We screened four B. thuringiensis strains whose parasporal inclusions contained the S-layer protein (SLP), and cloned two slp genes from each strain. Phylogenetic analysis indicated these SLPs could be divided into two groups, SLP1s and SLP2s. To confirm whether SLPs were present in the S-layer or as a parasporal inclusion, strains CTC and BMB1152 were chosen for further study. Western blots with isolated S-layer proteins from strains CTC and BMB1152 in the vegetative phase showed that SLP1s and SLP2s were constituents of the S-layer. Immunofluorescence utilizing spore-inclusion mixtures of strains CTC and BMB1152 in the sporulation phase showed that SLP1s and SLP2s were also constituents of parasporal inclusions. When heterogeneously expressed in the crystal negative strain BMB171, four SLPs from strains CTC and BMB1152 could also form parasporal inclusions. This temporal and spatial expression is not an occasional phenomenon but ubiquitous in B. thuringiensis strains. [BMB reports 2011; 44(5): 323-328]

Oriental orchids of the genus Cymbidium are famous for their flower fragrance and have great value in the horticultural market. To investigate the regulatory mechanisms of fragrance biosynthesis in Cymbidium faberi, the promoters of the CfAOC and CfJMT genes in the methyl jasmonate biosynthesis pathway were cloned and analyzed. Notably, the CfAOC promoter was transcriptionally active in all vegetative organs, which was not true for the CfJMT promoter, indicating that the latter is tissue-specific for the flowers of C. faberi. The dual-luciferase assay showed that the fragment between –629 and –1038 nt of the CfAOC promoter, as well as the fragment between –960 and –1121 nt of the CfJMT promoter, displayed specific interaction with the CfbHLH transcription factors. These results provide a theoretical basis for the genetic modification of the flower fragrance of Cymbidium based on specific CfbHLH transcription factors.

Background Non-voltage-gated sodium channel, also known as the epithelial sodium channel (ENaC), formed by heteromeric complexes consisting of SCNN1A, SCNN1B, and SCNN1G, is responsible for maintaining sodium ion and body fluid homeostasis in epithelial cells. However, no systematic study of SCNN1 family members has been conducted in renal clear cell carcinoma (ccRCC) to date. Objective To investigate the abnormal expression of SCNN1 family in ccRCC and its potential correlation with clinical parameters. Methods The transcription and protein expression levels of SCNN1 family members in ccRCC were analyzed based on the TCGA database, and were confirmed by quantitative RT-PCR and immunohistochemical staining assays, respectively. The area under curve (AUC) was used to evaluate the diagnostic value of SCNN1 family members for ccRCC patients. Results The mRNA and protein expression of SCNN1 family members was significantly downregulated in ccRCC compared with normal kidney tissues, which might be due to DNA hypermethylation in the promoter region. It is worth noting that the AUC of SCNN1A, SCNN1B, and SCNN1G were 0.965, 0.979, and 0.988 based on the TCGA database (p < 0.0001), respectively. The diagnostic value was even higher when combing these three members together (AUC = 0.997, p < 0.0001). Intriguingly, the mRNA level of SCNN1A was significantly lower in females compared with males, while SCNN1B and SCNN1G were increased with the progression of ccRCC and remarkably associated with a worse outcome for patients. Conclusion The aberrantly decrease of SCNN1 family members might serve as valuable biomarkers for the diagnosis of ccRCC.
Clinical Significance of Expression and Amplification of the DcR3 Gene in Pancreatic Carcinomas

This study aimed to investigate the clinical significance of expression and amplification of decoy receptor 3 (DcR3) in pancreatic carcinomas (PC). mRNA expression was detected by PQ-PCR, and amplification was determined. DcR3 protein expression was detected by immunohistochemistry and ELISA. Correlations between DcR3 expression and clinical pathological factors were analyzed. The relative amount of DcR3 in PC tissues and non-cancerous tissues showed a statistically significant difference, 21 cases displaying more than two fold DcR3 amplification, while no such amplification was found in normal pancreatic tissues. DcR3 positive cell staining was located in the cytoplasm. The positive rate of DcR3 in PC and non-cancerous tissues showed a significant difference. DcR3 mRNA expression was correlated with clinical staging, size of the tumor, lymph node metastasis and histological staging, while protein expression was correlated with clinical data like tumor size. DcR3 gene amplification only correlated with tumor size. The level of DcR3 in serum of the PC resectable group before operation was $72.2{\pm}10.2$ pg/ml, showing a significant difference compared to gallbladder carcinoma group (GC) or pancreatic benign tumor (PBT) group (P < 0.01). In conclusion, DcR3 amplification is correlated with DcR3 expression in PC tissues, especially those clinical pathological factors which reflect tumor progression. Assessment of DcR3 level in sera of PC patients may be helpful for the early diagnosis and prognostic judgement.
Residual behavior of SRRAC beam and column after exposure to high temperatures

Composite effect between steel and recycled aggregate concrete (RAC) in steel reinforced-RAC (SRRAC) structures can effectively improve RAC’s adverse mechanical properties due to the natural defects of recycled coarse aggregate (RCA). However, the performance of SRRAC after thermal exposure will have a great impact on the safety of the structure. In this paper, firstly, the mechanical properties of SRRAC structures after high temperatures exposure were tested, including 24 SRRAC columns and 32 SRRAC beams. Then, the change rules of beams and columns performance with the maximum temperature and replacement percentage were compared. Finally, the formulas to evaluate the residual bearing capacity of SRRAC beams and columns after exposure to high temperatures were established. The experimental results show that the maximum exposure temperature can be judged by the apparent phenomenon and mass loss ratio of RAC. After high temperatures exposure, the mechanical properties of SRRAC beams and columns change significantly, where the degradation of bearing capacity and stiffness is the most obvious. Moreover, it is found that the degradation degree of compression member is more serious than that of flexural member. The formulas of residual bearing capacity established by introducing influence coefficient of material strength agree well with the experimental results.
