Helicobacter pylori와 대장균의 Shuttle Vector 개발

In this study, a vehicle vector using cryptic plasmids was constructed for gene transfer in Helicobacter pylori. pHP51(3.9 kb) and pHP489(1.2 kb) were selected for constructing vectors from cryptic plasmid of H. pylori isolates in Korea. The HindⅢ-digestedDNA fragment(1.2kb) of pHP489 and 1.6kb DNA fragment of pHP51 were ligated with a kanamycin resistance gene(aph3'-Ⅲ) from C. jejuni to produce the recombinant plasmids pHP489K and pHP51K, respectively. Transformation frequency of pHP51K by electroporation was low. But pHP489K could be effectively transformed into various H. pylori strains. In order to design an intermdiate vehicle vector for gene transfer into H. pylori, pBlueHP489K was prepared by recloning pHP489K DNA into pBluescript and pTZ19R vector. This vector permitted the DNA fragment containing pHP489 sequence, aph3'-Ⅲ, and cloned DNA to be cut and self-ligated in the SacⅠ site after cloning. ureA and ureB gene were inserted into pBlueHP489K, resulting in pBlueHP489K/AB. The DNA fragment containing pHP489, kanamycin resistance gene(aph3'-Ⅲ), and urease structural gene was cut away from pBlueHP489K/AB and self-ligated to generate pBlueHP489K/AB. pBlueHP489K/AB made urease-negative H. pylori strains restore their urease activity. By this experiment, pBlueHP489K was confirmed to be the vehicle system for transferring H. pylori genes.

Site characterization is one of the most important performances in projects with regard to solving geotechnical problems. Piezocone penetration test is a fast, economic, and accurate method of site characterization. Static cone penetrometer and the piezocone represents the most versatile tools currently available for soil exploration. The cone penetration and piezocone tests provide continuous sounding capability and repeatability. Rare of consolidation parameter may be assessed form the Piezocone test by measuring the dissipation or decay of pore pressure with time after a stop in penetration. And many researches have been developed to determine the horizontal coefficent of consolidation from the Piezocone test. Many of these researches using a theoretical solutions which is determined from either the cavity expantion theory or the stress path method. However, depending on assumptions and analytical techniques, the results can be varied considerably. For that reason, many researchers make their effort to improve and complement the defect of already established methods. In this dissertation, the correlation between the Coefficient of Consolidation from Piezocone dissipation test and the Coefficient of Consolidation Rowe Cell horizontal consolidation test in many cases was studied. 특정한 이득여유와 위상여유를 이용한 PID 동조 알고리즘

이득여유와 위상여유를 보장하는 견실한 제어기의 설계는 arctan 함수의 근사화로 가능하게 되었다. 이 논문은 arctan 함수의 새로운 근사화로 특정한 이득 여유와 위상여유를 기초로한 PID 동조 알고리즘을 제안한다. arctan 함수의 새로운 근사화 방법은 arctan(0), arctan(0.5) 이용한 1차선형 보간식과 arctan(0.5), arctan(1)을 이용한 1차선형 보간식 2개를 선형 결합하여 근사화 하였다. 제안된 방법의 주파수 응답 특성에서 특정한 이득여유와 위상여유를 보장함을 알 수 있다. The robust design of controllers to ensure gain and phase margin is can be use approximation of arctan function. In this paper, We proposed a tuning algorithm PID controllers based on specified gain and phase margin by a new approximation of arctan function. This method have linear interpolation equations of two arctan interval instead of one arctan internal of arctan(x). It is shown that the frequency response of this method was to ensure specified gain and phase margin.

E. Rutherford proposed a new theory of the scattering of alpha-particles by a matter. This theory was based on a new atomic and was succesful in describing the experimental results. The curves in Ruterford's model an alpha-particle near a nucleus are hyper-bolas, all with the nucleus at the focus by the Coulomb's law, Newton's law of motion, and the detailed geometrical arrangement. The essentials of alpha-particle scattering reproduce the methods of inquiring learning through the following three kinds of mechanical models. Theses models were many times analyzed by means of feedback and compar-ed with the experimental results by precise appliances. 1) Conic and ball model. 2) Magnet and magneticpendulum model. 3) Electrostatic body and styrofoam ball pendulum model.

본 연구는 주형의 형상에 관계없이 균일한 건조성능을 가지는 장치의 도입을 목적으로 Microwave를이용한 주형건조장치 개발을 위한 기초 연구에 관한 것이다. 건조 성능의 검토를 위해 진공건조기와Lab-scale의 Microwave 출력장치를 가지는 Microwave oven을 이용하여 주물사의 무게별, 건조시간별 건조효율을 검토하였으며, CO₂주형법에서 물유리의 양과 CO₂통기시간에 따른 주형의 압축강도를 측정하였다.

국내에서 생산되고 있는 철근의 방청효과는 내식성기준(KS D 9520)으로 1시간 미만으로 생산된 철근의 국외 수출시 방청성에 대한 요구가 많은 실정이다. 이에 따라 철근의 내식성 향상을 위해 소요되는 비용과 과다한 방청포장은 제품 경쟁력에 악영향을 끼치고 있다. 내식성 향상을 위한 Cr 대체물질 개발은 많으나 철근에 고니식성 및 고내열성을 부여하기 위한 Cr 대체물질 개발은 미진하다. 본 연구에서는 고온안정성과 고내식성을 동시에 만족하고, 환경유해물질을 포함하지 않는 새로운 소재를 개발하기 위해 수행하였으며, 다양한 제법에 따라 제조된 물질의 내식성 실험을 통해 방청제로서의 가능성을 검토하기 위하여 수행하였다.
