This study investigated the changes in the growth and phenotype of human degenerated intervertebral disc cells depending on the frequency of subculture and the growth factors effects in vitro monolayer culture system. The twice passaged nucleus pulposus (NP) cells were replated six well plates at a density 1×104 cells/cm2. The replated NP cells were divided into four subgroups: no treatment (control group), the treatment of 10 ng/mL transforming growth factor (TGF)-β1 (TGF group), treatment of 5 ng/mL basic fibroblast growth factor (bFGF, FGF group), and treatment of TGF-β plus bFGF (TGF/FGF group). Analyses of proliferation, morphology, histological and reverse transcriptase - polymerase chain reaction (RT-PCR) were made at different time point. The NP cells gradually showed a fibroblast-like spindle shape from their irregular, polygonal shape while undergoing the third time of subculture. The cell proliferation was the highest at the second subculture time. On RT-PCR, the type II collagen expression was gradually decreased and almost unexpressed at the third subculture, but the type I collagen expression was gradually increased in the NP cells. The expression of proteoglycan was gradually decreased in NP cells and it was almost unexpressed at the third time of subculture. FGF can stimulate NP cell proliferation and TGF-β showed increase collagen type II and aggrecan synthesis, and the combination of TGF and FGF appeared both effects of proliferation and extracellular matrix (ECM) synthesis on the NP metabolism. To obtain a great number of cells using a small frequency of subculture and to increase the ECM synthesis, new culture methods based on growth factors or mechanical stimulation must be considered.

Objective: The aim of this study was to investigate the effects of Enteromorpha powder supplementation on reproduction-related hormones and genes in the late laying period of Zi geese. Methods: A total of 312 (1-year-old) Zi geese with similar laying rate were randomly divided into 2 groups with 6 replicates each, each with 21 female geese and 5 male geese. The control group was fed with a basal diet and the test group was fed with a diet containing 3% Enteromorpha powder. The trial period lasted for 7 weeks. Results: Our results showed that the laying rate was improved in the test group at each week of trial (p<0.01), and the levels of estradiol in serum and prolactin in ovary were increased compared with the control group (p<0.05). Conclusion: Based on above results, Enteromorpha powder supplementation at 3% could promote reproductive performance during the late laying period of Zi geese. Objective: Clozapine is one of the most commonly used antipsychotic drugs in China. To date, few studies have investigated the patterns the prescription of clozapine nationwide. The present study examined these patterns in China in 2006 and identified the demographic and clinical characteristics associated with the use of clozapine. Methods: Using a standardized protocol and data collection procedure, we surveyed 5,898 patients with schizophrenia in 10provinces with differing levels of economic development. Results: Overall, clozapine had been prescribed for 31.9% (n=1,883) of the patients; however we found considerable variation among the 10 provinces. The frequency of clozapine use was highest in Sichuan (39.3%) and lowest in Beijing (17.3%). The mean daily dose of clozapine was 210.36±128.72 mg/day, and 25.1% of the patients were treated with clozapine in combination with other antipsychotics. Compared with the group not receiving clozapine, clozapine-user had been treated for longer durations and had experienced a greater number of relapses and hospitalizations. Furthermore, those in the clozapine-user had lower family incomes, were less able to seek psychiatric services, and more likely to be male and have a positive family history of schizophrenia. A multiple logistic regression analysis revealed that age, sex, professional help-seeking behaviors, duration of illness, economic status, educational level, and clinical manifestations were associated with the use of clozapine. Conclusion: Clozapine use is common in China. However, use of the antipsychotic varies among provinces, and demographic and clinical factors play important roles in the prescription of clozapine.
