김유진,나예린,이현호,오계헌 순천향대학교 기초과학연구소 2021 순천향자연과학연구 논문집 Vol.27 No.1.2
The aim of this work was to investigate the physiological activities of capsaicin-resistant bacterium, Leuconostoc mesenteroides GK-10 isolated from fully fermented mustard leaf kimchi. Initially, the morphological, physiological and biochemical characteristics of strain GK-10 were examined. Based on phylogenetic analysis using 16S rRNA sequencing, the isolate was identified as L. mesenteroides GK-10 and phylogenetic tree of the strain was plotted. L. mesenteroides GK-10 grew well in the different concentrations of capsaicin (0-600 μg/mL). Various physiological activities (e.g., tyrosinase inhibitory activity, ACE inhibitory activity, antioxidant activity, SOD-like activity, depletion of sodium nitrite) of L. mesenteroides GK-10 cultures with different concentrations of capsaicin were evaluated and compared to the cultures of L. mesenteroides DK-3 as a standard strain. According to the results, all of the physiological activities evaluated in this work were considerably higher in L. mesenteroides GK-10 cultures with 600 μg/mL capsaicin compared to L. mesenteroides DK-3 cultures with the same capsaicin concentration, although the physiological activities of GK-10 were higher or lower compared to those of DK-3 in the absence of capsaicin. In consequence, this work showed that L. mesenteroides GK-10 strains can maintain their physiological activities despite the presence of high concentrations of capsaicin.

Background: Red ginseng polysaccharides (RGPs) have been acknowledged for their outstanding immunomodulation and anti-tumor activities. However, their studies are still limited by the complexity of their structural features, the absence of purification and enrichment methods, and the rarity of the analytical instruments that apply to the analysis of such macromolecules. Thus, this study is an attempt to establish a new mass spectrometry (MS)-based analysis procedure for RGPs. Methods: Saponin pre-excluded powder of RG (RG-SPEP, 10 mg) was treated with 200 mL of distilled water and centrifuged for 5 h at 1000 rpm and 85 _C. Ethanol-based precipitation and centrifugation were applied to obtain RGPs from the heated extracts. Further, endo-carbohydrase treatments were performed to produce specific saccharide fragments. Solid-phase extraction (SPE) processes were implemented to purify and enrich the enzyme-treated RGPs, while matrix-assisted laser desorption/ionization time-of-flight/time-of-flight (MALDI-TOF/TOF) MS was employed for the partial structural analysis of the obtained RGPs. Results: Utilizing cellulase, porous graphitized carbon (PGC), hydrophilic interaction chromatography (HILIC), and MALDI-TOF/TOF MS, the neutral and acidic RGPs were qualitatively analyzed. Hexn and Hexn-18 (cellulose analogs) were determined to be novel neutral RGPs. Additionally, the [Unknown + Hexn] species were also determined as new acidic RGPs. Furthermore, HexAn (H) was determined as another form of the acidic RGPs. Conclusion: Compared to the previous methods of analysis, these unprecedented applications of HILICSPE and MALDI-TOF/TOF MS to analyze RGPs proved to be fairly effective for fractionating and detecting neutral and acidic components. This new procedure exhibits great potential as a specific tool for searching and determining various polysaccharides in many herbal medicines.

In this paper, a separate collection guidance and compensation system for PET bottles using Google Teachable Machine was designed and produced. The designed system consists of an image detection unit, a control unit, an audio output unit, and a user recognition unit. The image detection unit uses a machine learning camera to detect PET bottles and detects the presence of labels and caps. If the detected result is out of a specific standard range, the result is transmitted to the control unit using Arduino. The control unit opens and closes the entrance of the collection box using a servo motor according to the result value received from the detection unit. It also controls the voice output unit and performs a function of notifying the user recognition unit. The user recognition unit recognizes the user through the NFC card so that points are accumulated. Through this, it is expected that users will be encouraged to actively separate collection.

Introduction: Coronary artery aneurysm (CAA) is defined as abnormal dilatation of coronary artery, found in 1.5% to 5% of patients undergoing coronary angiography. Coronary artery fistula (CAF) is found in 20% of CAA patients, 17% of CAF is connected to pulmonary artery. Rare case has been reported hemoptysis in patients with coronary aneurysm - pulmonary artery fistula. Case Report: Herein, we report a case of a 58-year-old woman complaining of recently aggravated hemoptysis. Her symptom started 13 years ago, she previously diagnosed with bronchiectasis and underwent bronchial artery embolization at the time. The electrocardiogram showed normal sinus rhythm and no active lung lesion was found in chest radiograph. From chest contrast-enhanced computed tomography, dilated left main coronary artery and left anterior descending artery were detected, which maximum diameter up to 12.17 mm (Fig. 1A). Echocardiogram showed normal systolic function with an ejection fraction of 63%, and normal valve function. But we found diastolic shunt flow towards the distal part of pulmonary valve, suggesting fistulous connection between pulmonary artery and left main coronary artery. Coronary angiography revealed coronary-to-pulmonary artery fistula associated with a giant aneurysm located at left main coronary artery (Fig. 1B) and anomalous branch from RCA os to pulmonary trunk. The patient was successfully by treated by surgical closure of fistula, and was discharged on postoperative day 5.

Recent studies have revealed that probiotics and their metabolites are present under various conditions; however, the role of probiotic metabolites (i.e., postbiotics in pathological states) is controversial. Natural killer (NK) cells play a key role in innate and adaptive immunity. In this study, we examined NK cell activation influenced by a postbiotics mixture in response to gut microbiome modulation in stress-induced mice. In vivo activation of NK cells increased in the postbiotics mixture treatment group in accordance with Th1/Th2 expression level. Meanwhile, the Red Ginseng treatment group, a reference group, showed very little expression of NK cell activation. Moreover, the postbiotics mixture treatment group in particular changed the gut microbiome composition. Although the exact role of the postbiotics mixture in regulating the immune system of stressinduced mice remains unclear, the postbiotics mixture-induced NK cell activation might have affected gut microbiome modulation.
