      • Immunomodulatory Effects of Self-Assembled RNA Microspheres

        Rationally designed micro-sized RNA particles, called RNA microspheres (RMSs), were generated with single or double strands via rolling circle transcription. The RMSs showed an exceptional stability in the presence of serum, while they were selectively degraded under endolysosomal conditions. With precisely controlled size, both RMSs were successfully taken up by macrophages. Unlike the nature of RNA fragments, RMSs induced only basal-level expression of inflammatory cytokines as well as type I interferon from macrophages, suggesting that RMSs are immunocompatible in the therapeutic dose range. This study could help accelerate clinical translation and broaden the applicability of the self-assembled RNA-based particles without being limited by their potential immunotoxicity, while a systematic controllability to release RNA fragments from RMSs would provide self-assembled RNA-based structures with a great potential for immunomodulation.

        White ginseng extract induces immunomodulatory effects via the MKK4-JNK pathway

        Panax ginseng Meyer (white ginseng) is a popular functional food and its biological effects on the human body have been noted for hundreds of years. In the present study, the underlying mechanisms responsible for the immunomodulatory effects of white ginseng extract (WGE) were investigated. WGE increased NO production via upregulation of iNOS expression levels. Mouse cytokine array results also revealed that the expression of 13 cytokines was elevated by WGE treatment in IFN-γ-primed macrophage cells. Although both MKK4-JNK and MEK-ERK signaling pathways were activated after treatment with WGE, only the MKK4-JNK signaling pathway appears to have any significant immunomodulatory significance. Oral administration of WGE for 28 days recovered cyclophosphamide (CY)-induced suppression of the immune system in mice via the MKK4- JNK pathway. Taken together, these findings suggest that the MKK4-JNK signaling pathway is a crucial mechanism of WGE-induced immunomodulation.

        불가사리 유래 콜라겐 펩타이드의 피부 면역 증진 효과

        불가사리 콜라겐으로부터 저분자 활성 펩타이드를 SDS-PAGE로 분리하고 MALDI-TOF MS를 이용한 분자량 측정을 통해 fraction별로 F1-F5의 5개 시료로 나누어 실험을 수행하였다. 인간 fibroblast를 이용한 세포독성 측정에서 1.0 ㎎/mL 농도의 시료 첨가를 통한 모든 조건에서 20%의 이하의 세포독성을 나타냄에 따라 불가사리 유래 펩타이드가 세포 수준에서 유의할만한 세포독성을 나타내지 않음 확인하였다. 대식세포를 이용한 NO 생성능 측정에서는 시료만 처리했을 경우 무처리 대조군과 비교하여 큰 변화를 관찰 할 수 없었으나, LPS와의 혼합 처리를 통해 F2가 최대 33.8 μM의 NO의 생성량을 나타냄에 따라 시료첨가를 통해 면역 기작이 획기적으로 증진되는 효과를 보였다. 인간 fibroblast인 CCD-986sk의 생육에서 UV 조사를 통한 PGE₂ 발현도 측정 실험에서는 시료첨가를 통해 PGE₂의 발현이 농도 의존적으로 감소되는 결과를 나타내었으며, 세포 증식에 따른 콜라겐 합성능의 비교 결과에서는 시료 첨가를 통해 모든 조건의 콜라겐 생성량이 농도 의존적으로 증가하는 경향을 나타내었다. 특히 F1과 F2가 1.0 ㎎/mL의 농도에서 각각 121.2±3.7%과 127.8±1.1%를 나타내며 양성 대조군에 비해 높은 콜라겐 합성량을 나타내었다. 면역 활성 탐색을 위한 hyaluronidase 저해효과 측정에서도 F2가 535.7 ㎍/mL로 가장 낮은 IC??을 나타내었다. 이상의 실험을 통해 불가사리 유래 콜라겐의 저분자 펩타이드는 세포독성 낮고 NO 생성 촉진 활성, PGE₂ 발현 저해 및 콜라겐의 생성 촉진 효과를 가지는 것을 확인할 수 있었다. 이러한 결과는 불가사리 유래 콜라겐 펩타이드의 기능성 향장 소재로서 활용 가능성을 확인해주는 긍정적인 결과로 향후 추가적인 연구를 통해 불가사리의 향장소재로서의 활용이 기대된다. Low molecular peptides were isolated from Asterias amurensis via SDS-PAGE. The peptides were separated via consecutive gel filtration as five fractions (F1-F5) according to molecular weights, based on the results of MALDITOF MS analysis. The molecular weight of the most active peptide was estimated as 15,000 daltons. The peptide showed cytotoxicity on normal human fibroblast cells at levels as low as 20% when 1.0 ㎎/mL of the samples was added. The peptide also exhibited higher levels of nitric oxide production from macrophages than the lipopolysaccaharides. It was determined that prostaglendin E₂ production was significantly inhibited, up to 127.8% as compared to the control. The low molecular peptide inhibited hyaluronidase activity as 535.7 ㎍/mL of IC??. It can be concluded that the relatively low molecular weight peptide, fucoidan, from A. amurensis has excellent cosmetic and immunomodulatory activities, which can be considered as a possible resource of new cosmetic agents for skin immunomodulation.

        Nonimmunologic targets of immunosuppressive agents in podocytes

        Proteinuria is a characteristic finding in glomerular diseases and is closely associated with renal outcomes. In addition, therapeutic interventions that reduce proteinuria improve renal prognosis. Accumulating evidence has demonstrated that podocytes act as key modulators of glomerular injury and proteinuria. The podocyte, or glomerular visceral epithelial cell, is a highly specialized and differentiated cell that forms interdigitated foot processes with neighboring podocytes, which are bridged together by an extracellular structure known as the “slit diaphragm” (SD). The SD acts as a size- and charge-selective barrier to plasma protein. Derangement of SD structure or loss of SD-associated protein results in podocyte injury and proteinuria. During the past decades, several immunemodulating agents have been used for the treatment of glomerular diseases and for the reduction of proteinuria. Interestingly, recent studies have demonstrated that immunosuppressive agents can have a direct effect on the SD-associated proteins and stabilize actin cytoskeleton in podocyte and have therefore introduced the concept of nonimmunologic mechanism of renoprotection by immunomodulators. This review focuses on the evidence that immuno-modulating agents directly target podocytes.

        Immunomodulation for maxillofacial reconstructive surgery

        Immunomodulation is a technique for the modulation of immune responses against graft material to improve surgical success rates. The main target cell for the immunomodulation is a macrophage because it is the reaction site of the graft and controls the healing process. Macrophages can be classified into M1 and M2 types. Most immunomodulation techniques focus on the rapid differentiation of M2-type macrophage. An M2 inducer, 4-hexylresorcinol, has been recently identified and is used for bone grafts and dental implant coatings.

        Scientific rationale of Indian AYUSH Ministry advisory for COVID-19 prevention, prophylaxis, and immunomodulation

        The current outbreak of COVID-19 is caused by the SARS-CoV-2 virus that has affected > 210 countries. Various steps are taken by different countries to tackle the current war-like health situation. In India, the Ministry of AYUSH released a self-care advisory for immunomodulation measures during the COVID-19 and this review article discusses the detailed scientific rationale associated with this advisory. Authors have spotted and presented in-depth insight of advisory in terms of immunomodulatory, antiviral, antibacterial, co-morbidity associated actions, and their probable mechanism of action. Immunomodulatory actions of advised herbs with no significant adverse drug reaction/toxicity strongly support the extension of advisory for COVID-19 prevention, prophylaxis, mitigations, and rehabilitation capacities. This advisory also emphasized Dhyana (meditation) and Yogasanas as a holistic approach in enhancing immunity, mental health, and quality of life. The present review may open-up new meadows for research and can provide better conceptual leads for future researches in immunomodulation, antiviral-development, psychoneuroimmunology, especially for COVID-19.

        파라바이오틱스의 면역조절능 및 항암효과

        Recently, the term ‘parabiotics’ has reported inactivated microbes with similar health properties of probiotics. Parabiotics can overcome the side effects, risks, safety, and functional limitations of viable probiotics, especially in vulnerable consumers who have difficulty consuming them. Parabiotics produced by heating, radiation, ultraviolet rays, high pressure, and sonication. The main components were known as peptidoglycan, lipopolysaccharides, teichoic acids, and biosurfactants. In addition, parabiotics have been reported the health functionality including immunomodulation, anticancer, antioxidant, neuroprotective effects, and other various functional effects. Therefore, this review summarizes the fundamentals of parabiotics and their representative functional effects, including immunomodulation and anticancer effects, and discusses their potential future applications as health functional materials in the food and pharmaceutical industries.
