학생의 리더십은 셀프리더십을 바탕으로 한 자기관리, 대인관계 관리, 공동체 참여로 구성된다. 본 연구에서는 대학생의 리더십을 향상시키기 위한 집단상담을 구안하였으며 구조화된 대학생 리더십향상 집단상담을 2005년 1월에 실시하였다. 참여한 총 10명의 집단원들을 대상으로 집단상담 전 후에 각 구성요소를 측정하는 설문조사를 실시하였으며, 통계적으로 유의미한 차이는 아니었으나 자기관리, 대인관계관리, 공동체참여 각 영역 모두에서 향상된 양상을 보였다. 또한 집단원들의 회기별 피드백을 토대로 본 프로그램의 효과를 검증하였으며 본 프로그램의 보다 효과적인 적용을 위한 논의를 덧붙였다. Leadership, nowadays, can be classified into four different kinds: self-leadership, followership, empowering-leadership, and servant leadership. Korean university students have developmental tasks as members of group such as setting a life goal, making a plan, making an action, associating with others, and being advanced. Therefore, the elements of leadership program for university students based on self-leadership are self management, interpersonal relationship management, and participating in a group. In this study, a structured program for developing leadership of university students was designed, and operated on Jan, 2005. Surveys which measured each elements of the program were given to ten students who were participated in a group before and after the program. A statistical result was not significant. However, each elements, self management, interpersonal relationship management, and participating in a group, showed a progress. Session reviews by students were also used to verify the effect of the program. Further suggestions and studies for more effective application of the program were discussed.

        Endoscopic submucosal tunnel dissection salvage technique for ulcerative early gastric cancer.

        <P>Endoscopic submucosal dissection is an effective treatment modality for early gastric cancer (EGC), though the submucosal fibrosis found in ulcerative EGC is an obstacle for successful treatment. This report presents two cases of ulcerative EGC in two males, 73- and 80-year-old, with severe fibrosis. As endoscopic ultrasonography suggested that the EGCs had invaded the submucosal layer, the endoscopic submucosal tunnel dissection salvage technique was utilized for complete resection of the lesions. Although surgical gastrectomy was originally scheduled, the two patients had severe coronary heart disease, and surgeries were refused because of the risks associated with their heart conditions. The endoscopic submucosal tunnel dissection salvage technique procedures described in these cases were performed under conscious sedation, and were completed within 30 min. The complete en bloc resection of EGC using endoscopic submucosal tunnel dissection salvage technique was possible with a free resection margin, and no other complications were noted during the procedure. This is the first known report concerning the use of the endoscopic submucosal tunnel dissection salvage technique salvage technique for treatment of ulcerative EGC. We demonstrate that endoscopic submucosal tunnel dissection salvage technique it is a feasible method showing several advantages over endoscopic submucosal dissection for cases of EGC with fibrosis.       • Use of Electroacupuncture at ST36 to Inhibit Anaphylactic and Inflammatory Reaction in Mice

        <P><I>Objective:</I> Electroacupuncture (EA) has been used to treat myalgia, allergy and gastroenteropathy in Korea. To determine whether EA can treat anaphylactic and inflammatory reactions, the effect of EA was investigated in a murine model. <I>Methods:</I> EA stimulation of the ST36 acupoint was performed for 10 min. Using a passive cutaneous anaphylaxis (PCA) model, the antianaphylactic effects of EA were examined. Interleukin-6 and tumor necrosis factor-α were measured using the ELISA method. The level of nuclear factor (NF)-κB/RelA protein and NF-κB DNA-binding activity was determined using the Western blot analysis and the transcription factor enzyme-linked immunoassay method. <I>Results:</I> EA inhibits PCA and β-hexosaminidase release, IL-6 secretion on the PCA, and in addition, EA reduces NF-κB DNA-binding activity. <I>Conclusion:</I> These results indicate that EA may possess antianaphylactic and antiinflammatory properties.</P><P>Copyright © 2007 S.         이순신 장군을 재현한 영화에 나타난 인물 변화 양상 연구: <명량>과 <한산: 용의 출연>을 중심으로

        In the Yi Sun-shin trilogy, consisting of “Roaring Currents” and “Hansan: Rising Dragon,” directed by Kim Han-min, we have examined the evolution of Yi Sun-shin’s portrayal in response to the shifting social landscape in South Korea since the 2010s. Specifically, we have analyzed how the representation of Yi Sun-shin transformed in these films against the backdrop of significant historical events, namely the Sewol Ferry sinking and the global COVID-19 pandemic, which were prominent concerns during the production of “Roaring Currents” and “Hansan: Rising Dragon,” respectively. In “Roaring Currents,” the film accentuates Yi Sun-shin’s heroic attributes as the primary determinant of victory in the Battle of Myeongnyang. The narrative places Yi Sun-shin in the role of an authoritative leader, with a spotlight on his self-sacrificial actions. This portrayal of Yi Sun-shin as a resolute and charismatic leader significantly contributed to the film’s box office success.In contrast, during the production of “Hansan: Rising Dragon,” the world grappled with the challenges posed by the COVID-19 pandemic. Consequently, the film shifts its focus away from Yi Sun-shin’s heroism and instead emphasizes the roles of ordinary people and sailors, portraying Yi Sun-shin as an inclusive leader. The film highlights the themes of cooperation and solidarity, particularly in contrast to the Japanese enemy. By means of a comparative and analytical approach applied to these two works, it becomes evident that the characterization of Yi Sun-shin is responsive to the changing socio-cultural context and subtly mirrors these societal shifts. Moreover, the films communicate the evolving significance of Yi Sun-shin’s role in Korean society, thereby imparting greater depth to his cinematic representation.

        트레드밀 운동이 고지방식이를 섭취한 비만 쥐 골격근의소포체 스트레스에 미치는 영향

        본 연구는 고지방식이로 유도된 비만 흰쥐를 대상으로 지구성 운동이 체중변화, 글루코스 내성 및 소포체 스트레스(ER-stress)에 반응에 관여하는 단백질 발현에 미치는 영향을 규명하는데 목적이 있다. 실험동물은 16주간 고지방식이를 먹인 흰쥐를 사용하였으며 집단은 Sprague-Dawley Control(SD-Con/n=4)집단, High Fat Control(HF-Con/n=4)집단, High Fat exercise(HF-Exe/n=4)으로 구분하고 트레드밀 운동을 하루 30분, 주 5회 총 8주간 21m/min의 속도로 실시하였다. 먼저, 체중과 글루코스 내성은 HF-Con 집단에서 유의하게 가장 높았지만 트레드밀 운동을 실시한 HF-Exe집단에서 유의한 감소를 나타났다. 또한 소포체 스트레스에 반응에 관여하는 glucose-regulated protein 78 (BiP/GRP78), activating transcription factor 6 (ATF6), Pancreatic ER kinase(PERK) 및 inositol-requiring 1α (IRE-1α)/X-box binding protein 1 (XBP-1)은 HF-Con 집단에서 유의하게 증가를 나타냈고, HF-Exe 집단에서 유의한 감소가 나타났다. 따라서 지구성 운동은 고지방 식이로 유도된 비만 휜쥐의 체중 감소와 글루코스 내성을 개선시키고, 소포체 스트레스를 감소시키는데 긍정적인 영향을 미치는 것으로 생각된다. This study was to performed to the effect of 8-week endurance exercise influences on body weight, glucose tolerance and ER-stress in soleus of 16weeks Rats fed High-Fat diet. Rats were randomly assigned to 3 group; (1)Sprague-Dawley Control diet (SD-Con/n=4), (2)High-Fat diet Control (HF-Con/n=4), (3)High-Fat diet Exercise (HF-Exe/n=4). Exercise group ran on the treadmill for 30min/day at the level of 21m/min for 5days/week during 8weeks. Results showed that body weight and glucose tolerance of the HF-Con group was remarkably increased(p<.05) compared to other groups. However, HF-Exe group significantly decreased body weight and glucose tolerance compared to HF-Con group. Moreover, level of GRP78, ATF6, PERK and IER1?, which are main proteins of ER-stress were significantly increased in HF-Con group higher than other group, whereas HF-Exe group significantly decreased the expression of GRP78, ATF6, PERK and IER1?. Taken together, these finding suggested that the reduction of the body weight, glucose tolerance and unfolded protein response by treadmill exercise may represent a positive adaptation protecting against high-fat diet-induced ER stress.
