        Clinical Prognostic Factors in 86 Chinese Patients with Primary Myelodysplastic Syndromes and Trisomy 8: A Single Institution Experience

        Purpose: The objective was to determine the characteristics and prognostic factors of 86 Chinese patients with trisomy 8 aberrationsand compare the prognostic value of International Prognostic System (IPSS) and Revised IPSS (IPSS-R) in this cohort. Materials and Methods: A total of 86 cases diagnosed with primary myelodysplastic syndromes (MDS) with isolated tr8 or with tr8 and other additional cytogenetic aberrations diagnosed and treated at the Union Hospital, Tongji Medical College of Huazhong University of Science and Technology between July 2002 and March 2013 were reviewed. Results: The median survival of the entire group was 23.0 months, and acute myeloid leukemia (AML) developed in 43% (37/86) patients within the follow up time. The univariate analysis revealed that overall survival (OS) was correlated with age, thrombocytopenia,absolute neutrophil count, marrow blasts, cytogenetic status and red blood cell transfusion at diagnosis, and the multivariateanalysis revealed that age, marrow blasts, cytogenetic status and transfusion dependence were independent parameters for the OS. The cytogenetic complexity and marrow blasts had the strongest impact on the AML transformation by multivariate analysis. Comparing the two prognostic systems, both two systems could successfully discriminate risk groups for survival. IPSS-R was more refined than IPSS for predicting OS, but had no advantage in predicting the risk of AML development. Conclusion: This study confirmed the influence of clinical factors on the prognosis of 86 Chinese MDS patients with trisomy 8. In addition, IPSS-R can further refine prognostic discrimination in the IPSS risk categories.

        Effect of Kai Xin San on Learning and Memory in a Rat Model of Paradoxical Sleep Deprivation

        The present study aimed to evaluate the effect of kai xin san (KXS, at doses of 500, 250, and 125 mg/kg body weight per day), a well-known traditional Chinese medicine, on learning and memory in paradoxical sleep deprivation (PSD)-induced cognition deficit rats. Two behavior tests (the Open Field test and the Morris water maze task) were used for testing the effects of KXS on a PSD-induced learning and memory deficit model. Furthermore, its effect on the glutamic acid (GLU) and γ-amino-butyric acid (GABA) levels in the brain tissue, brain-derived neurotrophic factor (BDNF), cyclic AMP response element binding protein (CREB), and phosphorylated-CREB (p-CREB) expression in the hippocampus was also tested. KXS exerted the greatest cognition against the 48 h PSD-induced cognitive deficit and these effects may be mediated by decreasing the GLU and GABA levels and increasing the levels of BDNF, CREB, and p-CREB. This study indicates that the effect of KXS on learning and memory in a rat model of PSD could be associated with the modulation of neurotransmitter levels and the expression of some genes in the brain that contribute to memory functions.

        Developing TCM clinical practice guidelines: A comparison between traditional Chinese medicine and western medicine

        Clinical practice guidelines in Traditional Chinese Medicine (CPG-TCM) is the recommendation that aims to provide the best service to users by identifying and summarizing the rules of prevention, diagnosis, treatment, rehabilitation, and regression of diseases based on systematic reviews of evidence and balancing the advantages and disadvantages of different interventions for clinical questions of Traditional Chinese Medicine. Over the past 30 years, the concept and methods of evidence-based medicine have had a significant impact on the development of clinical practice guidelines in Western Medicine (CPG-WM), and their standardized guideline development methods are being adapted and used in the development of CPG-TCM. However, the quality of CPG-TCM is far from CPG-WM, and the methodological system for developing CPG-TCM is not yet fully established. Therefore, this study aims to explore the methodological differences between CPG-TCM and CPG-WM and to inform the development of high-quality CPGTCM.

        Effects of Exogenous Organic Acids on the Growth and Antioxidant System of Cosmos bipinnatus under Cadmium Stress

        Cadmium pollution in China’s urban green spaces is extremely severe, causing a significant increase in the risk of urban environmental pollution. Applying organic acids under Cd stress conditions improves the activity of the major antioxidant enzymes in the AsA-GSH cycle signi- ficantly. In this study, Cosmos bipinnatus seedlings were cultured in a nutritive medium containing 40 µmol·L-1 Cd. They were subsequently supplemented with different concentrations of citric acid and malic acid (0, 0.5, 1.0, 2.0 mmol L-1 ) to analyze the roles of the organic acids in regulating the AsA-GSH cycle involved in plant resistance to Cd stress. Exogenous organic acids inhibited the growth of C. bipinnatus in a dose-dependent manner. AsA, GSH, and GR in the leaves decreased dramatically at high malic acid concentrations (2 mol L-1 ). The contents of GSH and GR and the APX activities showed a downward trend when the seedlings were treated with 0.5–2 mmol·L-1 of citric acid. However, AsA exhibited contrasting results under these conditions. Notably, both the antioxidant and enzyme activities were reduced considerably to a minimum when the citric acid concentration was 2 mmol·L-1 . Low concentrations of exogenous acids increased the activities of APX and GR in the leaves of C. bipinnatus, thus increasing the contents of ASA and GSH. Such increases boost the efficiency of the ASA-GSH cycle in relieving the toxic effect of cadmium on C. bipinnatus.         Diversity and Active Mechanism of Fengycin-Type Cyclopeptides from Bacillus subtilis XF-1 Against Plasmodiophora brassicae

        Bacillus subtilis XF-1, a strain with demonstrated ability to control clubroot disease caused by Plasmodiophora brassicae, was studied to elucidate its mechanism of antifungal activity against P. brassicae. Fengycin-type cyclopeptides (FTCPs), a well-known class of compounds with strong fungitoxic activity, were purified by acid precipitation, methanol extraction, and chromatographic separation. Eight homologs of fengycin, seven homologs of dehydroxyfengycin, and six unknown FTCPs were characterized with LC/ESI-MS, LC/ESI-MS/MS, and NMR. FTCPs (250μg/ml) were used to treat the resting spores of P. brassicae (107/ml) by detecting leakage of the cytoplasm components and cell destruction. After 12 h treatment, the absorbencies at 260 nm (A260) and at 280 nm (A280) increased gradually to approaching the maximum of absorbance, accompanying the collapse of P. brassicae resting spores, and nearly no complete cells were observed at 24 h treatment. The results suggested that the cells could be cleaved by the FTCPs of B. subtilis XF-1, and the diversity of FTCPs was mainly attributed to a mechanism of clubroot disease biocontrol.

        Phosphorylation of DYNLT1 at Serine 82 Regulates Microtubule Stability and Mitochondrial Permeabilization in Hypoxia

        Hypoxia-induced microtubule disruption and mitochondrial permeability transition (mPT) are crucial events leading to fatal cell damage and recent studies showed that microtubules (MTs) are involved in the modulation of mitochondrial function. Dynein light chain Tctex-type 1 (DYNLT1) is thought to be associated with MTs and mitochondria. Previously we demonstrated that DYNLT1 knockdown aggravates hypoxia-induced mitochondrial permeabilization, which indicates a role of DYNLT1 in hypoxic cytoprotection. But the underlying regulatory mechanism of DYNLT1 remains illusive. Here we aimed to investigate the phosphorylation alteration of DYNLT1 at serine 82 (S82) in hypoxia (1% O2). We therefore constructed recombinant adenoviruses to generate S82E and S82A mutants, used to transfect H9c2 and HeLa cell lines. Development of hypoxia-induced mPT (MMP examining, Cyt c release and mPT pore opening assay), hypoxic energy metabolism (cellular viability and ATP quantification), and stability of MTs were examined. Our results showed that phosph-S82 (S82-P) expression was increased in early hypoxia; S82E mutation (phosphomimic) aggravated mitochondrial damage, ele-vated the free tubulin in cytoplasm and decreased the cellular viability; S82A mutation (dephosphomimic) seemed to diminish the hypoxia-induced injury. These data suggest that DYNLT1 phosphorylation at S82 is involved in MTs and mitochondria regulation, and their interaction and cooperation contribute to the cellular hypoxic tolerance. Thus, we provide new insights into a DYNLT1 mechanism in stabilizing MTs and mitochondria, and propose a potential therapeutic target for hypoxia cytoprotective studies.

        Ketamine induces neuronal apoptosis and cognitive disorder via miR-199a-5p/HIF-1α in neonatal rats

        It reported that repeated administration of clinical doses of ketamine could induce neuronal apoptosis in the immature brain. In this study, SpragueDawley postnatal day 7 rats were treated with ketamine. As the rats grew, we found ketamine impaired the cognitive function of the rats. Ketamine increased HIF1α level in the brain tissues of rats as compared to the control group and YC-1 rescued those effects. The escape latency and the platform crossing time of Morris water maze decreased in the YC-1-treated rats compared to the control in ketamine-treated rats. We also found that ketamine decreased the expression of miR199a-5p and HIF-1α is a directly target of miR-199a5p. Administration of ketamine also decrease the cell activity while YC-1 and MiR-199a-5p mimics rescued the inhibition effect. In conclusion, we suggest that ketamine-induced neuronal apoptosis in neonatal rats, followed by learning and memory impairment, might be mediated via the miR-199a-5p/HIF-1α signalling pathway.
