        Diagnosis and Treatment of Contemporary Chinese College Students’ ‘Empty Heart Disease’: Based on the Theory and Practice of Philosophical Counseling and Therapy

        In their daily lives, with varying degrees, many Chinese college students are stuck in the predicaments of lacking life meanings and values. They have a strong sense of loneliness and meaninglessness, not knowing what they are living for. If such ethical predicaments are not solved efficiently and timely, then they 1may result in very serious psychological, mental or even physical problems. In fact, the so-called ‘empty heart disease’ (空心病) is not a kind of psychological or mental disease, but rather a ‘perplexity’ in mind which is very commonly seen during the growth of the youth. Based on the philosophical theories and methods, the trained philosophical workers can provide suggestions and guidances towards people’s concrete problems and situations, so that people can obtain larger space and also better capacity of thinking; by doing so, the perplexities that trouble the people will thus be hopefully solved, achieving the purpose of ‘disambiguation’. In the philosophical counselors’ opinions, it is the human beings themselves that give meanings to their lives. Therefore, to solve the ‘empty heart disease’ problem, we can not depend on just preaching or inculcating externally; rather, we should encourage and inspire the college students to reflect on the purposes of their lives and their self-identities, so that they can afterwards recreate and develop their values and senses of meanings. Finally, the differences of ‘empty heart disease’, ‘noogenic neurosis’ and ‘existential neurosis’ are discussed in this paper, while an analysis of the boundaries between the need for psychotherapy and the need for philosophical counseling in relation to the three ‘diseases’ is also made.

        Presupposition Disclosure: A Key Strategy in Philosophical Practice

        Philosophical practice, which is also sometimes called philosophical counseling or philosophical therapy, is at the leading edge of applied philosophy in the west, and it has revolutionarily constituted a new paradigm in philosophical research. As a “doctor of the thoughts”, a philosophical practitioner’s first and foremost target is to disclose and examine the rationality of the latent presuppositions that are hidden in the client’s web of beliefs. Philosophers such as G. Frege, P. F. Strawson and R. C. Stalnaker have investigated profoundly on semantic presupposition and pragmatic presupposition. To identify presuppositions, philosophical practitioners have to rely on presupposition triggers that appear in the clients’ words, which are usually some particular terms or syntactic structures. Presupposition and presumption are closely connected with each other, and the role of the latter is mainly realized by the former. Through examining on the rationality of the client’s implicit pragmatic presumptions, semantic presumptions, factual presumptions and mutual presumptions that are related to the client’s presuppositions, the philosophical practitioner may hopefully succeed in helping the client resolve or dissolve the painful perplexities in his mind, which are usually brought about by the client’s irrational presuppositions. In this way, philosophical practitioners actually restore the therapeutic function of philosophy, making philosophy return to the normal people’s daily life world.

        Microstructure evolution and effect on deuterium retention in oxide dispersion strengthened tungsten during He<sup>+</sup> irradiation

        Oxide dispersion-strengthened materials W-1wt%Pr<sub>2</sub>O<sub>3</sub> and W-1wt%La<sub>2</sub>O<sub>3</sub> were synthesized by wet chemical method and spark plasma sintering. The field emission scanning electron microscopy (FE-SEM) analysis, XRD and Vickers microhardness measurements were conducted to characterize the samples. The irradiations were carried out with a 5 keV helium ion beam to fluences up to 5.0 × 10<sup>21</sup> ions/m<sup>2</sup> under 600 ℃ using the low-energy ion irradiation system. Transmission electron microscopy (TEM) study was performed to investigate the microstructural evolution in W-1wt%Pr<sub>2</sub>O<sub>3</sub> and W-1wt%La<sub>2</sub>O<sub>3</sub>. At 1.0 × 10<sup>20</sup> He<sup>+</sup>/m<sup>2</sup>, the average loops size of the W-1wt%Pr<sub>2</sub>O<sub>3</sub> was 4.3 nm, much lower than W-1wt% La<sub>2</sub>O<sub>3</sub> of 8.5 nm. However, helium bubbles were not observed throughout in both doped W materials. The effects of pre-irradiation with 1.0 × 10<sup>21</sup> He<sup>+</sup>/m<sup>2</sup> on trapping of injected deuterium in doped W was studied by thermal desorption spectrometry (TDS) technique using quadrupole mass spectrometer. Compared with the samples without He<sup>+</sup> pre-irradiation, deuterium (D) retention of doped W materials increased after He<sup>+</sup> irradiation, whose retention was unsaturated at the damage level of 1.0 × 10<sup>22</sup>D<sub>2</sub><sup>+</sup>/m<sup>2</sup>. The present results implied that irradiation effect of He<sup>+</sup> ions must be taken into account to evaluate the deuterium retention in fusion material applications.

        Response of Saccharomyces cerevisiae to Ethanol Stress Involves Actions of Protein Asrlp

        During the fermentation process of Saccharomyces cerevisiae, yeast cells must rapidly respond to a wide variety of external stresses in order to survive the constantly changing environment, including ethanol stress. The accumulation of ethanol can severely inhibit cell growth activity and productivity. Thus, the response to changing ethanol concentrations is one of the most important stress reactions in S. cerevisiae and worthy of thorough investigation. Therefore, this study examined the relationship between ethanol tolerance in S. cerevisiae and a unique protein called alcohol sensitive RING/PHD finger 1 protein (Asr1p). A real-time PCR showed that upon exposure to 8% ethanol, the expression of Asr1 was continuously enhanced, reaching a peak 2 h after stimulation. This result was confirmed by monitoring the fluorescence levels using a strain with a green fluorescent protein tagged to the C-terminal of Asr1p. The fluorescent microscopy also revealed a change in the subcellular localization before and after stimulation. Furthermore, the disruption of the Asr1 gene resulted in hypersensitivity on the medium containing ethanol, when compared with the wild-type strain. Thus, when taken together, the present results suggest that Asr1 is involved in the response to ethanol stress in the yeast S. cerevisiae.

        The Dynamic Mechanical Properties for Recycled Aggregate Concrete under Tensile-Compressive States

        The experiments under biaxial tensile-compressive (T-C) complex stress were carried out for two kinds of substitution percentages of recycled coarse aggregate (RCA) on biaxial proportional loading (α = σ1:σ3 =1:0, 0: -1, 0.05: -1, 0.1: -1, 0.25: -1, 0.5: -1, 1: -1 and-1: -1) and four loading deformation rates of 10−5/s, 10−4/s , 10−3/s, and 10−2/s. The research was completed in the static-dynamic true triaxial machine. The experimental phenomena indicate the tensile failure appears in these specimens of recycled aggregate concrete (RAC) under biaxial T-C stress states. The dynamic mechanical behavior for RAC under biaxial T-C loadings was affected by the strain rates, stress ratios and its substitution percentages of RCA. Accompanied by the increase of strain rates, its strength of RAC is improved. As the tensile stress increases, its compressive strength decreases. The T-C strengths for RAC under biaxial T-C states are less than its corresponding uniaxial T-C strengths at a certain identical strain rates, respectively. Considering the effect of loading rates, a new tensile-compressive failure criterion is established under complex stress states.

        Cloning and functional characterization of two GsSnRK1 gene promoters from wild soybean

        Sucrose non-fermenting-related protein kinase 1 (SnRK1) belongs to a plant serine/threonine protein kinase family, and is part of a large interactive network of signaling pathways in plants to regulate many aspects of growth and development, and its gene expressions are highly responsive to multiple external stimuli. To functionally characterize the role of different SnRK1 members in wild soybean and identify the expression patterns of novel SnRK1 promoters for transgenic research, in this study, we performed an in silico analysis of the SnRK1 gene family in wild soybean and found GsSnRK1.1 and GsSnRK1.2 are main SnRK1 isoforms in wild soybean. The promoter fragments of both GsSnRK1.1 and GsSnRK1.2 were isolated, and a series of 5′-deletion and the full-length versions of both GsSnRK1.1 and GsSnRK1.2 promoters were fused with GUS reporter gene to generate expression vectors. The GUS activities in different tissues of seedling and adult growth stages of transgenic Arabidopsis plants were determined, and the GUS activities in transgenic Arabidopsis plants under NaCl, NaHCO3, PEG, ABA, MeJA and SA were also analyzed. The results showed that GsSnRK1.1 and GsSnRK1.2 genes have different expression patterns and both promoters may contain enhancers and silencers. In addition, NaCl, NaHCO3, ABA, MeJA, and PEG treatments enhanced, whereas SA treatment decreased the GUS activity under the control of both GsSnRK1.1 and GsSnRK1.2 promoters, which may be due to the distributions of various cis-elements in both GsSnRK1.1 and GsSnRK1.2 promoter regions. These results will contribute to a better understanding of the underlying mechanisms of GsSnRK1 genes in response to various abiotic stresses, and as stress-inducible promoters, GsSnRK1.1 and GsSnRK1.2 promoters can be candidates for practical application in agriculture.

        Activated Protein C Protects Myocardium Via Activation of Anti-apoptotic Pathways of Survival in Ischemia-reperfused Rat Heart

        Activated protein C (APC) is known to be beneficial on ischemia reperfusion injury in myocardium. However, the protection mechanism of APC is not fully understood. The purpose of this study was to investigate the effects and possible mechanisms of APC on myocardial ischemic damage. Artificially ventilated anaesthetized Sprague-Dawley rats were subjected to a 30 min of left anterior descending coronary artery occlusion followed by 2 hr of reperfusion. Rats were randomly divided into four groups; Sham, I/R, APC preconditioning and postconditioning group. Myocardial infarct size, apoptosis index, the phosphorylation of ERK1/2, Bcl-2, Bax and cytochrome c genes and proteins were assessed. In APC-administrated rat hearts, regardless of the timing of administration, infarct size was consistently reduced compared to ischemia/reperfusion (I/R) rats. APC improved the expression of ERK1/2 and anti-apoptotic protein Bcl-2 which were significantly reduced in the I/R rats. APC reduced the expression of pro-apoptotic genes, Bax and cytochrome c. These findings suggest that APC produces cardioprotective effect by preserving the expression of proteins and genes involved in anti-apoptotic pathways, regardless of the timing of administration.

        Clinicopathologic and Prognostic Significance of the Zinc Finger of the Cerebellum Family in Invasive Breast Cancer

        Purpose: Five members of the zinc finger of the cerebellum (ZIC) family—ZIC1, ZIC2, ZIC3, ZIC4, and ZIC5—have been shown to be involved in various carcinomas. Here, we aimed to explore the clinicopathologic and prognostic roles of ZIC family members in invasive breast cancer patients using immunohistochemical analysis, western blotting analysis, and real-time quantitative polymerase chain reaction (RT-qPCR). Methods: A total of 241 female invasive breast cancer patients who underwent radical mastectomy between 2009 and 2011 were enrolled. ZIC proteins in 241 pairs of breast tumors and corresponding normal tissues were investigated using immunohistochemistry and the clinicopathologic roles of proteins were analyzed using Pearson’s chi-square test. Kaplan-Meier curves and Cox regression analysis were also used to analyze the prognostic value of the ZIC proteins. In addition, 12 pairs of fresh-frozen breast tumors and matched normal tissues were used in the western blotting analysis and RT-qPCR. Results: Only ZIC1 expression in normal tissues was obviously higher than that in tumors (p<0.001). On multivariate analysis, ZIC1 expression (in overall survival analysis: hazard ratio [HR], 0.405, 95% confidence interval [CI], 0.233–0.702, p=0.001; in disease-free survival analysis: HR, 0.395, 95% CI, 0.234–0.669, p=0.001) was identified as a prognostic indicator of invasive breast cancer. Conclusion: ZIC1, but not the other proteins, was obviously decreased in breast tumors and associated with clinicopathologic factors. Thus, ZIC1 might be a novel indicator to predict the overall and disease-free survival of invasive breast cancer patients.

        Net micromineral requirements for maintenance and growth of ewe lambs at the latter fattening period

        Objective: The objective of this study was to investigate the net micromineral (Cu, Fe, Mn, Zn) requirements for maintenance and growth of Dorper×Jinzhong crossbred ewe lambs at their latter fattening period. Methods: Thirty 1/2 Dorper × 1/2 Jinzhong crossed F1 ewe lambs (35±0.5 kg of body weight [BW]) were used and divided into five groups in a randomized design for a comparative slaughter trial. At the beginning of the experiment, six lambs were randomly selected and slaughtered at 35 kg BW to determine their initial body composition. When their BWs reached 43 kg, another six lambs fed ad libitum were slaughtered to serve as an intermediate slaughter group. The retained eighteen lambs were randomly distributed into three groups and offered one type of feed at 100%, 65%, and 40% of the ad libitum intake. When the lambs fed ad libitum reached a BW of 50 kg, the three groups were slaughtered. The body composition (muscle, fat, bone, blood with viscera, skin, and wool) were weighted, ground, mixed, and subsampled for mineral content analysis. Results: The net maintenance requirements of Cu, Fe, Mn, and Zn were 0.017, 0.160, 0.004, and 0.067 mg/kg BW/d, respectively, and the net growth requirements per 100 grams of average daily gain ranged from 0.48 to 0.51 mg of Cu, 2.63 to 2.17 mg of Fe, 0.12 to 0.15 mg of Mn, and 2.07 to 2.00 mg of Zn, respectively, for Dorper × Jinzhong crossed ewes from 35 to 50 kg BW. Conclusion: Our results suggest that the micromineral requirements for both maintenance and growth of Dorper × Jinzhong crossbred ewe lambs were quite different from the recommendations of NRC (2007), except for Zn.

        Production of 3-hydroxypropionic acid by recombinant Klebsiella pneumoniae based on aeration and ORP controlled strategy

        A biosynthetic pathway for the production of 3-hydroxypropionic acid (3-HP) from glycerol was established in recombinant Klebsiella pneumoniae by introducing the aldehyde dehydrogenase gene from Escherichia coli. The activity of aldehyde dehydrogenase, which oxidized 3-hydroxypropionaldehyde (3-HPA) to 3-HP, was detected and 3-HP was produced by the recombinant strains. Three different oxygen supply strategies, associated with measuring the oxidoreduction potential (ORP) during the fermentation under these conditions, were adopted for higher production of 3-HP by the recombinant cells. About 0.8 g/l 3-HP and more 1,3-propanediol production by the recombinant Klebsiella pneumoniae were obtained under completely aerobic conditions. Under micro-aerobic conditions, 3-HP production could be increased to 2.2 g/l and 1,3-propanediol production was almost the same as in the original strain. Under the anaerobic conditions, 1,3-propanediol was the main product and about 1.3 g/l 3-HP was produced. Finally, 3-HP production of the recombinant strain was increased to 2.8 g/l under micro-aerobic condition with a further two-stage ORP controlled strategy.
