        6-(3.4-Dichlorophenyl)amino-7-chloro-5.8-quinolinedione (RCK50) was tested for antifungal activities in mice systemically infected with Candida albicans. The therapeutic potential of RCK50 was also assessed in comparison with ketoconazole. RCK50 had ED_50 0.22±0.01 ㎎/㎏. Ketoconazole as a positive control had ED_50 6.00±1.70 ㎎/㎏. Intraperitoneally administered RCK50 at the ED_50 for 7 days and 14 days reduced Candida albicans colony count in the kidneys and liver. And administered RCK50 at the ED_50 for 14 days improved survival rates. The genotoxicities of RCK50 had been evaluated. RCK50 was negative in Ames test with Salmonella typhimurium and chromosomal aberration test in CHL cells. RCK50 did not show any clastogenic effect in mouse peripheral blood and was negative in mouse micronucleus assay. These results indicate that RCK50 has no genotoxic potential under these experimental conditions. Acute oral toxicity studies of RCK50 were carried out in ICR mice of both sexes. RCK50 did not show acute oral toxicities and LD_50 values were over 2.850 ㎎/㎏ in ICR mice.

      • 백삼에 있어 수침처리가 체형 및 사포닌 함량에 미치는 영향

        본 연구는 수침 처리가 백삼제품의 원형복원에 미치는 영향을 조사하여 백삼제품 제조에 사용한 원료수삼의 품질을 평가하기 위한 기초자료를 얻고자 수행하였던 바 결과를 요약하면 다음과 같다. 백삼의 수침처리는 원료수삼과 같은 수준으로 원형을 복원할 수 있어 제품에 사용된 원료수삼의 뇌두의 경흔적, 동체부위의 피해정도 그리고 내부조직의 나이테 관찰이 용이하여 연근판별 및 품질평가가 가능하였다. 백삼제품의 ginsenoside 함량은 제조회사에 따라 다소 차이가 있었으며 panaxadiol계 사포닌은 ginsenoside-Rb₁이 많고, panaxatriol계 사포닌은 ginsenoside-Rg₁이 높게 나타났다. 그리고 수침에 의한 백삼의ginsenoside 함량은 수침온도가 증가할수록 감소하는 경향이었다. This study was conducted to investigate the effects of soaking treatment in white ginseng and evaluated quality evaluation of raw ginseng and ginsenoside. Restoration to the original state of the raw ginseng can be achieved by soaking treatment in white ginseng. Stem vestige at rhizome and damage of main root of the swelled white ginseng was examined by eyes and annual rings by microscope easily. Soaking treatment made it easy to be examined the identification of root age and quality evaluation of white ginseng. The content of ginsenoside showed difference among the three products. The content of ginsenoside-Rb1 was highest among the constituents of protopanaxadiol saponin and the content of ginsenoside-Rg1 among that of protopanaxatriol saponin. The higher the soaking temperature was, the lower the content of ginsenoside in the white ginseng was.

      • 백삼에 있어 수침처리가 원형복원에 미치는 영향

        본 연구는 시중에서 거래되고 있는 백삼을 구입하여 수침 처리가 백삼의 원형복원 및 사포닌 성분에 미치는 영향을 구명하고자 수행하였다. 시판백삼은 같은 등급 및 편급의 제품이었으나 뇌두, 근장, 근직경 등 조사 형질에서 제품간 차이가 있었다. 그리고 수침처리에 의해 근중, 동체, 근직경, 근장 등은 증가하여 원료삼으로서 수삼상태를 확인할 수 있었다. 또한 이들 백삼은 온도가 증가할수록 체적이 빨리 증가하여 온도에 의한 영향을 크게 받았으며 다시 건조하여 본래 백삼의 상태로 환원하는 것이 가능하였다. 그리고 수침에 의한 근중의 변화는 2배 이상 증가하였는데 근중이 최고도에 달하는 시간은 대략 24시간 후 이었다. This study was conducted to investigate the effects of soaking treatment in white ginseng and evaluated white ginseng on the restoration to the original shape and the quality of raw ginseng. KW-1 white ginseng had longer rhizome length and main root length. KW-2 had larger rhizome diameter, main root diameter and root weight. KW-3 showed lower quality than the other two white ginseng products. Soaking treatment made the root weight, main root diameter and main root length of raw ginseng increased. The decrease of root weight was significant by drying. The effects of soaking and drying treatment on the characters investigated of the raw ginseng and white ginseng showed same tendency. The higher the soaking temperature was, the faster the increasing rate was. The increasing of white ginseng stopped after around 12 hours from soaking at any soaking temperature.

      • Candida albicans의 형태변환 조절과정 규명의 최근 연구 동향

        The opportunistic fungal pathogen Candida albicans can grow as yeast, pseudohyphae or true hyphae, and morphology of each cell is essential for efficient dissemination and tissue invasion. Therefore the ability to switch in C. albicans is thought to be an important virulence factor. The morphological conversion of C. albicans can be triggered in vitro by a various environmental stimuli including serum, pH, temperature, and nutrient depletion. These inducers regulate the expression of hypha-specific genes, which are involved in morphogenesis, through multiple signaling pathways such as MAPK pathway and cAMP/PKA pathway. In order to investigate Signal transduction pathways, disruption or over-expression of the component gene(s) were performed and then change(s) in morphological transition as well as the expression of hypha-specific genes are observed in general. As the approach to unravel new roles of already-known signaling pathways in fungal pathogenicity, responses toward external stimuli such as hypoxic condition and iron utilization are performed these days. The new approaches revealed that the morphological conversion in C. albicans is resulted from the consorted action of multiple signaling pathways rather than from a single pathway. Consequently, the existence of a novel Signaling pathway for dimorphic switch in C. albicans could not be excluded. In this context, it is noteworthy that the involvement of LAMMER kinase in fungal development; LAMMER kinase is known to be involved in oxidative-stress response, filamentous growth, and flocculation in fission yeast; S. cerevisiae LAMMER kinase, ScKNS1, is also reported to phosphorylate Slt2, MAPK of cell wall integrity pathway, and to interact with dual-specificity phosphatase Sdp1, which regulates the activity of Slt2. Therefore, studies on the function of LAMMER kinase in C. albicans will be helpful to reveal novel regulatory mechanisms for virulence in pathogenic fungi.

      • CW Laser Annealing 과정중 SOS 상의 시간적 온도변화 측정에 관한 연구

        The purpose of this experiment is to demonstrate the new method of TSRTM during CW Ar laser annealing using a He-Ne laser probe beam. Optical interference phenomena by a small changes of refractive index of Si, which is induced by the temperature change during short time laser annealing on the Si wafer surface, was used. The samples, SOS(Silicon on Sapphire), used in this work were epi. <100> Si grown on a <1012> insulator sapphire substrate with 2.0μm thickness. From the experimental temperature dependent equation of the refraction of index of Si with respect to the probe beam(λ=6328 Å) in furnace experiment, n(T)=3.98+7×10-4T (℃), the temperature on the destructtve or constructive interference was calculated. Incident Ar laser beam whose power ranges from 1.8W to 2.8W was irradiated and the effect of temperature rise of Si wafer on the elliptical spot whose major and minor axis is 200μm, 15μm respectively is investigated by the He-Ne laser probe beam which is focused 30μm. The annealing time ranges from 8 msec~250 msec to 500 msec~20 sec was adopted to estimate that the first constructive interference is occurred at least total irradiated power density 5.46×102Jㆍsec/cm2, and it is inferred that more than 180msec is needed for the first constructive interference when the total incident energy is 2.8 W. For the longtime annealing, about more than 1 sec, the interference pattern was coincided with the result produced by the furnace experiment. It is investigated from the space-resolved temperature gradient that the irradiated laser beam was focused in center.

        Platinum sensitive 재발성 난소암에서 Carboplatin/Paclitaxel의 효과

        목적 : 재발성 난소암에서 platinum 감수성을 기준으로 2차 항암요법으로 사용한 Carboplatin과 Paclitaxel의 효과를 알아보기 위하여 본 연구를 진행하였다. 연구방법 : 1998년 1월 1일부터 2003년 8월 31일까지 만 5년 8개월 간 아주대학교병원 산부인과에 재발성 난소암으로 입원한 환자 중에서 최초 수술로 난소암을 확진한 후 Cisplatin 또는 Carboplatin 등 platinum 제재가 포함된 항암치료를 받은 후 6개월 이후에 재발한 환자 29명을 대상으로 Carboplatin은 5 AUC를 그리고 Paclitaxel은 175 mg/m^(2)를 투여하였으며, 최소 3회에서 최고 16회까지 투여하였다. 치료효과의 판정은 CT, MRI, Ultrasonography등 영상진단상 암종괴의 크기가 완전히 소실되었을 경우를 완전반응(complete response)으로, 50%이상 감소되었을 경우를 부분반응(partial response)으로, 50% 미만으로 감소되었거나 20% 미만 증가하였을 경우를 불변(stable disease)으로, 그리고 20% 이상 증가되었을 경우를 진행(progressive disease)로 정의하였다. 결과 : 대상 환자에서 완전반응은 6명(21%), 부분반응은 11명(38%), 그리고 불변은 5명(17%)로 전체 반응율 (overall response rate)은 59% 그리고 임상적 유용성(clinical benefit=CR+PR+SD)은 76%이었다. 결론 : 재발성 난소암환자에서 기왕에 사용한 Platinum 제재에 민감한 환자에서는 Carboplatin과 Paclitaxel의 병행 투여가 효과적이었다. Objective : To ascertain the effect of Carboplatin and Paclitaxel combination as a second line regimen for recurrent ovarian carcinoma based on platinum sensitivity. Methods : From January 1st, 1998 through to August 31st, 2003, 29 patients were retrospectively reviewed who had previously underwent primary cytoreductive surgery and adjuvant chemotherapy with Cisplatin or Carboplatin for ovarian carcinoma, and who had later recurred during follow up. All 29 patients had recurred at least 6 months after therapy, and thereafter received second line chemotherapy consisting of Carboplatin (5 AUC) and Paclitaxel (175 mg/㎡) for a minimum of least 3 cycles and a maximum of 16 cycles. Treatment efficacy was assessed by imaging studies including Computerized tomography, Magnetic Resonance Imaging, or Ultrasonography. Complete response was determined as complete disappearance of tumor, partial response at least than 50% reduction in tumor volume, stable disease as less than 50% reduction or less than 20% increase in tumor volume, and progressive disease as more than 20% increase. Results : There was complete response in 6 (21%) patients, partial response in 11 (38%), stable disease in 5 (17%), showing an overall response rate of 59%. The clinical benefit (CR+PR+SD) was 76%. Conclusion : The combination of Carboplatin and Paclitaxel administration seems to be an effective mode of second line therapy in patients with recurrent ovarian carcinoma who had been sensitive to prior platinum based chemotherapy.

      • 주철과 오스테나이트계 스테인리스강과의 확산접합에 관한 연구

        Effects of the diffusion bonding on the mechanical properties for the GCD45 /austenitic stainless steel(STS304) bonding specimens have been investigated. The results are obtained as follows ; 1) In the case of GCD45/STS304 specimen made on air, the bonding tensile strength increased with increasing bonding temperature. The maximun bonding tensile strength was 467.5MPa at 1373K. 2) The bonding tensile strength of GCD45/STS304 made by using insert metal MBF35 increased with increasing bonding time, and was about 470MPa at the conditions of bonding temperature 1323K, bonding time 1.8Ks and bonding pressure 9.8MPa. 3) The bonding tensile strength of GCD45/STS304 made by using the insert metal was higher than that of direct bonding specimen.
