        Study of the Application of Fructooligosaccharides in Piglets

        In this study, 90 crossbred weaned pigs (Duroc횞Landrace횞Large White) weighing ~ 7.86짹0.06 kg each were randomly allotted to one of three dietary treatments. Control pigs were a fed corn-soybean meal diet with no additives. The two treatment groups were fed the basal diet supplemented either with 75 mg/kg Aureomycin or 0.4% fructooligosaccharides (FOS) in order to study the effects on performance, serological indices, and enteric morphology in addition to examining the content of volatile fatty acids in intestinal digesta. The results indicate that the diets containing FOS and antibiotics had a significant effect on feed conversion ratios (FCR) and diarrhea incidence, as well as increasing the concentrations of isobutyric and butyric acid and total VFAs in the caecum, and acetic acid, isovaleric acid, and total VFAs in feces. Supplementation with FOS also resulted in significantly longer mucosal villi height and a higher percentage of goblet cells compared with the control. No difference was found in crypt depth among the three treatments. While serum glucose levels were significantly higher following FOS supplement, differences in serum total protein, albumin, globulin, and urea nitrogen levels were not significant.

      • Rapid Identification of the Layer Number of Large-Area Graphene on Copper

        <P>Chemical vapor deposition (CVD) on Cu foils emerged as an important method for preparing high-quality and large-area graphene films for practical applications. However, to date it remains challenging to rapidly identify the structural features, especially the layer numbers, of CVD-graphene directly on Cu substrate. Herein, we report an O<SUB>2</SUB>-plasma-assisted approach for identifying the coverage, wrinkles, domain size, and layer number of large-area graphene films on Cu foils by optical microscopy. The wrinkles and grain boundaries of five-layer graphene can be observed with a grayscale increment of ∼23.4% per one graphene layer after O<SUB>2</SUB>-plasma treatment for only 15 s, which allows for checking graphene on Cu foils with a sample size of 17 cm × 20 cm in a few minutes. The Raman spectroscopy and X-ray photoelectron spectroscopy presents a strong layer number dependence of both the plasma induced graphene defects and Cu oxides, which, as indicated by molecular dynamic simulation, is responsible for the improved image contrast as a result of the interaction between O-ions and graphene with different layer numbers. We expect that this O<SUB>2</SUB>-plasma-assisted method would be applied to meter-scale samples if atmospheric-pressure plasma is used and therefore will be beneficial for the fast evaluation of CVD-graphene in both laboratory and industry.</P> [FIG OMISSION]</BR>

        Hepatoprotective effect of phenylethanoid glycosides from Incarvillea compacta against CCl<sub>4</sub>-induced cytotoxicity in HepG2 cells

        The extraction and solvent partition of roots of Incarvillea compacta, a traditional Tibetan folk medicine, and repeated column chromatography and preparative high-performance liquid chromatography for n-butanol fraction yielded four phenylethanoid glycosides, crenatoside (1), 3'''-O-methylcrenatoside (2), leucoseceptoside A (3), and martynoside (4). The chemical structures were identified on the basis of spectroscopic data analyses including NMR and MS. All compounds were isolated for the first time from the plant. Compound 1 exerted better 1,1-diphenyl-2-picrylhydrazyl free radical scavenging activity. In addition, compounds 1-4 were evaluated for their hepatoprotective activity on carbon tetrachloride ($CCl_4$)-induced liver injury in HepG2 cells. Pretreatment of HepG2 cells with compound 1-4 significantly increased the viability on $CCl_4$-induced cell death. Furthermore, compounds 1-4 also alleviated $CCl_4$-induced hepatotoxicity by enhancement of the antioxidant enzyme activities of superoxide dismutase and reduction of the malondialdehyde content, intracellular ROS as well as NF-${\kappa}B$ transactivation. Our results suggest that phenylethanoid glycosides ameliorate $CCl_4$-induced cell injury, and this protection was likely due to antioxidative activity and down-regulation of NF-${\kappa}B$.

        Alkyl Polyglucosides stabilized foam for gas controlling in high-temperature and high-salinity environments

        APG can be employed as a promising foaming agent due to its largely independent of temperature and salinity. Aiming at the APGs foam, we conducted the experiments about adsorption behaviors, surface viscoelasticity, bulk foam properties and core flooding under conditions of high-temperature and -salinity. Our results demonstrated that APG (n = 10) foam presents a good foam performance and foam properties are highly depended on the surface viscoelasticity and are not completely determined by the adsorbing capacity at the interface of gas–water. In the porous media, an appropriate foam quality of APG foam is contributed to increase the foam strength and gas trapping.

        A Novel Superoxide Anion Radical Nonenzymatic Electrochemical Sensor Based on PtRuCu Ternary Alloy Nanoparticles/Graphene Composite Modified Electrode

        A superoxide anion radical (O2 ˙-) nonenzymatic electrochemical sensor based on PtRuCu ternary alloy nanoparticles/electrochemically reduced graphene oxide (ERGO) composite modified glassy carbon electrode (PtRuCu/ERGO/GCE) was developed. ERGO and PtRuCu/ERGO composites were characterized by scanning electron microscopy. The prepared PtRuCu/ERGO/ GCE showed remarkable performance toward the electrocatalytic reduction of (O2 ˙-). The sensor possessed a high sensitivity of 1725µA · mM-1 · cm-2, a wide linear range of 7.5–330 µM (R2 = 0.993) and a low detection limit of 0.7 µM (S/N = 3), as well as excellent selectivity, reproducibility and stability, which can be attributed to the synergistic effect of ERGO and PtRuCu ternary alloy nanoparticles. Therefore, the prepared sensor is promising for electrochemical determination of O2˙-.

        piRNA-1742 promotes renal cell carcinoma malignancy by regulating USP8 stability through binding to hnRNPU and thereby inhibiting MUC12 ubiquitination

        Accumulating studies have confirmed that PIWI-interacting RNAs (piRNAs) are considered epigenetic effectors in cancer. We performed piRNA microarray expression analysis on renal cell carcinoma (RCC) tumor tissues and paired normal tissues and performed a series of in vivo and in vitro experiments to explore piRNAs associated with RCC progression and investigate their functional mechanisms. We found that piR-1742 was highly expressed in RCC tumors and that patients with high piR-1742 expression had a poor prognosis. Inhibition of piR-1742 significantly reduced tumor growth in RCC xenograft and organoid models. Mechanistically, piRNA-1742 regulates the stability of USP8 mRNA by binding directly to hnRNPU, which acts as a deubiquitinating enzyme that inhibits the ubiquitination of MUC12 and promotes the development of malignant RCC. Subsequently, nanotherapeutic systems loaded with piRNA-1742 inhibitors were found to effectively inhibit the metastasis and growth of RCC in vivo. Therefore, this study highlights the functional importance of piRNA-related ubiquitination in RCC and demonstrates the development of a related nanotherapeutic system, possibly contributing to the development of therapeutic approaches for RCC.
