        Pomegranate extract on eroded dentin: antioxidant action, bond strength and morphology of the adhesive interface after aging

        Objectives This study aimed to evaluate the effect of pomegranate solution (Punica granatum) on eroded dentin through antioxidant action, shear bond strength (SBS) and interface morphology. Materials and Methods The 10% pomegranate peel extract was prepared by the lyophilization method. Punicalagin polyphenol was confirmed by high-performance liquid chromatography. Antioxidant activity was evaluated by capturing the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical. For the SBS, 48 dentin fragments were divided into sound or eroded, and subdivided according to the pretreatment (n = 12): water or P. granatum. The surfaces were restored with self-etch adhesive and a bulk-fill resin (Ecosite; DMG). The SBS was done immediately (24 hours) and after thermal cycling + water storage (12 months). For scanning electron microscopy, 48 dentin fragments (24 sound and 24 eroded) received the same treatments as for SBS (n = 6), and they were analyzed after 24 hours and 12 months. Results The P. granatum had antioxidant action similar (p = 0.246) to the phenolic standard antioxidants. After 24 hours, eroded dentin had lower SBS than sound dentin (p < 0.001), regardless of the pretreatment. After 12 months, P. granatum maintained the SBS of sound dentin (13.46 ± 3.42 MPa) and eroded dentin (10.96 ± 1.90 MPa) statistically similar. The lowest values were found on eroded dentin treated with water (5.75 ± 1.65 MPa) (p < 0.001). P. granatum on eroded dentin caused peritubular demineralization and hybrid layer with resin tags. Conclusions The pomegranate extract had antioxidant action and preserved the adhesive interface of the eroded dentin.         화피, 석류피, 염부수백피 추출물의 항산화 및 항균 효과

        Objectives : This study was conducted to measure the antioxidative and antimicrobial activities of water- and ethanol-extracts from Betula platyphylla var. japonica, Punica granatum and Rhus javanica against various species of anaerobic bacteria. Methods : In order to evaluate the antioxidative and antimicrobial activities of water- and ethanol-extracts, DPPH radical scavenging and superoxide dismutase (SOD)-like activities were measured in various species such as Staphylococcus epidermidis, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans and Propionibacterium acnes that induce skin inflammation. Also the total amount of phenol in each water- and ethanol-extract was measured to identify its role in the antioxidative and antimicrobial activities of water- and ethanol-extracts from Betula platyphylla var. japonica, Punica granatum and Rhus javanica. The antibacterial activity of the extracts was measured by confirming the lowest concentrations in disk diffusion and minimum inhibitory concentration (MIC) tests. Results : It was observed that RJE (Rhus javanica-EtOH extracts) show the highest content of total phenol. In addition, for RJE the total phenol content was higher from samples taken from domestic sources than from samples taken from foreign sources. DPPH radical scavenging activity was increased by treatment with PGE (Punica granatum-EtOH extracts), RJE and RJW (Rhus javanica-water extracts). It was observed that SOD-like activity was highest in the treatment with PGE. All of the extracts showed antimicrobial activity on S. epidermidis, S. aureus, P. acnes and E. coli, including those from Punica granatum, and it was noted that the activity was higher with RJE than with RJW. Conclusions : These results provide evidence that ethanol extracts of Punica granatum and Rhus javanica may have a beneficial role as antioxidants and antibiotics. Extracts from domestic samples of Betula platyphylla var japonica appeared to have a greater efficacy than extracts from foreign samples.

        A new record of Aphis punicae Passerini, 1863 (Hemiptera: Aphididae) from Korea

        The pomegranate aphid, Aphis punicae Passerini, 1863 (Hemiptera: Aphididae), was firstly collected on Punica granatum L. (Lythraceae) in Korea. For accurate species identification of A. punicae, we compared it with four other pomegranate-feeding Aphis species using molecular and morphological characteristics. In the morphometric analysis, A. punicae is distinguished from Aphis gossypii, which is morphologically similar to A. punicae, by shorter lengths of Ant.IV, Ant.V, and URS and a lower number of hairs on GP. In the sequence analysis, A. punicae reveals genetic differences ranging from 2.1% to 2.4% from A. gossypii in COI gene. In this study, we describe apterous and alate viviparous females of A. punicae and present and identification key to the subgenus Aphis on P. granatum.

      • Report of an Exotic Aphid pest Aphis punicae (Hemiptera:Aphididae) Recently Introduced into Korea

        The exotic aphid pest, pomegranate aphid, Aphis punicae Parsserini, 1863 (Hemiptera: Aphididae), is newly recognized in the Korean peninsula. This species is originally distributed throughout the Mediterranean regions, the Middle East, Ethiopia, India, Pakistan, Indonesia, and Turkey. In the major producers of pomegranate such as India and the Mediterranean regions, this species is regarded as one of the most serious insect pests inflicting injury on pomegranate, Punica granatum Linnaeus. By sucking plant sap and excreting honeydew, aphids cause various plant diseases and virus infections which are more likely to occur as population size increases. Aphis punicae infests together commonly with Aphis gosspii Glover, and rarely with Aphis craccivora Koch and Aphis spiraecola Patch on P. granatum in Korea. In this study, we first compared two closely related species, A. gossypii and A. punicae, based on morphometric analysis to confirm their morphological relationships. Second molecular identification using mitochondrial COI barcode region was performed for the aphid samples collected from P. granatum.

        Biofabrication, characterization, and possible bio-reduction mechanism of platinum nanoparticles mediated by agro-industrial waste and their catalytic activity

        The present study showed biofabrication of platinum nanoparticles (Pt-NPs) using agro-industrial wastePunica granatum’s peel extract. Appearance of the broad spectrum from visible to the ultraviolet region,confirmed the biofabrication of Pt-NPs. Pt-NPs were spherical, within size range of 16–23 nm. XRDsuggested the fabrication of crystalline Pt-NPs with (111) plane in predominant orientation. Thenegative z potential value of colloidal Pt-NPs revealed high stability. FTIR confirmed the role of hydroxyland carbonyl groups of polyphenolic compounds of peel extract for biofabrication. The reduction ofanthropogenic pollutant, 3-nitrophenol, by NaBH4 using colloidal Pt-NPs established it as an efficient‘‘green catalyst.’’

        석곡(石斛), 석류(石榴)의 항산화, 항염증, 주름, 미백에 미치는 영향

        Objective : The aim of this study is to determine the effects of Dendrobii herba extract and Punica granatum extract on skin disease and skin beauty. Methods : To investigate in vitro anti-oxidant activity assay, ethanol extracts of medicinal plants tested by DPPH radical, xanthine oxidase activity. In the next experiment, to investigate anti-inflammatory activity assay, examined by relations in NO synthesis, IL-$1{\beta}$, IL-6, TNF-${\alpha}$, NF-${\kappa}B$, COX-2, MAP kinase. To study Skin wrinkle formation effect, we were examined by tyrosinase activities, melanin synthesis in MNT-1 cell. Results : 1. In an anti-oxidant test, Dendrobii and Punica granatum extract showed high radical scavenging activity. 2. In an anti-inflammatory test, Dendrobii herba and Punica granatum extract weakly inhibited the lipopolysaccharide(LPS)-induced nitric oxide(NO) release from RAW 246.7 macrophage cells. Dendrobii herba and Punica granatum extract also inhibited LPS-induced IL-$1{\beta}$ and COX-2 expressions. The inhibitory effect of Dendrobii herba and Punica granatum extract on macrophage activation were via the inhibition of NF-${\kappa}B$, evidenced by transient transfection assay. however, Dendrobii herba and Punica granatum extract did not have any effects about activation of Jun-N-terminal kinase(JNK) and inhibition of p38 MAP kinase in RAW 264.7 cells. 3. In the skin wrinkle formation assay, Dendrobii herba and Punica granatum extract weakly inhibited collagenase and elastase, however it was not statistically significant. 4. In the skin whitening assay, Dendrobii herba and Punica granatum extract weakly inhibited tyrosinase activity, however, it was not statistically significant. They did not have any effect on melanin synthesis, indicating that they could not be applicable for skin whitening. Conclusion : Dendrobii herba extract and Punica granatum extract may play a significant role in skin disease and skin beauty.

        석류 소비자 구매의식과 구매특성 분석

        In this study, we examined the awareness of consumers purchasing Punica granatum by conducting a survey on con-sumption of Punica granatum for the consumer panel of the Rural Development Administration (RDA) and derived the purchasing characteristics from the actual purchase date analyzed in a Linear regression model and Tobit model. Mostconsumers had been purchasing Punica granatum for health and beauty, and the proportion of that consumers were willingto repurchase Punica granatum was 93.1%. The result of examining the biggest considerations in 5 point scale whenchoosing a Punica granatum was in the order of freshness (4.37)> price (4.15)> safety (4.13)> size(3.86)> brand (3.27)>discount event (2.76). When we compared the results between a linear regression model and tobit model, the signs of all variables are consistent with each other. However, it was estimated that all absolute values of the coefficient valuesin the results of the tobit model analysis were larger than the values in the linear regression model, except for the “favoritepurchasing place” of a weekday traditional markets. Punica granatum is known as a good fruit for postmenopausal womenand it seems that the higher age is, the more purchase there will be. The more income a housewife had, the greaterpurchase there was. In the case of the purchase amount, a selecting for a eating pleasure was bigger than a selecting for a need of health. Therefore, it is necessary to develop Punica granatum with a taste in consumer preferences.

        석류추출물의 항산화와 MMPs 단백질 발현 억제 및 액정 유화물에서의 안정화에 관한 연구

        본 연구에서는 석류추출물의 기능성 활성 검증 및 화장품 개발을 위한 이용 가능성을 확인하였다. 전자공여능을 측정한 결과, 1,000 ㎍/ml의 농도에서 60.6%의 효과를 나타내었고, ABTS 소거능을 측정한 결과, 1,000 ㎍/ml의 농도에서 93.9%의 소거능을 보였다. 또한 elastase 억제 활성과 collagenase 억제 활성은 1,000 ㎍/ml의 농도에서 각각 30%와 47.2%의 억제 활성을 보였다. 석류추출물이 섬유아세포(CCD-986sk)의 증식에 미치는 영향을 조사하기 위해 MTT법으로 세포 생존성을 측정한 결과, 500 ㎍/ml 이하의 농도에서 130% 이상의 생존율이 확인됐다. 석류추출물을 처리한 섬유아세포에서 MMP-1, MMP-2, MMP-3의 단백질 발현은 100 ㎍/ml의 농도에서 각각 23.2%, 81.9%, 69.2%의 저해율을 나타내었다. 위 결과를 바탕으로 석류추출물이 0.1% (w/v)의 농도로 함유된 O/W 액정크림을 제조하여 4, 25, 45, 50℃에서 pH, 시간에 따른 변화, 온도별 안정성 등의 안정성을 조사한 결과, 모두 1개월간 안정성이 있는 것으로 확인되었으며 석류추출물의 화장품 소재로서의 가능성을 확인하였다. This study confirmed the potential of Punica granatum L. extract for functional activity verification and cosmetic development. The electron-donating ability of Punica granatum L. extract was shown 60.6% at a 1,000 ㎍/ml concentration. Its ABTS+ radical scavenging ability was shown 93.9% at a 1,000 ㎍/ml concentration. Additionally, the inhibitive effects of elastase and collagenase inhibition effects were measured as 30% and 47.2%, respectively, at a 1,000 ㎍/ml concentration. To determine the effect of Punica granatum L. extract on the proliferation of fibroblasts (CCD-986sk), cell viability was measured using a 3-[4,5-dimethyl-thiazol-2-yl]-2,5-diphenyl-tetrazoliumbromide (MTT) assay. As a result, survival rates of 130% or higher at a 500 ㎍/ml concentration or less were confirmed. According to the results of Western blot with Punica granatum L. extract, the expression inhibition rates of matrix metalloproteinase-1 (MMP-1), matrix metalloproteinase-2 (MMP-2), and matrix metalloproteinase-3 (MMP-3) were decreased by 23.2%, 81.9%, and 69.2%, respectively, at a 100 μg/ml concentration. Based on the results above, O/W liquid crystal cream with 0.1% Punica granatum L. extract was prepared. The stabilities were tested at 4, 25, 45, and 50℃. By checking the pH, change over time, and stability by temperature, it was confirmed that all were stable for one month. Thus, Punica granatum L. extract shows potential as a natural material for cosmetics.

        Punica granatum Flower 추출물을 함유한 립글로스 제조 및 플럼핑 효과

        Purpose: Lip plumpers are cosmetic agents for producing a temporary increase in lip volume. The topical action of lip plumpers has been rarely discussed in the dermatology literature. This study was conducted to assess the volumizing effect of lip cosmetic products. Methods: We prepared the three different lip glosses: the control (base lip gloss), sample A (2% Zingiber officinale root oil), and sample B (2% Punica granatum flower extract). To assess their efficacy, 30 Korean female subjects (age range 30–49 years) with no negative clinical history of lip conditions were recruited. The impressions of lips were prepared at 0 min, 30 min, and 3 weeks after applying the lip gloss. Scanning electron microscopy was used to scan the impressions. The volume changes in the impressions were assessed by 3D image analysis software. Results: For sample A, the intensity of wrinkles on the lip was decreased at 30 minutes after application, and the volumizing effect was seen at the same time. Both results were statistically significant. However, there was no statistically significant difference in wrinkle relief and volumizing effect 3 weeks later, as compared to the baseline. On the other hand, sample B both decreased wrinkle intensity and showed volumizing effects at 30 min that persisted 3 weeks later. Also, the wrinkle grade was measured by the experts. Conclusion: In conclusion, the sample containing the Punica granatum flower extract had long-term effects in relieving wrinkles and increasing the volume of the lips. These results support the possibility of developing functional cosmetics using Punica granatum flower extract. 목적: 입술 플럼핑은 입술의 볼륨감을 일시적으로 증가시키기 위한 화장품이다. 입술 플럼핑의 국소작용 효과는 피부학 문헌에서거의 논의되지 않았다. 본 연구는 립 화장품의 볼륨감 효과에 대한 평가를 위해 실시되었다. 방법: 컨트롤(베이스 립글로스), 샘플 A (2% 생강 오일), 샘플 B (2% 석류꽃 추출물) 3가지 립글로스를 준비했다. 그 효능을 평가하기 위해 입술에 피부질환이 없는 30명의한국인 여성 피험자(30-49세)를 모집했다. 입술의 모형은 0 min, 30 min, 3주의 시간에 립글로스를 바른 후 제작되었다. SEM은모형을 스캔하는 데 사용되었다. 모형의 볼륨 변화는 3D 이미지 분석 소프트웨어에 의해 평가되었다. 결과: 샘플 A의 경우 바르고30분 후 입술 주름이 감소하였다. 그리고 볼륨감의 효과가 동시에 나타났다. 두 결과 모두 통계적으로 유의미했다. 그러나 기준선에비해 주름 완화 및 볼륨 증가 효과는 3주 만에 통계적으로 차이가 없었다. 반면 샘플 B는 주름 완화 효과와 볼륨 증가 효과가 모두30분대에 나타났고 3주 후에도 지속됐다. 또한 주름 등급은 전문가들에 의해 측정되었다. 그리고, 비슷하게 샘플 B가 입술 주름의강도를 효과적으로 감소시킨다는 것을 발견했다. 결론: 결론적으로 석류꽃 추출물이 함유된 샘플 B는 장기적으로 주름 완화 효과와입술 부피를 증가시키는 효과를 보였다. 이 연구에서는 석류꽃 추출물에 의한 기능성 화장품의 개발 가능성을 제시한다.
