<B>Background</B><P>Glucagon-like peptide-1 (GLP-1) is a gut-derived incretin hormone that plays an important role in glucose homeostasis. Its functions include glucose-stimulated insulin secretion, suppression of glucagon secretion, deceleration of gastric emptying, and reduction in appetite and food intake. Despite the numerous antidiabetic properties of GLP-1, its therapeutic potential is limited by its short biological half-life due to rapid enzymatic degradation by dipeptidyl peptidase IV. The present study aimed to demonstrate the therapeutic effects of constitutively expressed GLP-1 in an overt type 2 diabetic animal model using an adenoviral vector system.</P><B>Methods</B><P>A novel plasmid (pAAV-ILGLP-1) and recombinant adenoviral vector (Ad-ILGLP-1) were constructed with the cytomegalovirus promoter and insulin leader sequence followed by GLP-1(7–37) cDNA.</P><B>Results</B><P>The results of an enzyme-linked immunosorbent assay showed significantly elevated levels of GLP-1(7–37) secreted by human embryonic kidney cells transfected with the construct containing the leader sequence. A single intravenous administration of Ad-ILGLP-1 into 12-week-old Zucker diabetic fatty (ZDF) rats, which have overt type 2 diabetes mellitus (T2DM), achieved near normoglycemia for 3 weeks and improved utilization of blood glucose in glucose tolerance tests. Circulating plasma levels of GLP-1 increased in GLP-1-treated ZDF rats, but diminished 21 days after treatment. When compared with controls, Ad-ILGLP-1-treated ZDF rats had a lower homeostasis model assessment for insulin resistance score indicating amelioration in insulin resistance. Immunohistochemical staining showed that cells expressing GLP-1 were found in the livers of GLP-1-treated ZDF rats.</P><B>Conclusions</B><P>These data suggest that GLP-1 gene therapy can improve glucose homeostasis in fully developed diabetic animal models and may be a promising treatment modality for T2DM in humans. Mp Criterion Based Multiloop PID Controllers Tuning for Desired Closed Loop Responses

A tuning method of multiloop PID controllers is developed based on the generalized IMC PID tuning rule by Lee et al. (1998a). To extend the SISO PID tuning method to MIMO systems, a new tuning criterion is proposed. The criterion is based on the closed loop frequency response method to meet desired performance and robustness as close as possible. Examples for 2*2, 3*3 and 4*4 systems are used to illustrate the proposed method. The results show that the proposed method is superior to conventional method such as the BLT tuning method.

Background: As self-employed workers are vulnerable to health problems, this study aimed to analyze mental health problems and sleep disturbances among self-employed workers compared with paid workers in Korea. Methods: A total of 34,750 workers (23,938 paid workers and 10,812 self-employed workers) were analyzed from the fifth Korean Working Condition Survey, which included 50,205 households collected by stratified sampling in 2017. To compare mental health problems and sleep disturbance among self-employed workers and paid workers, multivariate logistic regression analyses were performed. Results: The odds ratio in self-employed workers compared with paid workers was 1.25 (95% confidence interval [CI]: 1.09–1.42) for anxiety, 1.11 (95% CI: 1.04–1.17) for overall fatigue, 1.11 (95% CI: 1.04–1.20) for difficulty falling asleep, 1.10 (95% CI: 1.02–1.18) for difficulty maintaining sleep and 1.24 (95% CI: 1.16–1.32) for extreme fatigue after waking up. Conclusions: Self-employed workers in Korea have a higher risk of self-reported mental health problems and sleep disturbances than paid workers. Further studies with a longitudinal design and structured evaluation are required to investigate the causal relationship between health problems and self-employment.

<P>A screening method for simultaneous analysis of 379 pesticides in human serum was developed using liquid chromatography-tandem mass spectrometry (LC-MS/MS). Electrospray ionization with positive/negative switching mode of LC-MS/MS was adopted, and scheduled multiple reaction monitoring for each target compound was established. The limit of quantitation was 10 ng/mL for 94.5% of the total pesticides, and the correlation coefficients of calibration were ≥0.990 for 93.9% of the pesticides. For the sample preparation, scaled-down QuEChERS were used. Serum (100 μL) was extracted with acetonitrile (400 μL), partitioned with magnesium sulfate (40 mg) and sodium chloride (10 mg), and the upper layer was used for analysis without further cleanup steps. For the accuracy and precision tests, most of the pesticides showed excellent results in intra- and interday conditions. In the recovery tests at 10, 50, and 250 ng/mL, 85.8-91.8% of all target compounds satisfied the recovery range of 70-120% (relative standard deviation ≤20%).</P> [FIG OMISSION]</BR>

Objectives: To identify and investigate through qualitative and quantitative analysis the hazardous substances generated when compressed wood was burned at a live fire-training center. Methods: Four types of compressed wood that are actually used in live fire training were burned in a chamber according to KS F2271. The gaseous material was sampled with a gas detector tube and conventional personal samplers. Results: 1,3-butadiene, benzene, toluene, xylene, formaldehyde, hydrogen chloride, hydrogen cyanide, ammonia, carbon monoxide, and nitric acid were detected. In particular, 1,3-butadiene (497.04-680.44 ppm), benzene (97.79-125.02 ppm), formaldehyde (1.72-13.03 ppm), hydrogen chloride (4.71-15.66 ppm), hydrogen cyanide (3.64-8.57 ppm), and sulfuric acid (3.85-5.01 ppm) exceeded the Korean Occupational Exposure Limit as measured by sampling pump according to the type of compressed wood. Conclusions: We found through the chamber testing that firefighters could be exposed to toxic substances during live fire training. Therefore, firefighter protection is needed and more research is required in the field.

<P>A multiresidue method for the simultaneous and rapid analysis of 360 pesticides in representative agricultural produce (brown rice, orange, spinach, and potato) was developed using a modified QuEChERS procedure combined with gas chromatography tandem mass spectrometry (GC-MS/MS). Selected reaction monitoring transition parameters (e.g., collision energy, precursor and product ions) in MS/MS were optimized to achieve the best selectivity and sensitivity for a wide range of GC-amenable pesticides. A short (20 m) microbore (0.18 mm i.d.) column resulted in better signal-to-noise ratio with reduced analysis time than a conventional narrowbore column (30 m X 0.25 mm i.d.). The priming injection dramatically increased peak areas by masking effect on a new GC liner. The limit of quantitation was <0.01 mg/kg, and the correlation coefficients (r(2)) of matrix-matched standards were >0.99 within the range of 0.0025-0.1 mg/kg. Acetonitrile with 0.1% formic acid without additional buffer salts was used for pesticide extraction, whereas only primary secondary amine (PSA) was used for dispersive solid phase extraction (dSPE) cleanup, to achieve good recoveries for most of the target analytes. The recoveries ranged from 70 to 120% with relative standard deviations of <= 20% at 0.01 and 0.05 mg/kg spiking levels (n = 6) in all samples, indicating acceptable accuracy and precision of the method. Seventeen real samples from local markets were analyzed by using the optimized method, and 14 pesticides in 11 incurred samples were found at below the maximum residue limits.</P>

<P><B>Abstract</B></P> <P>In this study, we developed a multiresidue method for the analysis of 310 pesticides in representative agricultural produce (brown rice, orange, and spinach) using ultra high performance liquid chromatography–tandem mass spectrometry (UHPLC-MS/MS) combined with a modified QuEChERS procedure. The optimal mobile phase composition (Methanol containing 5 mM ammonium formate and 0.1% formic acid) produced high sensitivity and reliable results. Also, the relationship between injection volume and repeatability of peak area was investigated. Most of the target pesticides had a limit of quantitation under 10 ng g<SUP>−1</SUP>, and correlation coefficients (<I>r</I> <SUP>2</SUP>) > 0.99 in matrix-matched standards within the range of 1–100 ng g<SUP>−1</SUP>. To validate the optimized method, recovery tests were performed with each of the crops at 10 and 50 ng g<SUP>−1</SUP> spiking levels (<I>n</I> = <I>5</I>). Satisfactory recoveries were achieved showing that 86.8–88.7% (at 10 ng g<SUP>−1</SUP>) and 91.9–96.1% (at 50 ng g<SUP>−1</SUP>) of the pesticides met the validation criteria (recoveries in the range of 70–120% and relative standard deviation ≤ 20%). Fifteen compounds were found to show a loss of recovery due to adsorption by primary and secondary amine or graphite carbon black. In the case of brown rice, 86.1% of pesticides showed an insignificant matrix effect (<±20%), while 35.2% and 41.6% of pesticides in orange and spinach were in that range, respectively. Sixteen apple samples from local markets were analyzed to evaluate the applicability of the optimized method. Nineteen pesticides were detected, of which the concentrations were lower than the maximum residue limit.</P> <P><B>Highlights</B></P> <P> <UL> <LI> The protocol can analyze large number of pesticides (310 analytes) with modified QuEChERS and LC-MS/MS. </LI> <LI> A relationship between injection volumes and repeatability in LC-MS/MS analysis. </LI> <LI> Mobile phase compositions were compared for best sensitivity in LC-MS/MS. </LI> <LI> The method is reproducible and sensitive. </LI> <LI> It was successfully used to determine pesticide residues in market samples. </LI> </UL> </P>
