      • BRCA1 Gene Exon 11 Mutations in Uighur and Han Women with Early-onset Sporadic Breast Cancer in the Northwest Region of China

        The prevalence of BRCA1 gene mutations in breast cancer differs between diverse ethnic groups. Relatively little information is known about patterns of BRCA1 mutations in early-onset breast cancer in women of Uighur or Han descent, the major ethnic populations of the Xinjiang region in China. The aim of this study was to identify BRCA1 mutations in Uighur and Han patients with early-onset (age <35 years), and sporadic breast cancer for genetic predisposition to breast cancer. For detection of BRCA1 mutations, we used a polymerase chain reaction single-stranded conformation polymorphism approach, followed by direct DNA sequencing in 22 Uighur and 13 Han women with early-onset sporadic breast cancer, and 32 women with benign breast diseases. The prevalence of BRCA1 mutations in this population was 22.9% (8/35) among early-onset sporadic breast cancer cases. Of these, 31.8% (7/22) of Uighur patients and 7.69% (1/13) of Han patients were found to have BRCA1 mutations. In 7 Uighur patients with BRCA1 mutations, there were 11 unique sequence alterations in the BRCA1 gene, including 4 clearly disease-associated mutations on exon 11 and 3 variants of uncertain clinical significance on exon 11, meanwhile 4 neutral variants on intron 20 or 2. None of the 11 BRCA1 mutations identified have been previously reported in the Breast Cancer Information Core database. These findings reflect the prevalence of BRCA1 mutations in Uighur women with early-onset and sporadic breast cancer, which will allow for provision of appropriate genetic counseling and treatment for Uighur patients in the Xinjiang region.

        Protective effect of hepatocyte-enriched lncRNA-Mir122hg by promoting hepatocyte proliferation in acute liver injury

        Some long noncoding RNAs (lncRNAs), which harbor microRNAs in their gene sequence and are also known as microRNA host gene derived lncRNAs (lnc-MIRHGs), play a dominant role alongside miRNAs, or both perform biological functions synergistically or independently. However, only a small number of lnc-MIRHGs have been identified. Here, multiple liver injury datasets were analyzed to screen and identify the target lncRNA Mir122hg. Mir122hg was mainly enriched in liver tissues with human-mouse homology. In both CCl4-induced acute liver injury and Dgal/LPS-induced fulminant liver failure in mice, Mir122hg was sharply downregulated at the early stage, while a subsequent significant increase was only found in the CCl4 group with liver recovery. Overexpression and silencing assays confirmed that Mir122hg played a protective role in acute injury by promoting hepatocyte proliferation in vivo and in vitro. Consistent with the results of gene enrichment analysis, Mir122hg binding to C/EBPα affected its transcriptional repression, promoted gene transcription of downstream chemokines, Cxcl2, Cxcl3, and Cxcl5, and exerted pro-proliferative effects on hepatocytes through activation of the AKT/GSK-3β/p27 signaling pathway by CXC/CXCR2 complexes. This study identifies a novel lncRNA with protective effects in acute liver injury and demonstrates that the binding of Mir122hg-C/EBPα promotes hepatocyte proliferation via upregulation of CXC chemokine and activation of AKT signaling.

      • Acute Normovolemic Hemodilution Effects on Perioperative Coagulation in Elderly Patients Undergoing Hepatic Carcinectomy

        Background: Acute normovolemic hemodilution (ANH) has been widely used to prevent the massive blood loss during hepatic carcinoma. The influences of ANH on coagulation function are still controversy, especially in elderly patients. The study observed ANH effects on coagulation function and fibrinolysis in elderly patients undergoing the disease. Materials and Methods: Thirty elderly patients (aged 60-70 yr) with liver cancer (ASA I or II) taken hepatic carcinectomy from February 2007 to February 2008 were randomly divided into ANH group (n=15) and control group (n=15). After tracheal intubation, patients in ANH group and control group were infused with 6% hydroxyethyl starch (130/0.4) and Ringer's solution, respectively. Blood samples were drawn from patients in both groups at five different time points: before anesthesia induction (T1), 30 min after ANH (T2), 1 h after start of operation (T3), immediately after operation (T4), and 24 h after operation (T5). Then coagulation function, soluble fibrin monomer complex (SFMC), prothrombin fragment (F1+2), and platelet membrane glycoprotein (CD62P and activated GP IIb/GP IIIa) were measured. Results: The perioperative blood loss and allogeneic blood transfusion were recorded during the surgery. The perioperative blood loss was not significantly different between two groups (p>0.05), but the volume of allogeneic blood transfusion in ANH group was significantly less than in control group ($350.0{\pm}70.7$) mL vs. ($457.0{\pm}181.3$) mL (p<0.01). Compared with the data of T1, the prothrombin time (PT) and activated partial thromboplastin time (APTT) measured after T3 were significantly longer (p<0.05) in both groups, but within normal range. There were no significant changes of thrombin time (TT) and D-dimer between two groups at different time points (p>0.05). SFMC and F1+2 increased in both groups, but were not statistically significant. PAC-1-positive cells and CD62P expressions in patients of ANH group were significantly lower than those at T1 (p<0.05) and T2-T5 (p>0.05). Conclusions: ANH has no obvious impact on fibrinolysis and coagulation function in elderly patients undergoing resection of liver cancer. The study suggested that ANH is safe to use in elderly patients and it could reduce allogeneic blood transfusion.

        Mechanical behavior of rock-coal-rock specimens with different coal thicknesses

        To explore the influence of coal thickness on the mechanical behavior and the failure characteristics of rock-coal-rock (RCR) mass, the experimental investigation of uniaxial compressive tests was conducted first and then a systematic numerical simulation by particle flow code (PFC2D) was performed to deeply analyze the failure mechanical behavior of RCR specimens with different coal thicknesses in conventional compression tests. The overall elastic modulus and peak stress of RCR specimens lie between the rock and the coal. Inter-particle properties were calibrated to match the physical sample strength and the stiffness response. Numerical simulation results show that the deformation and strength behaviors of RCR specimens depend not only on the coal thickness, but also on the confining pressure. Under low confining pressures, the overall failure mechanism of RCR specimen is the serious damage of coal section when the coal thickness is smaller than 30 mm, but it is shear failure of coal section when the coal thickness is larger than 30 mm. Whereas under high confining pressures, obvious shear bands exist in both the coal section and the rock section when the coal thickness is larger than 30 mm, but when the coal thickness is smaller than 30mm, the failure mechanism is serious damage of coal section and shear failure of rock section.

        Prevalence, Risk Factors, and Psychological Effects of Overactive Bladder in Chinese University Students

        Purpose: The purpose of this study was to investigate the prevalence and risk factors of overactive bladder (OAB) in young adults and to explore the influence of OAB on mental health. Methods: Between October 2019 and January 2020, 14,010 anonymous questionnaires were distributed to freshmen at 2 universities in Henan, China. The students came from all over the country. The questionnaire included general items and information necessary to calculate the overactive bladder symptom score, the Chinese version of the Pittsburgh Sleep Quality Index (PSQI) score, Self-Esteem Scale (SES) score, and Self-Rating Depression Scale (SDS) score. The relationships between the prevalence of OAB and its risk factors were evaluated. Results: The overall prevalence of OAB was 6.0%, with 4.3% of participants characterized as having dry OAB and 1.7% as having wet OAB. The prevalence of mild OAB was 5.5%, and that of moderate OAB was 0.5%; no severe OAB was observed. Higher prevalence rates of OAB were found among women, respondents with constipation, and respondents with primary nocturnal enuresis (PNE) (P <0.05). Compared to healthy controls, the OAB group exhibited a higher mean SDS score (52.12±8.986 vs. 47.71±9.399, P<0.001) and mean PSQI score (5.28±2.486 vs. 4.27±2.431, P<0.001), but a lower mean SES score (27.78±3.599 vs. 29.57±4.109, P<0.001). Conclusions: OAB significantly affects the mental health of young adults. Female sex, constipation, and PNE are risk factors for OAB.

      • Knockdown of Cdc25B in Renal Cell Carcinoma is Associated with Decreased Malignant Features

        Cdc25 phosphatases are important regulators of the cell cycle. Their abnormal expression detected in a number of tumors implies that their dysregulation is involved in malignant transformation. However, the role of Cdc25B in renal cell carcinomas remains unknown. To shed light on influence on renal cell carcinogenesis and subsequent progression, Cdc25B expression was examined by real-time RT-PCR and western blotting in renal cell carcinoma and normal tissues. 65 kDa Cdc25B expression was higher in carcinomas than in the adjacent normal tissues (P<0.05), positive correlations being noted with clinical stage and histopathologic grade (P<0.05). To additionally investigate the role of Cdc25B alteration in the development of renal cell carcinoma, Cdc25B siRNA was used to knockdown the expression of Cdc25B. Down-regulation resulted in slower growth, more G2/M cells, weaker capacity for migration and invasion, and induction of apoptosis in 769-P transfectants. Reduction of 14-3-3 protein expression appeared related to Cdc25B knockdown. These findings suggest an important role of Cdc25B in renal cell carcinoma development and provide a rationale for investigation of Cdc2B-based gene therapy.

        Prediction and Monitoring of the Construction Vibration Effect on an Adjacent Old Long Span Double-Convex Arch Bridge

        Ground vibrations caused by the construction of impact drilling piles may affect adjacent bridges, so corresponding prediction and monitoring are necessary to ensure the safety of bridges. In this study, the construction near an existing long-span double-convex arch bridge is taken as a case study to investigate different layouts of construction machines emanating harmful ground vibrations to the existing adjacent bridge by numerical simulations and monitoring. The peak acceleration and velocity acquired from numerical simulations, and monitoring were successfully used to implement impact drilling pile construction near the existing bridge. The results show that there are good consistencies between the monitoring and numerical simulation results, and the vibration acceleration of the foundation of abutments and piers are mainly derived from the vibration caused by the construction within a distance of 50 m, while the construction beyond 50 m has little effect on the old bridge. The numbers and locations of machines near the same foundation may have a weakening or strengthening effect on the peak acceleration and velocity due to destructive and constructive interference of waves, which can help to control the effects of the vibrations of the existing adjacent bridge.

        Modelling of the Hot Flow Behaviors for Ti-13Nb-13Zr Alloy by BP-ANN Model and Its Application

        The plastic deformation mechanisms and the constitutive model of flow behaviors at different deformation conditions in biomedical titanium alloy are an essential step to optimize the design of any forging process for implant productions. A series of isothermal compressions tests on Ti-13Nb-13Zr alloy in a wide range of true strain, temperature and strain rate were conducted on a thermomechanical simulator. The hot flow behaviors with different softening mechanisms, including dynamic recrystallization and dynamic recovery, were characterized based on true strain-stress curves. A back-propagational artificial neural network (BP-ANN) method was conducted to evaluate and predict this non-linear problem by self-training to be adaptable to the material characteristics. The flow stress of this material a wide deformation condition range can be predicted accurately by the BP-ANN model obtained in this study. The prediction ability of this BP-ANN Model was evaluated by three accuracy indexes, Absolute error, Relative error and Average absolute relative error. Sequently, the developed BP-ANN model was programed and implanted into the finite element (FE) analysis platform, Msc.Marc software. The results have sufficiently articulated that the well-trained ANN model has excellent capability to deal with the complex flow behaviors and has great application potentiality in hot deformation processes.

        A novel M2e-multiple antigenic peptide providing heterologous protection in mice

        Swine influenza viruses (SwIVs) cause considerable morbidity and mortality in domestic pigs, resulting in a significant economic burden. Moreover, pigs have been considered to be a possible mixing vessel in which novel strains loom. Here, we developed and evaluated a novel M2e-multiple antigenic peptide (M2e-MAP) as a supplemental antigen for inactivated H3N2 vaccine to provide cross-protection against two main subtypes of SwIVs, H1N1 and H3N2. The novel tetra-branched MAP was constructed by fusing four copies of M2e to one copy of foreign T helper cell epitopes. A high-yield reassortant H3N2 virus was generated by plasmid based reverse genetics. The efficacy of the novel H3N2 inactivated vaccines with or without M2e-MAP supplementation was evaluated in a mouse model. M2e-MAP conjugated vaccine induced strong antibody responses in mice. Complete protection against the heterologous swine H1N1 virus was observed in mice vaccinated with M2e-MAP combined vaccine. Moreover, this novel peptide confers protection against lethal challenge of A/Puerto Rico/8/34 (H1N1). Taken together, our results suggest the combined immunization of reassortant inactivated H3N2 vaccine and the novel M2e-MAP provided cross-protection against swine and human viruses and may serve as a promising approach for influenza vaccine development.
