Flammulina velutipe의 국내 균주와 외래 균주 간의 ITS region을 이용한 계통학적 유연관계 분석

본 연구는 팽이버섯(Flammulina velutipes)의 국내 균주와 외래 균주간의 유전적 유연관계를 조사하기 위하여 수행되었으며, 계통수 측정 결과 3개 그룹으로 나눠짐을 확인하였다. 20개 F. velutipes 균주들의 ITS region 염기서열을 확보하였으며, 이들 서열을 바탕으로 multiple alignment를 보았으며, Neighbor-joining method를 이용하여 계통수를 작성하였으며, 2개 그룹으로 나눠짐을 확인하였다. 또한 F. velutipes 4154 균주의 rDNA-cluster를 최초로 분석한 결과, 총 10,974 bp임을 밝혔다. SSU는 1,806 bp, ITS region은 553 bp의 염기배열이 결정되었다. ITS 1 부분은 245 bp이고 ITS 2는 308 bp였으며, LSU에 해당되는 염기서열은 3,402 bp, IGS 1은 1,400 bp, 5S는 83 bp, IGS 2는 3,571 bp임을 확인하였다. This study was carried out to investigate the genetic relationship of Flammulina velutipes with other species. The ribosomal DNA cluster containing 4 rRNA genes from F. velutipes 4154 were sequenced. The length of the rDNA cluster sequence was estimated at 7,403 bp long and consisted of 1,806 bp of SSU rDNA, 245 bp of ITS 1 region, 159 bp of 5.8S rDNA, 308 bp of ITS 2 region, 3,402 bp of LSU rDNA, 1,400 bp of IGS 1 region, and 83 bp of 5S rDNA. The F. velutipes 4154 genes were contained in the rDNA cluster of F. velutipes in the order of SSU rDNA - ITS 1 - 5.8S rDNA - ITS 2 - LSU rDNA - IGS 1 - 5S rDNA. The phylogenetic relationships of 20 strains of Tricholomataceae and Physalacriaceae were analyzed by conducting distance analysis using the Neighbor-joining (NJ) method. The 20 strains used in this study were divided into three groups and the strains of the genus Flammulina were related very closely to strains of Physalacria bambusae.

많은 제조 또는 서비스 산업에서는 각 주문에 대해 계약에 의해 허용될 수 있는 최대의 납기지연 일자가 존재하며, 이에 따라 주문자는 허용된 납기지연 일자가 초과된 주문은 취소 할 수 있다. 본 논문에서는 총 가중 납기지연 최소화를 위한 단일기계 일정계획문제를 다루며, 납기가 지연된 주문들에 대해 두 가지 종류의 벌칙금을 고려한다. 첫 번째는 허용한도 내에서 지연된 주문에 대한 납기지연 벌칙금이며, 두 번째는 허용한도를 초과한 납기지연에 의해 발생한 주문취소에 대한 벌칙금이다. 본 연구에서는 기존의 총 납기지연 최소화 단일기계 일정계획문제를 위해 개발된 우선순위규칙을 확장하여 납기지연 한도가 있는 상황에 적합한 우선순위규칙들을 제시하도록 한다.
Regional Distribution of Interstitial Cells of Cajal, (ICC) in Human Stomach

We elucidated the distribution of interstitial cells of Cajal (ICC) in human stomach, using cryosection and $c-Kit$ immunohistochemistry to identify $c-Kit$ positive ICC. Before $c-Kit$ staining, we routinely used hematoxylin and eosin (HE) staining to identify every structure of human stomach, from mucosa to longitudinal muscle. HE staining revealed that the fundus greater curvature (GC) had prominent oblique muscle layer, and $c-Kit$ immunostaining $c-Kit$ positive ICC cells were found to have typical morphology of dense fusiform cell body with multiple processes protruding from the central cell body. In particular, we could observe dense processes and ramifications of ICC in myenteric area and longitudinal muscle layer of corpus GC. Interestingly, $c-Kit$ positive ICC-like cells which had morphology very similar to ICC were found in gastric mucosa. We could not find any significant difference in the distribution of ICC between fundus and corpus, except for submucosa where the density of ICC was much higher in gastric fundus than corpus. Furthermore, there was no significant difference in the density of ICC between each area of fundus and corpus, except for muscularis mucosa. Finally, we also found similar distribution of ICC in normal and cancerous tissue obtained from a patient who underwent pancreotomy and gastrectomy. In conclusion, ICC was found ubiquitously in human stomach and the density of ICC was significantly lower in the muscularis mucosa of both fundus/corpus and higher in the submucosa of gastric fundus than corpus.

The effect of cathode flow pulsation on the performance enhancement of a 10-cell proton-exchange membrane fuel cell is investigated. In the case of cathode pulsating flow with mean flow, the fuel cell power output and the limiting current density dramatically increase as pulsating frequency increases at given pulsating amplitude, while the fuel cell efficiency slightly decreases. This performance enhancement by cathode pulsating flow is more distinct at low cathode mean flow rates. This result is contributed that the pulsating flow enhances the dispersion inside the cathode channels, and then improving the oxygen and temperature distributions. In the case of cathode pulsating flow without mean flow, the fuel cell stack is operated despite cathode mean flow is absent. The limiting current density is extended as the pulsating frequency and swept distance (amplitude) increase. When the pulsating frequency and swept distance are 2.38Hz and 13.65mm respectively, the fuel cell performance is equal to that the cathode mean flow rate is 1.29 lpm. Also, the case of pulsating flow is more stable at the concentration loss region than the case of non-pulsating flow for the same performance conditions.

Licochalcone (LC), a major phenolic retrochalcone from licorice, has anti-inflammatory activity. This study investigated the effects of licochalcone A (LCA) and licochalcone E (LCE) on Liver X receptor-α (LXRα)-mediated lipogenic gene expression and the molecular mechanisms underlying those effects. LCA and LCE antagonized the ability of LXRα agonists (T0901317 or GW3965) to increase sterol regulatory element binding protein-1c (SREBP-1c) expression and thereby inhibited target gene expression (e.g., FAS and ACC) in HepG2 cells. Moreover, treatment with LCA and LCE impaired LXRα/RXRα-induced CYP7A1-LXRE-luciferase (CYP7A1) transactivation. The AMPK-Sirt1 signaling pathway is an important regulator of energy metabolism and, therefore, a potential therapeutic target for metabolic diseases, including hepatic steatosis. We found here that LCE increased AMPK phosphorylation and Sirt1 expression. We conclude that LC inhibits SREBP-1c-mediated hepatic lipogenesis via activation of the AMPK/Sirt1 signaling pathway.
