국내 설사환자로부터 분리된 Escherichia coli 균의 혈청군 및 Verocytotoxin 생성능

목적: 국내 설사 환자에게서 분리 동정된 인체 감염 유래 E. coli 균의 혈청군, verocytotoxin 생성능을 구명하여 EHEC 균의 인체 감염 실태를 구명하고, 동물 및 식품 유래 분리균주의 성상과 비교 검토하여 인체 감염 위험성에 대한 기초적 자료를 의학계에 제공하고자 하였다. 방법: EHEC 감염의심 설사 환자 분변 검체로부터 분리된 균주 중 생화확적 성상에 따라 대장균을 분리하였고 이들 균주 중 E. coli O157:H7를 분리하기 위해서는 Dsorbitol 음성균을 선별하였으며, 분리균에 대하여 항혈청으로 응집 시험을 실시하여 혈청군을 구명하였다. 혈청군이 확인된 균주에 대하여 중합효소 연쇄반응 (PCR) 및 역수동라텍스 응집시험 (Reversed Passive Latex Agglutination:RPLA) 으로 verocytotoxin 생성능을 구명하였다. 결과: 장출혈성 대장균 감염 의심설사 환자의 분변검체로부터 25주의 E. coli 균을 분리 동정하였으나 이들 균주는 모두 verocytotoxin을 생성하지 않았다. 분리 동정된 대장균으로부터 확인된 혈청군은 16종으로서 E. coli O1, O6, O8, O15, O20, O25 O26, O28, O29, O44, O86a, O119, O126, O128, O152, 및 O157:H-였으며 E. coli O157:H-혈청형 균주와 E.coli O25 혈청군 균주가 각각 3주씩 분리되어 비교적 높은 분리율을 보였다. 결론: 장출혈성 대장균 감염 의심설사 환자 분변 가검물로부터 총 25주의 E. coli균을 분리 동정하였으나 verocytotoxin을 모두 생산하지 않았다. Background: Since 1982, many countries has reported outbreaks or sporadic cases caused by enterohaemorrhagic Escherichia coli (EHEC) serogroup strains, mainly E. coli O157:H7 type strain. However, systemic investigation about EHEC agents, including E. coli O157:H7, have not been done in Korea. Therefore, we investigated serogroup and verotoxin productivity of E. coli strains isolated from diarrheal patients and estimated risk of human infection in comparison with EHEC strains isolated from cow, pig, and food material in Korea. Methods: Diarrheal patient stool samples were collected and E. coli strains were isolated, according to biochemical characteristics. In order to isolate E. coli O157:H7, D-Sorbitol negative strains were selected. Serogrouping of the E. coli isolates was done by agglutination test. Verocytotoxin productivity was investigated by polymerase chain reaction (PCR) and reversed passive latex agglutination (RPLA). Human infection risk was estimated in comparison with EHEC strains isolated from cow, pig and food materials in Korea. Results: Twenty-five E. coli strains were isolated from the diarrheal patients who were suspected to be infected with EHEC. However, none of these E. coli strains produced verocytotoxin. Out of 25 E. coli isolates, 16 serogroups of E. coli O1, O6, O8, O15, O20, O25, O26, O28, O29, O44, O86a, O119, O126, O128, O152 and 157:H-were found. In each of the E. coli O157:H-and O25 serogroups 3 strains were found. Conclusion: None of 25 E. coli isolated from diarrheal patients who were suspected of EHEC infection produced verocytotoxin producing E. coli have been reported recently in Korea.

The purpose of this study is to evaluate for application of the new type of floor heating system with an improved performance than an existed floor heating system in apartment houses. The warmer and moderate part Ondol system suggested in this study were applied to the experimental apartment house and the mock-up room. The field measurements for this system were executed in winter climates. As a result of field measurement, when the load differences between perimeter zone and central zone are increased, the separately controled Ondol system divided in the warmer and moderate part can be evaluated as the new Ondol system with a capability for suppling proper heating energy for each zone. The user can have the freedom for changing warmer parts and the moderate parts and controlling each temperature according to their needs.
cDNA Library Construction of Aphis gossypii Using Gateway Cloning System

Aphis gossypii was widely distributed throughout the tropical, subtropical and temperate zone. The chemical control of A. gossypii is becoming problem because it was rapidly appeared resistance expression to chemicals. We will attempt to resolve the this problem using RNAi technique. Besides, RNAi technology can be helpful to study the target genes of A. gossypii. In this study we produce cDNA library construction using gateway cloning system for selecting target gene in order to control of A. gossypii using RNAi. As a result, the 100-400bp of insert size, which is appropriate for RNAi was confirmed. Most of insert gene is associated with A. gossypii, after that insert sequence was compared with DNA databases and EST databases using NCBI blast search. Consequentially, A. gossypii of cDNA library with the titer of 3.15x105 clones were completed. And we will perform the LR recombination to transfer cDNA library into TRV2 (tobacco rattle virus) vector with att site. Then, after performing transformation using Agrobacterium tumefaciens (GV 2260), we inoculated to cucumber with A. tumefaciens. An insecticidal effect or a repellent activity against A. gossypii by changing behavior in transgenic cucumber plants were conformed. Also, the selecting target gene in order to control A. gossypii using RNAi may be provided.

목적: 최근 조기대장암의 내시경 치료의 가능성에 대한 관심이 높아지고 있다. 이에 저자들은 조기대장암의 임상양상 및 치료결과에 대하여 알아보고자 하였다. 대상 및 방법: 1999년 7월부터 2004년 6월까지 대장암으로 진단 받은 969명 중 117명(120병소)의 조기대장암 환자의 의무기록을 후향적으로 조사하였다. 결과: 전체 대장암 중 조기대장암의 빈도는 12.1%였고 점막암이 80예, 점막하층암이 40예였다. 조기대장암에서 병변의 평균크기는 1.8 cm, 암조직의 평균크기는 1.1 cm였다. 병변의 평균크기는 점막하층암이 2.2 cm로 점막암의 1.7 cm보다 컸다(p=0.03). 육안적 분류로 편평형에서 점막하층암이 30%로 점막암의 15%보다 높은 경향을 보였다(p=0.07). 조직학적 분화도는 고분화선암이 77.5%였으며, 중등도 분화를 보인 경우는 점막하층암에서 63%로 점막암의 3%보다 유의하게 높았다(p<0.001). 조기대장암에 대한 치료는 점막암 80예중 초치료로 10예에서 수술을 시행하였고, 70예에서 내시경 절제를 하였으며, 이 중 2예에서 추가 수술적 절제를 시행하였다. 점막하층암 40예에서는 초치료로 19예에서 수술을 시행하였으며, 21예가 내시경 절제를 받았고 이 중 13예에서 추가 수술적 절제를 받았다. 결론: 본 연구에서 조기대장암의 빈도는 12.1%로 국내 과거 보고보다 증가하고 있으며, 조기대장암에서 내시경 치료를 고려하는 경우 크기가 2 cm 이상이며 중등도 분화도를 보이는 편평형 조기대장암에서는 점막하층암일 가능성을 고려하는 것이 치료에 도움이 될 것으로 생각한다. Background/Aims: Recently, it has been more interested in the early colorectal cancer (ECC) with possibility of endoscopic treatment. We evaluated the clinicopathological characteristics and the treatment results of the ECC. Methods: 120 cases of ECC from 969 patients with colon cancer were analyzed. The cases of concomitant advanced colon cancer were excluded. Results: 12.1% of total colon cancer was ECC. 80 and 40 cases were mucosal (M) and submucosal (SM) cancer, respectively. The size of lesion and cancer tissue of SM cancer was significantly larger than that of M cancer (lesion size, 2.2 vs. 1.7 cm, p=0.03; cancer size, 1.7 vs. 0.7 cm, p<0.001). The prevalence of flat type in SM cancer was higher than that of M cancer (30% vs. 15%, p=0.07). Moderately-differentiated type was more common in SM cancer than M cancer (63% vs. 3%, p<0.001). 15 cases received the additional operation (M cancer; 2, SM cancer; 13). The metastasis of lymph nodes was verified in 2 cases of SM cancer. Conclusions: The ECC with flat type, more than 2 cm in size and moderately-differentiated type had the possibility of SM cancer. Therefore endoscopic treatment in patients with those characteristics should be considered with caution. (Intestinal Research 2005;3:133-139)
Synthesis of α-Ketobutyrolactones and γ-Hydroxy-α-Keto Acids

In connection with the studies for developing new enzymes that could be useful in organic synthesis, practical preparation of racemic and enantiopure forms of ${\gamma}$-hydroxy-${\alpha}$-keto acids has been successfully achieved. For racemic form of ${\gamma}$-hydroxy-${\alpha}$-keto acids, indium-mediated allylation of aldehydes with 2-(bromomethyl)acrylic acid has been employed as a key step. Oxidative cleavage of the thus formed 2-methylenebutyrolactones provided the desired ${\alpha}$-ketobutyrolactones. Enzymatic resolution of the ${\gamma}$-hydroxy-${\alpha}$-methylene esters provided the desired${\gamma}$-hydroxy-${\alpha}$-methylene acids which were successfully converted to ${\gamma}$-hydroxy-${\alpha}$-ketobutyrolactones in optically pure forms.

To elucidate the putative role of the amido group in the metal binding of the fuculose-l-phosphate aldolase from Methanococcus jannaschii, we have examined a potential target using site-directed mutagenesis. The replacement of asparagine 25 with leucine or threonine was shown to have a negative effect, not only on catalytic efficiency, but also on substrate recognition as well. The Hill coefficient values yielded a value of 1. All metals used with the wild-type aldolases exhibited higher activity than that of the mutants. The spectra of the mutants were quite different from the wildtype aldolase. A highly conserved amino acid of asparagine 25 in a related family of aldolase does not appear to provide sufficient evidence for evolution.

Purpose: This study evaluated differences and shifting patterns in the health-related quality of life (HRQoL) of 5-year gastric cancer survivors after either a distal subtotal gastrectomy (DSG) or total gastrectomy (TG). Materials and Methods: We analyzed the prospectively collected HRQoL data of 528 patients who survived 5 years without recurrence using the European Organization for the Research and Treatment of Cancer (EORTC) Core Quality of Life Questionnaire and the EORTC Quality of Life Questionnaire-Stomach module according to the type of surgery. The purpose was to identify the proportion of patients with deteriorating HRQoL and to assess the clinical significance of these changes. Results: Deteriorating HRQoL was prevalent in both groups, including a large proportion of the DSG group. Decreased overall health status and scores on several function scales were less in the DSG group, while increases on the symptom scales were higher in the TG group. For most of the scales, gaps in HRQoL during the early postoperative period did not merge within the 5 years. Scores on the diarrhea and body image scales revealed "moderate changes" in both groups. Conclusions: During the 5-year period after surgery, the TG group suffered from inferior HRQoL compared to the DSG group. However, a large proportion of the DSG group also suffered HRQoL deterioration. In general, the TG group experienced more HRQoL decline, with diarrhea and body image being the major concerns for both groups. To improve HRQoL after gastrectomy, patients must be better informed about post-gastrectomy symptoms. These symptoms must be vigorously investigated, and medical interventions should be available parallel to nutritional support. Favorable evidence of function-preserving gastrectomy should be established and disseminated to improve the HRQoL of early gastric cancer patients.
