      • Fault Analysis in Software with the Data Interaction of Classes

        With the development of science, software becomes more and more complex, which makes it difficult to do fault propagation analysis. Software Network is a general tool in studying fault propagation, but because of the diversification of software’s architecture, we can no longer oversimplify the data flow of nodes in the software network which is universal in conventional method to analyze fault propagation. This paper put forward a new structure Class Interactive Network based on the data interaction of classes in software to analyze software’s fault propagation in the class hierarchy and simulate software classes’ performance in fault propagation with quantitative analysis in Class Interactive Network.

        Morphology and transcriptome differences between the haploid and diploid drones of Apis cerana

        In general, drone honey bees are haploid and develop from unfertilized eggs. However, a diploid drone can arise in an inbred colony. In this study, the morphological characteristics and gene expression profile of the haploid and diploid drones of Apis cerana were analyzed to reveal the differences between them. The ploidy level of the droneswas identified by FlowCytometry (FCM). The characters of the forewings,wetweight of reproductive organs and of newly emerged drones, were investigated. Then, a high throughput transcriptomic analysis was performed using RNA-seq in diploid and haploid drones. The results showed that the wet weight and reproductive organs of diploid droneswere significantly lighter than those of haploid drones. About 201 million high-quality reads were generated from RNA-seq, and 75.99–78.12% of the data weremapped to Apis cerana genome. 360 genes were differentially expressed between diploid and haploid drone, with 152 up-regulated and 208 downregulated in the diploid drones. Functional analysis identified that these genes were significantly enriched in 28 pathways. Comparative transcriptomic analysis detected several differentially expressed genes, which lay a foundation for future studies on molecular mechanisms underlying biology difference in drones in Apis cerana.

        Transcriptome comparison between newly emerged and sexually matured bees of Apis mellifera

        In order to understand the transcriptome characteristics of queens and drones of honeybee Apis mellifera, the transcriptome differences between newly emerged stage and sexually matured stage of queens and drones of A. mellifera L. were compared using high-throughput RNA-Seq. In drones, a total of 1618 DEGs were detected between the two stages. Out of these, 782 geneswere up-regulated and 836 geneswere down-regulated in sexually matured drones compared to newly emerged drones. In queens, the DEGs between the two stages were 1340, with 667 up-regulated and 673 down-regulated genes in matured queens compared to newly emerged queens. 411 genes showed the same expression trend in drones and queens during sexual maturing, with 233 (56.69%) up-regulated genes and 178 (43.31%) down-regulated at newly emerged stage. We found that genes encoding cuticular proteins (CP), cytochrome P450 (CYP), odorant binding proteins (OBP) and odorant receptor (OR), which are related to developments of bones, reproductive system and olfaction system, were differentially expressed between the sexually matured bees and the newly emerged bees. The results indicated that the expression levels of a large number of genes changed during sexual maturing of A. mellifera L. bees, which give us an insight into the characteristics of the gene expression during sexual maturing of adult queens and drones.

        Effect of the Outer Diameter on the Behavior of Square RC Columns Strengthened with Self-Compacting Concrete Filled Circular Steel Tube

        This paper aimed to investigate the optimum outer diameter of square RC columns strengthened with self-compacting concrete fi lled circular steel tube. An experiment of twelve strengthened RC columns (SRC) and two referential RC columns subjected to axial load was presented. The variables considered in the test were the confi nement index and the diameter of the strengthened section. The experimental results showed that, with the same confi nement index, the material strength utilization of SRC columns dropped with the increasing of strengthened section diameter. There existed an optimum outer diameter to take full use of material strength. With the outer diameter below the optimum value, the material strength utilization increased with increasing confi nement index. As the outer diameter beyond the optimum one, the material strength utilization decreased with improving confi nement index.

        Differential proteomic studies of the genic male-sterile line and fertile line anthers of upland cotton (Gossypium hirsutum L.)

        Pollen development is disturbed in the microsporedevelopment stage of the double-recessive nuclearmale-sterile line ms5ms6 (Gossypium hirsutum L.). Thisstudy aimed to identify differentially expressed antherproteins and their potential roles in pollen development andmale sterility. We compared the proteomes of sterile andfertile anthers of the double recessive nuclear male-sterileline ms5ms6. Approximately 1,390 protein spots weredetected by two-dimensional differential gel electrophoresis. Proteins with altered accumulation levels in sterileanthers compared with fertile anthers were identified bymass spectrometry and the NCBInr and Viridiplantae ESTdatabases. Down-regulated proteins in the sterile anthersincluded cytosolic ascorbate peroxidase 1 and glutaminyltRNAsynthetase (glutamine-tRNA ligase). Several carbohydratemetabolism- and photosynthesis-related enzymeswere also present at lower levels in the mutant anthers. Bycontrast, ATP-dependent RNA helicase eIF4A-13, NADHdehydrogenase subunit 1, enolase, gibberellin 20-oxidase,gibberellin 3-hydroxylase 1, alcohol dehydrogenase 2d,3-ketoacyl-CoA synthase, and trehalose 6-phosphate synthasewere expressed at higher levels in sterile anthers thanin fertile anthers. The regulation of upland cotton pollendevelopment involves a complex network of differentiallyexpressed genes. This study provides the foundation forfuture investigations of gene function in upland cottonpollen development and male sterility.

        Suppression of Aurora-A oncogenic potential by c-Myc downregulation

        The abnormality of serine/threonine kinase Aurora-A is seen in many types of cancers. Although in physiological context it has been shown to play a vital role in cellular mitosis, how this oncogene contributes to tumorigenesis remains unclear. Here we demonstrate that Aurora-A overexpression enhances both the expression level and transcriptional activity of c-Myc. The inhibition of c-Myc expression by RNA interference significantly impaired the oncogenic potential of Aurora-A, resulting in attenuated cellular proliferation and transformation rates as well as fewer centrosomal aberrations. Furthermore, downregulation of c-Myc effectively overcame Aurora-A-induced resistance to cisplatin in esophageal cancer cells. Taken together,our results suggest an important role for c-Myc in mediating the oncogenic activity of Aurora-A, which may in turn allow for future targeting of c-Myc as a potential therapeutic strategy for tumors with Aurora-A overexpression.

        Optimal Scheduling of Electric Vehicles Charging in low-Voltage Distribution Systems

        Uncoordinated charging of large-scale electric vehicles (EVs) will have a negative impact on the secure and economic operation of the power system, especially at the distribution level. Given that the charging load of EVs can be controlled to some extent, research on the optimal charging control of EVs has been extensively carried out. In this paper, two possible smart charging scenarios in China are studied: centralized optimal charging operated by an aggregator and decentralized optimal charging managed by individual users. Under the assumption that the aggregators and individual users only concern the economic benefits, new load peaks will arise under time of use (TOU) pricing which is extensively employed in China. To solve this problem, a simple incentive mechanism is proposed for centralized optimal charging while a rolling-update pricing scheme is devised for decentralized optimal charging. The original optimal charging models are modified to account for the developed schemes. Simulated tests corroborate the efficacy of optimal scheduling for charging EVs in various scenarios.

        Experimental and Numerical Study on Fatigue Behavior of RC Beams Strengthened with U-Shaped Steel Casing

        To achieve a rapid and great enhancement in the fl exural performance of reinforced concrete beams, this study proposes a new strengthening method using U-shaped steel casing and focuses on studying the fatigue performance of strengthened beams. In this paper, ten specimens were tested under fatigue loads. Testing results demonstrated that no obvious damage was observed from the strengthened beam after 2 million fatigue cycles compared with the reference beam which failed at 58.97 × 10 4 fatigue cycles, indicating the good joint performance of the U-shaped steel casing and the RC beam. The strengthened beams generally failed by the sudden rupture of the bottom steel plate accompanied by concrete crushing. The mid-span defl ection and material strains all increased rapidly at the beginning, then steadily developed during nearly 80% of the fatigue life, and grew rapidly again after the weld toe cracking until the strengthened beam failed. The fatigue life was signifi cantly extended with the increase of either the height of the steel box or the thickness of the bottom plate, while it was dramatically shortened with the increase in the fatigue load amplitude. To facilitate the use of the outcomes, a simplifi ed formula for predicting the concrete residual strain of strengthened beams was derived. The predicted results matched the experimental ones well. The derived formula is theoretically only applicable to strengthened beams with dimensions same as the specimens in the experimental tests under similar loading conditions. Reasonably good agreements were found between the fi nite element analyses and experimental results. The developed fi nite element model can well simulate the failure mode under static load and the damage evolution under fatigue load. Based on the stress analysis results, the fatigue lives of the strengthened beams were predicted relatively accurately using the Fe-safe software, with the deviations mostly less than 15%.
