        Identification of polymorphic loci in the deiodinase 2 gene and their associations with head dimensions in geese

        Objective: This study was conducted to clone and compare the molecular characteristics of the deiodinase 2 (DIO2) gene between Sichuan White geese and Landes geese, and to analyze the association between polymorphisms of the DIO2 gene and head dimensions in Tianfu meat geese. Methods: The coding sequence of the DIO2 gene was cloned by polymerase chain reaction and vector ligation and aligned by DNAMAN software. A total of 350 Tianfu meat geese were used to genotype the polymorphisms of the DIO2 gene and measure the head dimensions. Association analysis between the polymorphisms of the DIO2 gene and head dimensions was carried out. Results: An 840-bp coding sequence of the DIO2 gene was obtained and comparison analysis identified four polymorphic loci between Sichuan White geese and Landes geese. Further analysis showed that the dominant alleles for the four polymorphic loci were G, G, A, and T and the frequency of the heterozygous genotype was higher than that of the homozygous genotype in Tianfu meat geese. Compared to that in the population of non-knob geese of Tianfu meat geese, the head dimensions in the population of knob geese were significantly higher except for nostril height. However, in the non-knob geese, beak width 1, beak width 2, nostril length, cranial width 1, and maxillary length had significant differences among different genotypes or haplotypes/diplotypes. Conclusion: These results suggested that polymorphisms of the DIO2 gene could be considered molecular markers to select larger heads of geese in the population of non-knob geese.

      • Well-designed Te/SnS<sub>2</sub>/Ag artificial nanoleaves for enabling and enhancing visible-light driven overall splitting of pure water

        <P><B>Abstract</B></P> <P>To produce hydrogen and oxygen from photocatalytic overall splitting of pure water provides a promising green route to directly convert solar energy to clean fuel. However, the design and fabrication of high-efficiency photocatalyst is challenging. Here we present that by connecting different nanostructures together in a rational fashion, components that cannot individually split water into H<SUB>2</SUB> and O<SUB>2</SUB> can work together as efficient photocatalyst with high solar-to-hydrogen (STH) energy conversion efficiency and avoid the use of any sacrificial reagent. Specifically, Te/SnS<SUB>2</SUB>/Ag artificial nanoleaves (ANLs) consist of ultrathin SnS<SUB>2</SUB> nanoplates grown on Te nanowires and decorated with numerous Ag nanoparticles. The appropriate band structure of Te/SnS<SUB>2</SUB> p-n junctions and the surface plasmon resonance of Ag nanoparticles synergistically enhance the quantum yield and separation efficiency of electron-hole pairs. As a result, Te/SnS<SUB>2</SUB>/Ag ANLs enable visible-light driven overall water-splitting without any sacrificial reagent and exhibit high H<SUB>2</SUB> and O<SUB>2</SUB> production rates of 332.4 and 166.2μmolh<SUP>−1</SUP>, respectively. Well-preserved structure after long-term measurement indicates its high stability.       • Concurrent Weekly Cisplatin Versus Triweekly Cisplatin with Radiotherapy in the Treatment of Cervical Cancer: A Meta-analysis Result

        Aims: To evaluate the adverse effect and survival outcome of weekly and triweekly cisplatin with radiotherapy in treatment of cervical cancer. Methods: After an extensive literature search between 1995-2011, we analyzed 7 studies to compare weekly cisplatin and triweekly cisplatin combined radiotherapy. Results: Our analysis established that weekly cisplatin has a lower risk of hematologic toxicity than triweekly cisplatin with concurrent radiotherapy in the treatment of cervical cancer. However, there were no differences in progression free survival and overall survival between weekly cisplatin and triweekly cisplatin (p>0.05). Conclusions: Weekly cisplatin combined with concurrent radiation has lower risk in hematologic toxicity than triweekly cisplatin, but does not improve survival. Triweekly cisplatin treatment has longer intervals and is therefore more convenient. Clinicians and patients can choose either weekly cisplatin or triweekly cisplatin combined radiotherapy for cervical cancer.

        Fusion Expression and Immunogenicity of EHEC EspA-Stx2A1 Protein: Implications for the Vaccine Development

        Shiga toxin 2 (Stx2) is a major virulence factor for enterohemorrhagic Escherichia coli (EHEC), which is encoded by λ lysogenic phage integrated into EHEC chromosome. Stx2A1, A1 subunit of Stx2 toxin has gathered extensive concerns due to its potential of being developed into a vaccine candidate. However, the substantial progress is hampered in part for the lack of a suitable in vitro expression system. Here we report use of the prokaryotic system pET-28a::espA-Stx2A1/BL21 to carry out the fusion expression of Stx2A1 which is linked to E. coli secreted protein A (EspA) at its N-terminus. Under the IPTG induction, EspA- Stx2A1 fusion protein in the form of inclusion body was obtained successfully, whose expression level can reach about 40% of total bacterial protein at 25°C, much higher than that at 37°C. Western blot test suggested the refolded fusion protein is of excellent immuno-reactivity with both monoclonal antibodies, which are specific to EspA and Stx2A1, respectively. Anti-sera from Balb/c mice immunized with the EspA-Stx2A1 fusion protein were found to exhibit strong neutralization activity and protection capability in vitro and in vivo. These data have provided a novel feasible method to produce Stx2A1 in large scale in vitro, which is implicated for the development of multivalent subunit vaccines candidate against EHEC O157:H7 infections.

        Synthesis and In Vitro Antitumor Activities of Xanthone Derivatives Containing 1,4-Disubstituted-1,2,3-triazole Moiety

        To explore the more active antitumor compounds, two series of new xanthones, containing 1,4-disubstituted-1,2,3-triazole moiety were designed and synthesized. Eaton’s Reagent and “click reaction” were used in the synthesis. Most of the title compounds showed good inhibitory activity against the hepatoma carcinoma cell line (Bel-7402) and human cervical carcinoma cell line (HeLa) in vitro. Compounds 10a, 10e, 10f, 11r and 11t had potent activity with IC50 values, ranging from 2.2 ± 0.17 to 7.1 ± 0.27 μM, which was equivalent to Doxorubicin.

        Chemical Compositions and Antimicrobial Activities of Essential Oils Extracted from Acanthopanax brachypus

        The essential oils from the flowers, leaves, barks, roots and fruits of A. brachypus were individually extracted by hydrodistillation, and their chemical constituents were isolated and characterized by means of gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS). A total of 160 different constituents accounting for 86.45-96.54% of the total essential oils were identified, and 3 constituents were unidentified (< 0.8%), and significant qualitative and quantitative differences were observed among the oils, in which the yields varied between 0.61-1.83 (mL/100 g dried materials) under the same operational conditions. The oils from flowers, leaves, barks and roots consisted mainly of monoterpenes and sesquiterpene hydrocarbons as well as oxygenated monoterpenes and sesquiterpenes, while the fruit oil consisted mainly of fatty acids and esters. The antimicrobial activities of the essential oils was evaluated against 11 microorganisms (9 for bacteria and 2 for yeast) using agar disc diffusion and broth microdilution methods. The bacteria, including gram-positive bacteria and gramnegative bacteria, were more sensitive to the oils than yeasts.

        Development of a novel reconstruction method for two-phase flow CT with improved simulated annealing algorithm

        Two-phase flow, especially gas-liquid two-phase flow, has a wide application in industrial field. The diagnosis of two-phase flow parameters, which directly determine the flow and heat transfer characteristics, plays an important role in providing the design reference and ensuring the security of online operation of two-phase flow system. Computer tomography (CT) is a good way to diagnose such parameters with imaging method. This paper has proposed a novel image reconstruction method for thermal neutron CT of two-phase flow with improved simulated annealing (ISA) algorithm, which makes full use of the prior information of two-phase flow and the advantage of stochastic searching algorithm. The reconstruction results demonstrate that its reconstruction accuracy is much higher than that of the reconstruction algorithm based on weighted total difference minimization with soft-threshold filtering (WTDM-STF). The proposed method can also be applied to other types of two-phase flow CT modalities (such as X(𝛄)-ray, capacitance, resistance and ultrasound).

        Analysis of Photoluminescence for N-doped and undoped p-type ZnO Thin Films Fabricated by RF Magnetron Sputtering Method

        N-doped ZnO thin films were deposited on n-type Si(100) and homo-buffer layer, and undoped ZnO thin film was also deposited on homo-buffer layer by RF magnetron sputtering method. After deposition, all films were in-situ annealed at 800 oC for 5 minutes in ambient of O2 with pressure of 10 Torr. X-ray diffraction shows that the homo-buffer layer is beneficial to the crystalline of N-doped ZnO thin films and all films have preferable c-axis orientation. Atomic force microscopy shows that undoped ZnO thin film grown on homo-buffer layer has an evident improvement of smoothness compared with N-dope ZnO thin films. Hall-effect measurements show that all ZnO films annealed at 800 oC possess p-type conductivities. The undoped ZnO film has the highest carrier concentration of 1.145x1017 cm-3. The photoluminescence spectra show the emissions related to FE, DAP and many defects such as VZn, ZnO, Oi and OZn. The p-type defects (Oi, VZn, and OZn) are dominant. The undoped ZnO thin film has a better p-type conductivity compared with N-doped ZnO thin film.

        Fabrication of Stiffness Gradient Nanocomposite Hydrogels for Mimicking Cell Microenvironment

        It has attracted much attention to develop a gradient hydrogel for real mimic of the extracellular matrix. Despite simplicity and ease of electrophoresis preparation method, some drawbacks limit the application of the method including toxic organic crosslinker residual, specific ionic monomer types, as well as narrow gradient gaps. In this work, we successfully prepared gradiently crosslinked nonionic PNIPAm/Laponite nanocomposite (NC) hydrogels with controllable gradient structures by adjusting the intensity of the direct current electric field. The results showed that the stiffness, surface property and thermosensitivity of the hydrogel changed with gradient distributions of Laponite. The average stiffness gradients varied from 0.18 kPa/mm, 0.41 kPa/mm to 1.28 kPa/mm by simply adjusting the intensity of the direct current electric field from 0.13 V/mm, 0.20 V/mm to 0.27 V/mm accordingly. The stiffness reached in a range of 13.8~33.0 kPa. The growth behaviors of human cervical cancer cells (HeLa cells) on the gradient NC hydrogel surface were discussed. Spreading and proliferation, as well as morphology of HeLa cells were closely related to the stiffness of the gradient NC hydrogels. This work develops a stiffness-controllable gradient NC hydrogel and opens up a new insight into the better understanding of cells behaviors in different tissues and organs in vivo.
