        Phosphorylation of DYNLT1 at Serine 82 Regulates Microtubule Stability and Mitochondrial Permeabilization in Hypoxia

        Hypoxia-induced microtubule disruption and mitochondrial permeability transition (mPT) are crucial events leading to fatal cell damage and recent studies showed that microtubules (MTs) are involved in the modulation of mitochondrial function. Dynein light chain Tctex-type 1 (DYNLT1) is thought to be associated with MTs and mitochondria. Previously we demonstrated that DYNLT1 knockdown aggravates hypoxia-induced mitochondrial permeabilization, which indicates a role of DYNLT1 in hypoxic cytoprotection. But the underlying regulatory mechanism of DYNLT1 remains illusive. Here we aimed to investigate the phosphorylation alteration of DYNLT1 at serine 82 (S82) in hypoxia (1% O2). We therefore constructed recombinant adenoviruses to generate S82E and S82A mutants, used to transfect H9c2 and HeLa cell lines. Development of hypoxia-induced mPT (MMP examining, Cyt c release and mPT pore opening assay), hypoxic energy metabolism (cellular viability and ATP quantification), and stability of MTs were examined. Our results showed that phosph-S82 (S82-P) expression was increased in early hypoxia; S82E mutation (phosphomimic) aggravated mitochondrial damage, ele-vated the free tubulin in cytoplasm and decreased the cellular viability; S82A mutation (dephosphomimic) seemed to diminish the hypoxia-induced injury. These data suggest that DYNLT1 phosphorylation at S82 is involved in MTs and mitochondria regulation, and their interaction and cooperation contribute to the cellular hypoxic tolerance. Thus, we provide new insights into a DYNLT1 mechanism in stabilizing MTs and mitochondria, and propose a potential therapeutic target for hypoxia cytoprotective studies.

        Pratensein glycoside attenuates respiratory syncytial virus infection-induced oxidative and inflammatory injury via TGF-β signaling pathway

        Background Respiratory syncytial virus (RSV)-induced oxidative stress and inflammation are the leading causes of many severe diseases, especially in infants and children. Although many studies focused on the mechanism during RSV infection, an effective therapy is still urgently needed. Objectives In our study, we examined the function of pratensein glycoside (PG) in the progression of RSV-induced infection in vitro. Results PG treatment significantly inhibited RSV-induced oxidative stress and inflammation in a dose-dependent manner. We found that 40 μM of PG had no signifi cant cytotoxicity and exerted an anti-viral eff ect. RSV infection led to the activation of the TGF-β signaling pathway, which in turn promoted RSV infection. We hyperactivated the TGF-β signaling pathway by TGF-β and found that PG treatment can repress the pro-inflammation eff ect of TGF-β, suggesting that PG inhibits RSV infection through inhibiting TGF-β signaling pathway. Conclusions PG attenuated RSV infection-induced oxidative stress and inflammation through inhibiting the TGF-β signaling pathway. This study provides evidence for the application of traditional Chinese herbal medicine in the treatment of viral infection.

      • Prevalence of Human Papillomavirus 16 in Esophageal Cancer Among the Chinese Population: a Systematic Review and Meta-analysis

        Background and Aim: No firm evidence of HPV infection in esophageal cancer has been established to date. The aim of this meta-analysis was to investigate the prevalence of HPV 16 in esophageal cancer in China, which had a high burden of the disease. Materials and Methods: Studies on HPV infection and esophageal cancer were identified and a random-effects model was used to pool the summary prevalence and corresponding 95% confidence intervals (CIs). Results: A total of 3,429 esophageal cancer cases were evaluated from 26 eligible studies in this meta-analysis. The summary estimate for HPV16 prevalence was 0.381 (95% CI: 0.283, 0.479). The prevalence varied by geographical areas of the study, publication year, HPV detection method and types of specimen. In sensitivity analysis, HPV 16 prevalence ranged from 0.368 (95% CI: 0.276, 0.460) to 0.397 (95% CI: 0.286, 0.508). Conclusions: The results indicate a relatively high level of HPV 16 prevalence in esophageal cancer among Chinese population, although there was variation between different variables. Further studies are needed to elucidate the role of HPV in esophageal carcinogenesis with careful consideration of study design and laboratory detection method, providing more accurate assessment of the HPV status in esophageal cancer.

      • Novel Trust Model in E-Commerce

        This paper theoretically studied the issue of trust in E-Commerce business. Firstly, we established a dynamic game model to analyze conditions required for reaching a long-term equilibrium of trust, by introducing the concept of ESS equilibrium. Secondly, we designed a diffusion mechanism of trust with an algorithm, which helped us understand the process of how the stable equilibrium of trust would be reached. Our findings are as follows: (1) under unfavorable conditions like weak trust incentives, distrust in E-Commerce business is prevalent, even after a trust evaluating system has been established; (2) some factors, including payoff difference between honest traders and dishonest traders, the number of traders choosing the trust strategy and convenience of information exchange between traders, determine the diffusion process of trust in this business. With regard to the findings aforementioned and the sustainable development of E-Commerce business in China, we provide some policy suggestions, like increasing comparative advantage in payoffs received by honest traders, reinforcing regulation on entry permission, and investing more in technology or devices to promote information exchange between traders.

      • Indoor Location Algorithm Based on Kalman Filter and Multi-Source Data Integration

        For onboard single-station passive direction-finding and location, if there is any abnormal error in the observation data, the extended Kalman filter (EKF) algorithm adopted thereby will cause inaccurate location result. In order to improve algorithm robustness, the robust equivalent gain matrix is constructed according to the standardized prediction residual error and the robust EKF algorithm is applied to the onboard single-station passive direction-finding and location. In allusion to the low efficiency of the robust EKF algorithm, the single-station passive location algorithm based on the improved extended Kalman filter is proposed in this article on the basis of combining F distribution statistic, and meanwhile single abnormal error and continuous abnormal error are added in the observation value to test the algorithm resistance to different abnormal errors. The simulation shows that the algorithm proposed in this article can well weaken the influence of abnormal errors on position estimation and the algorithm based on F distribution discriminant can improve location efficiency.

      • Matrix Metalloproteinase-9 as a Prognostic Factor in Gastric Cancer: A Meta-Analysis

        Background: Matrix metalloproteinase-9 (MMP-9) is associated with disruption of basement membranes of blood vessels and promotion of metastasis through the lymphatics. However, its prognostic value for survival in patients with gastric cancer remains controversial. Method: We therefore conducted a meta-analysis of the published literature in order to clarify the impact of MMP-9. Clinical studies were selected for further analysis if they provided an independent assessment of MMP-9 in gastric cancer and reported analysis of survival data according to MMP-9 expression. Results: A total of 11 studies, covering 1700 patients, were included for meta-analysis. A summary hazard ratio (HR) of all studies and sub-group hazard ratios were calculated. The combined HR suggested that a positive MMP-9 expression had an impact on overall survival: 1.25 (95% confidence interval 1.11-1.40) in all eligible studies; 1.13 (1.06-1.20) in 8 studies detecting MMP-9 by immunohistochemistry; 1.36 (1.12-1.65) in 7 studies from Asia. Only one study for DFS showed a significant impact on disease free survival (HR 1.73, 95%CI 1.27-2.34). Conclusions: Our findings suggested that MMP-9 protein expression might be a factor for a poor prognosis in patients with gastric cancer. However, the association was rather weak, so that more prospective studies should further explore the prognostic impact of MMP-9 mRNA and correlations between MMP-9 and clinicopathological characteristics.

        A Vegetable Dietary Pattern Is Associated with Lowered Risk of Gestational Diabetes Mellitus in Chinese Women

        Background Identification of modifiable dietary factors, which are involved in the development of gestational diabetes mellitus (GDM), could inform strategies to prevent GDM. Methods We examined the dietary patterns in a Chinese population and evaluated their relationship with GDM risk using a case-control study including 1,464 cases and 8,092 control subjects. Propensity score matching was used to reduce the imbalance of covariates between cases and controls. Dietary patterns were identified using factor analysis while their associations with GDM risk were evaluated using logistic regression models. Results A “vegetable” dietary pattern was characterized as the consumption of green leafy vegetables (Chinese little greens and bean seedling), other vegetables (cabbages, carrots, tomatoes, eggplants, potatoes, mushrooms, peppers, bamboo shoots, agarics, and garlic), and bean products (soybean milk, tofu, kidney beans, and cowpea). For every quartile increase in the vegetables factor score during 1 year prior to conception, the first trimester, and the second trimester of pregnancy, the GDM risk lowered by 6% (odds ratio [OR], 0.94; 95% confidence interval [CI], 0.89 to 0.99), 7% (OR, 0.94; 95% CI, 0.88 to 0.99), and 9% (OR, 0.91; 95% CI, 0.86 to 0.96). Conclusion In conclusion, our study suggests that the vegetable dietary pattern is associated with lower GDM risk; however, the interpretation of the result should with caution due to the limitations in our study, and additional studies are necessary to explore the underlying mechanism of this relationship.

        Full Waveguide-band E-plane Power Divider with High Output Power Ratio

        This paper presents a new E-plane power divider with high output power ratio that operates over the full rectangular waveguide band. The proposed divider consists of an E-plane junction with rounded corners and a smoothly tapered septum. Output ratios of up to 10 are realized by offsetting the septum while output power ratios greater than 10 are achieved by additionally reducing the broad-wall width in the output waveguides with smaller power. Optimum dimensions of the proposed divider are obtained using a commercial electromagnetic-wave simulation tool. A design is given for a power divider in the WR-10 waveguide that has reflection coefficient of less than –20 ㏈ over 75-110 ㎓ and output power ratios up to 100 at 75 ㎓.

        Nanosheets of copolymerized ionic liquid-based polyelectrolyte complexes regulated at oil–water interface and their emulsification capability

        The emulsification by polyelectrolyte complexes is easy to handle, and could be considered as an effectivestrategy for introducing functional materials to liquid–liquid interfaces, but the related studies are stillinsufficient. Herein, a kind of copolymerized ionic liquid (PIL-co-PVIm) which is synthesized from 1-vinyl-3-ethylimidazolium bromide and 1-vinylimidazole, are used as polycation to form polyelectrolytecomplexes type Pickering emulsifiers with sodium alginate, poly(sodium 4-styrenesulfonate) and poly(acrylic acid) sodium salt respectively as polyanion. For toluene-water emulsion systems andhydrophobic ionic liquid 1-butyl-3-methylimidazolium hexafluorophosphate ([BMIm]PF6)-watersystem, the polyelectrolyte complexes formed by PIL-co-PVIm and sodium alginate or poly (sodium4-styrenesulfonate) showed best emulsification capability. The emulsions stabilized by the polyelectro-lyte complexes at stoichiometric ratio are relatively less stable compared to non-stoichiometriccomplexes, while PIL-co-PVIm without complexation has no emulsification capability. The polyelectro-lyte complexes based on homopolymerized PIL give an unsatisfied emulsification capability, while theemulsification performance could be remarkably improved by copolymerizing ionic liquids with theuncharged comonomer, but excessive contents of the uncharged unit make the polyelectrolytecomplexes less interface active. The sheet-like PECs are formed in aqueous dispersions, and theirthickness is remarkably reduced at oil–water interface after emulsification, approximately ranging from200 nm to 10 nm by manipulating the polyanion.
