        Excessive mechanical strain accelerates intervertebral disc degeneration by disrupting intrinsic circadian rhythm

        Night shift workers with disordered rhythmic mechanical loading are more prone to intervertebral disc degeneration (IDD). Our results showed that circadian rhythm (CR) was dampened in degenerated and aged NP cells. Long-term environmental CR disruption promoted IDD in rats. Excessive mechanical strain disrupted the CR and inhibited the expression of core clock proteins. The inhibitory effect of mechanical loading on the expression of extracellular matrix genes could be reversed by BMAL1 overexpression in NP cells. The Rho/ROCK pathway was demonstrated to mediate the effect of mechanical stimulation on CR. Prolonged mechanical loading for 12 months affected intrinsic CR genes and induced IDD in a model of upright posture in a normal environment. Unexpectedly, mechanical loading further accelerated the IDD in an Light-Dark (LD) cycle-disrupted environment. These results indicated that intrinsic CR disruption might be a mechanism involved in overloading-induced IDD and a potential drug target for night shift workers.

        Value of contrast-enhanced ultrasonography in microwave ablation treatment of symptomatic focal uterine adenomyosis

        Purpose: This study evaluated the value of contrast-enhanced ultrasonography (CEUS) in the ultrasound-guided microwave ablation (MWA) treatment of symptomatic focal uterine adenomyosis.Methods: This retrospective study was conducted between March 2020 and January 2023, enrolling 52 patients with symptomatic focal uterine adenomyosis who had undergone MWA. All patients were examined with CEUS before and after MWA. The non-perfused volume (NPV) was compared between CEUS and dynamic contrast-enhanced magnetic resonance imaging (DCEMRI) following ablation. Therapeutic efficacy and safety were evaluated at 3-, 6-, and 12-month follow-ups. Additionally, this study explored the correlations between pre-treatment CEUS features and a volume reduction ratio indicating sufficient ablation, defined as 50% or more at the 3-month follow-up.Results: No significant differences in NPV were noted between CEUS and DCE-MRI immediately after MWA and during follow-up (all P>0.05). At the 3-month follow-up, the median VRRs for the uterus and adenomyosis were 33.2% and 63.9%, respectively. Sufficient ablation was achieved in 69.2% (36/52) of adenomyosis cases, while partial ablation was observed in the remaining 30.8% (16/52). The identification of non-enhancing areas on pre-treatment CEUS was associated with sufficient ablation (P=0.016). At the 12-month follow-up, significant decreases were observed in both the uterine and adenomyosis volumes (all P<0.001). Dysmenorrhea and menorrhagia were significantly alleviated at 12 months, and no major complications were encountered.Conclusion: CEUS can be used to evaluate the ablation zone of focal adenomyosis that has been treated with MWA, similarly to DCE-MRI. The identification of non-enhancing areas on pretreatment CEUS indicates satisfactory treatment outcomes.         A New Method for Determination of 3D Active Earth Pressure of Unsaturated Backfills

        Determinations of active earth pressures are commonly performed two-dimensionally (2D) based on completely saturated and/or dry assumptions, though the soil in cases of geotechnical interest is mostly unsaturated and the earth pressures are usually of conspicuous three-dimensional (3D) features. In this paper, a novel finite prismoid element method (FPEM) for calculating the lateral earth pressures acting against the retaining wall is suggested. The main feature of the FPEM is that the whole backfill is discretized into numerous horizontally distributed prismoid elements that might characterized with different soil properties. For unsaturated backfills, the prismoid elements are characterized with various soil cohesions and unit soil weights. Upper bound solutions to active earth pressures under 2D and 3D conditions with and without suction are both calculated and compared with several other analytical ones, indicating the reliability and applicability of the proposed method. The responses of unsaturated backfills to surcharge loads on the crest are numerically studied and discussed. An illustrative example is reexamined to further demonstrate the practical use of the technique.

        Mixed organic acids improve nutrients digestibility, volatile fatty acids composition and intestinal microbiota in growing-finishing pigs fed high-fiber diet

        Objective: The objective of this study was to investigate effects of mixed organic acids (MOA) on nutrient digestibility, volatile fatty acids composition and intestinal microbiota in growing-finishing pigs fed high wheat bran diet. Methods: Six crossbred barrows (Duroc×Landrace×Yorkshire), with an average body weight 78.8±4.21 kg, fitted with T-cannulas at the distal ileum, were allotted to a double 3×3 Latin square design with 3 periods and 3 diets. Each period consisted of a 5-d adjustment period followed by a 2-d total collection of feces and then a 2-d collection of ileal digesta. The dietary treatments included a corn-soybean-wheat bran basal diet (CTR), mixed organic acid 1 diet (MOA1; CTR+3,000 mg/kg OA1), mixed organic acid 2 diet (MOA2; CTR+2,000 mg/kg OA2). Results: Pigs fed MOA (MOA1 or MOA2) showed improved (p<0.05) apparent total tract digestibility (ATTD) of gross energy, dry matter and organic matter, and pigs fed MOA2 had increased (p<0.05) ATTD of neutral detergent fiber compared to CTR. Dietary MOA supplementation decreased (p<0.05) pH value, and improved (p<0.01) concentrations of lactic acid and total volatile fatty acids (TVFA) in ileum compared to CTR. Pigs fed MOA showed higher (p<0.05) concentration of acetic acid, and lower (p<0.05) content of formic acid in feces compared to CTR. Pigs fed MOA1 had increased (p<0.05) concentration of TVFA and butyric acid in feces. Pigs fed MOA1 showed higher concentration of Lactobacillus and lower concentration of Escherichia in feces compared to CTR. Conclusion: Dietary supplementation of MOA 1 or 2 could improve nutrients digestibility, TVFA concentration and intestinal flora in growing-finishing pigs fed high fiber diet.

        Characterization of a Thermophilic Hemoglobin-degrading Protease from Streptomyces rutgersensis SCSIO 11720 and Its Application in Antibacterial Peptides Production

        High value utilization of byproduct of slaughterhousesis regarded as beneficial to both economy andenvironment. A hemoglobin degrading protease PA720was purified from fermentation broth of a Porites luteaassociated strain Streptomyces rutgersensis SCSIO 11720(SCSIO 11720) and exhibited maximal activity at 70°C,pH 10.5. The isolated strain was identified by genotypicmethod and 16 s RNA gene sequence was deposited inGenBank under accession No. KC414842. The enzymeexhibited excellent thermostability since it was stable at 70°Cwith t1/2 value of 1,386.3 min. Kinetic and thermodynamicstudy showed the activation energy (Ea), ΔG* (free energy),ΔH* (enthalpy) and ΔS* (entropy) for protease deactivationwere 62.71 kJ/mol, 74.26 kJ/mol, 59.86 kJ/mol, and-41.97 J/mol/K, respectively. Highest specific activity ofprotease PA720 was observed when using casein (9,953 U/mg)and hemoglobin (9,854 U/mg) as substrates. Hemoglobinhydrolysate prepared by protease PA720 showed significantantibacterial activity towards Escherichia coli, Staphylococcusaureus and Bacillus subtilis, indicating this protease couldbe used as an instrumental enzyme for production ofhemoglobin-based antibacterial peptides. Based on partialamino acid sequences of the enzyme PA720, the full geneencoding this protease was obtained by degenerate primerPCR and has been deposited in GenBank under accessionNo. KC414842. The deduced amino acid sequence exhibitshomology with other microbial thermophilic proteases insubtilisin family.

      • Seed-Mediated Synthesis of Ag Nanocubes with Controllable Edge Lengths in the Range of 30−200 nm and Comparison of Their Optical Properties

        <P>Silver nanocubes with edge lengths controllable in the range of 30−200 nm were synthesized using an approach based on seeded growth. The keys to the success of this synthesis are the use of single-crystal Ag seeds to direct the growth and the use of AgNO<SUB>3</SUB> as a precursor to elemental Ag, where the byproduct HNO<SUB>3</SUB> can block both the homogeneous nucleation and evolution of single-crystal seeds into twinned nanoparticles. Either spherical (in the shape of a cuboctahedron) or cubic seeds could be employed for this growth process. The edge length of the resultant Ag nanocubes can be readily controlled by varying the amount of Ag seeds used, the amount of AgNO<SUB>3</SUB> added, or both.       • A Fault Diagnosis of ZPW-2000A Tuning Area based on the WNN

        With the rapid development of china railway, ZPW-2000A track circuit has been widely used. Tuning area, which is an important part of the ZPW-2000A track circuit, is not only the relationship between the signal transmission quality, and determine the effect of electrical insulation between adjacent sections. Therefore, the study of fault diagnosis aspects for tuning area is urgent and significant. In this paper, using the theory of transmission line a model of track circuit is built, the comparison of the actual data and experimental data of the track surface voltage envelope curve shows the correctness of this model. Owning to the good time-frequency characteristics of wavelet and the nonlinear mapping features of neural network, a fault diagnosis of ZPW-2000A tuning area based on the wavelet neural network(WNN) is proposed. Combined with the practical failure situation of railway site, the fault diagnosis method in this paper can accurately identify failure modes of tuning area.

        TCP10L synergizes with MAD1 in transcriptional suppression and cell cycle arrest through mutual interaction

        T-complex protein 10A homolog 2 (TCP10L) was previously demonstrated to be a potential tumor suppressor in human hepatocellular carcinoma (HCC). However, little is known about the molecular mechanism. MAX dimerization protein 1 (MAD1) is a key transcription suppressor that is involved in regulating cell cycle progression and Myc-mediated cell transformation. In this study, we identified MAD1 as a novel TCP10L-interacting protein. The interaction depends on the leucine zipper domain of both TCP10L and MAD1. TCP10L, but not the interaction-deficient TCP10L mutant, synergizes with MAD1 in transcriptional repression, cell cycle G1 arrest and cell growth suppression. Mechanistic exploration further revealed that TCP10L is able to stabilize intracellular MAD1 protein level. Consistently, the MAD1-interaction-deficient TCP10L mutant exerts no effect on stabilizing the MAD1 protein. Taken together, our results strongly indicate that TCP10L stabilizes MAD1 protein level through direct interaction, and they cooperatively regulate cell cycle progression.
